Background Digital is the new blood, where collection, use, and storage of raw digital data streams enabled by technology-embedded sensors are replacing analog methods. Analyses of digital data extend beyond current applications, analogous to previously stored blood currently be used for omics/biomarker research. Today digital technologies are used to measure Alzheimer’s disease (AD)-related clinical symptoms (e.g., cognition, function, and behavior), but future possibilities remain to be determined. Importantly, digital data collection can go beyond current intermittent sampling by employing internet-connected applications and devices that allow real-time monitoring and accurate detection of changes, even within a widely heterogeneous spectrum. Method Beginning in 2005, the Framingham Heart Study (FHS) began digitally recording participants’ spoken responses to neuropsychological (NP) test questions, and in 2011 extended to a digital pen to record written responses. The emergence of wearable devices and smartphone applications has led to an expanded use of digital technologies beyond recordings during NP assessments. The Boston University Alzheimer’s Disease Research Center (BU ADRC) leverages digital sensors in multiple internet-connected devices that longitudinally collect digital data, in both in-person and remote settings. Result The first publication involving FHS digital data was in 2016; the remaining (n> 17) have been from 2020-current. Digital data measuring cognition, function and behavior are being collected through the BU ADRC, where results to date indicate nearly 100% and 75% uptake of active and passive engagement applications, respectively, and 95% uptake for wearable devices (n = 60). Longitudinal assessments include > 3 months for ∼50 participants; > 1 year for n∼21 participants. Chemotherapy, radiotherapy, targeted therapy, and immunotherapy are established cancer treatment modalities that are widely used due to their demonstrated efficacy against tumors and favorable safety profiles or tolerability. Nevertheless, treatment resistance continues to be one of the most pressing unsolved conundrums in cancer treatment. Hypoxia-inducible factors (HIFs) are a family of transcription factors that regulate cellular responses to hypoxia by activating genes involved in various adaptations, including erythropoiesis, glucose metabolism, angiogenesis, cell proliferation, and apoptosis. Despite this critical function, overexpression of HIFs has been observed in numerous cancers, leading to resistance to therapy and disease progression. In recent years, much effort has been poured into developing innovative cancer treatments that target the HIF pathway. Combining HIF inhibitors with current cancer therapies to increase anti-tumor activity and diminish treatment resistance is one strategy for combating therapeutic resistance. This review focuses on how HIF inhibitors could be applied in conjunction with current cancer treatments, including those now being evaluated in clinical trials, to usher in a new era of cancer therapy.

Imaging materials and inner structures with resolution below the diffraction limit has become of fundamental importance in recent years for a wide variety of applications. In this work, we report sub-diffractive internal structure diagnosis of hexagonal boron nitride by exciting and imaging hyperbolic phonon polaritons. Based on their unique propagation properties, we are able to accurately locate defects in the crystal interior with nanometer resolution. The precise location, size and geometry of the concealed defects is reconstructed by analyzing the polariton wavelength, reflection coefficient and their dispersion. We have also studied the evolution of polariton reflection, transmission and scattering as a function of defect size and photon frequency. The nondestructive high-precision polaritonic structure diagnosis technique introduced here can be also applied to other hyperbolic or waveguide systems, and may be deployed in the next-generation bio-medical imaging, sensing and fine structure analysis. Infections can lead to persistent symptoms and diseases such as shingles after varicella zoster or rheumatic fever after streptococcal infections. Similarly, severe acute respiratory syndrome coronavirus 2 (SARS‑CoV‑2) infection can result in long coronavirus disease (COVID), typically manifesting as fatigue, pulmonary symptoms and cognitive dysfunction. The biological mechanisms behind long COVID remain unclear. We performed a genome-wide association study for long COVID including up to 6,450 long COVID cases and 1,093,995 population controls from 24 studies across 16 countries. We discovered an association of FOXP4 with long COVID, independent of its previously identified association with severe COVID-19. The signal was replicated in 9,500 long COVID cases and 798,835 population controls. Given the transcription factor FOXP4’s role in lung physiology and pathology, our findings highlight the importance of lung function in the pathophysiology of long COVID. © The Author(s) 2025.

As the only endemic Neotropical parrot to have recently lived in the northern hemisphere, the Carolina parakeet (Conuropsis carolinensis) was an iconic North American bird. The last surviving specimen died in the Cincinnati Zoo in 1918 (Elphick et al., 2010). The cause of its extinction remains contentious: besides excessive mortality associated to habitat destruction and active hunting, their survival could have been negatively affected by its range having become increasingly patchy (Burgio et al., 2017), or by the exposure to poultry pathogens (Snyder, 2004;Snyder and Russel, 2002). In addition, the Carolina parakeet showed a predilection for cockleburs, an herbaceous plant that contains a powerful toxin, carboxyatractyloside, or CAT (Stuart et al., 1981), which did not seem to affect them but made the birds notoriously toxic to most predators (Snyder, 2004). To explore the demographic history of this bird we generated the complete genomic sequence of a preserved specimen held in a private collection in Spain, as well as of a close extant relative, Aratinga solstitialis. We identified non-synonymous genetic changes in two highly conserved proteins known to interact with CAT that could underlie a specific dietary adaptation to this toxin. Our genomic analyses did not reveal evidence of a long-term demographic decline in the Carolina parakeet;also, its genome did not exhibit the long runs of homozygosity that are typically found in endangered species and are signals of recent inbreeding. As such, our results suggest its extinction was an abrupt process, and thus likely solely attributable to human causes. © 2019, The Authors. All rights reserved.

We determined the complete mitochondrial genome ofMetapenaeopsis dalei(Rathbun, 1902), which is collected from East China Sea (124°E and 33.5°N). Total mitochondrial genome length ofM. daleiwas 15 939 bp, in which 13 proteins, two ribosomal RNAs, 22 transfer RNAs and a putative control region were encoded. The gene arrangement ofM. daleiwas well conserved with nine known Penaeidae mitochondrial genomes fromCOX1totRNATyr. The protein-coding genes started with ATN except forCOX1in which ACG is used. Four genes (COX2, COX3, ND3andND5) exhibited an incomplete stop codon. Nucleotide sequence identity ofM. daleimitochondrial genome to those of nine Penaeidae species ranged from 78% to 80%. Based on the COI region,M. daleiis most closely related toPenaeus notialis.

Ultrashort pulse fiber lasers (FLs) are required to have higher power to achieve high quality laser processing of difficult-to-cut materials such as carbon fiber reinforced plastics (CFRPs). Our present work develops an all-fiber coherent addition system to achieve a high power ultrashort pulse FL beyond pulse-power limits due to nonlinear effects in rare-earth-doped fiber [1]. Coherent addition is a technology that divide a single pulse into multiple pulses, and amplify the power by fiber amplifiers, and combine the amplified pulses again into a single pulse [2]. However, the coherent addition of ultrashort pulses is technically difficult because ultrashort pulse lasers have very low coherence due to their broad spectrums. Therefore, we have developed a novel all-fiber coherent addition system that can passive-matching of phase and polarization, and attempted high-efficiency and high-power by femtosecond pulse.

Background: Hearing loss affects access to spoken language, which can affect cognition and development, and can negatively affect social wellbeing. We present updated estimates from the Global Burden of Disease (GBD) study on the prevalence of hearing loss in 2019, as well as the condition's associated disability. Methods: We did systematic reviews of population-representative surveys on hearing loss prevalence from 1990 to 2019. We fitted nested meta-regression models for severity-specific prevalence, accounting for hearing aid coverage, cause, and the presence of tinnitus. We also forecasted the prevalence of hearing loss until 2050. Findings: An estimated 1·57 billion (95% uncertainty interval 1·51–1·64) people globally had hearing loss in 2019, accounting for one in five people (20·3% [19·5–21·1]). Of these, 403·3 million (357·3–449·5) people had hearing loss that was moderate or higher in severity after adjusting for hearing aid use, and 430·4 million (381·7–479·6) without adjustment. The largest number of people with moderate-to-complete hearing loss resided in the Western Pacific region (127·1 million people [112·3–142·6]). Of all people with a hearing impairment, 62·1% (60·2–63·9) were older than 50 years. The Healthcare Access and Quality (HAQ) Index explained 65·8% of the variation in national age-standardised rates of years lived with disability, because countries with a low HAQ Index had higher rates of years lived with disability. By 2050, a projected 2·45 billion (2·35–2·56) people will have hearing loss, a 56·1% (47·3–65·2) increase from 2019, despite stable age-standardised prevalence. Interpretation: As populations age, the number of people with hearing loss will increase. Interventions such as childhood screening, hearing aids, effective management of otitis media and meningitis, and cochlear implants have the potential to ameliorate this burden.
