A tissue bath system, to be used as an alternative to complex perfusion chambers, was constructed for use in cardiac electrophysiological studies. This system consists of an acrylic chamber to hold circulating physiological medium such as DMEM, suspended in a water bath warmed by a hot plate. Temperature and pH were controlled to mimic physiological conditions. Rat and porcine cardiac tissues, were used to test viability of the conditions presented in the bath system. Using a cardiac mapping system, the tissues were stimulated and responses recorded. From the recordings we were able to calculate conduction velocities and spatial dispersion of activation indices. The results are comparable to previous in-vivo work, which suggests that the tissue bath system design can maintain tissue viability. This tissue bath system is a relatively simple alternative for ex-vivo testing of cardiac tissues.

A resolution measure that overcomes the limitations of Rayleigh's criterion was proposed that predicted the resolution of an optical microscope was not limited and that it can be improved by increasing the number of detected photons. An interdisciplinary approach was used to obtain the new resolution measure in which tools of advanced statistical image processing address a classical problem in optical microscopy. Single-molecule imaging experiments have verified that distances between single molecules that are well below Rayleigh's criterion can be measured with an accuracy in accordance with that prediction. An analytical expression for the fundamental resolution measure (FREM) was obtained by assuming imaging conditions analogous to those of Rayleigh's criterion. The proposed resolution measure shows that certain single-molecule experiments can be performed that were once unfeasible with optical microscope.

The optical microscope is a powerful instrument for observing cellular events. Recently, the increased use of microscopy in quantitative biological research, including single molecule microscopy, has generated significant interest in determining the performance limits of an optical microscope. Here, we formulate this problem in the context of a parameter estimation approach in which the acquired imaging data is modeled as a spatio-temporal stochastic process. We derive formulations of the Fisher information matrix for models that allow both stationary and moving objects. The effects of background signal, detector size, pixelation and noise sources are also considered. Further, formulations are given that allow the study of defocused objects. Applications are discussed for the special case of the estimation of the location of objects, especially single molecules. Specific emphasis is placed on the derivation of conditions that guarantee block diagonal or diagonal Fisher information matrices.

Mentoring is significant personal and professional assistance given by a more experienced person to a less experienced person during a time of transition. Transitions from high school to university, from university to graduate school are difficult. Organizing and running mentoring programs in schools or in professional societies provide good recruitment and retention of female students in engineering. biomedicalengineering (BME) is the engineering discipline that has the highest percentage of female degree recipients and tenure/tenure-track teaching faculty. Thus there is a great potential for female role models, mentors and mentees in BME. Recently, I have a developed a mentoring program for women at the jointgraduatebiomedicalengineeringprogram of University of Memphis (UM) and University of Tennessee (UT). Currently we are following the book "Giving Much/Gaining More: Mentoring for Success" by Dr. Wadsworth for our meetings and activities to provide a support and discussion group, and environment to women in their transition time of the BME graduate studies. Our future goal is to expand mentoring to female students in our engineering school.
