Ameliorative Effect of Combined Placenta-Derived Mesenchymal Stem Cells plus Platelet-rich Plasma on Polycystic Ovarian Model in Rats: A Biochemical and Histological Study

Polycystic ovary syndrome (PCOS) is a common cause of infertility in women, characterized by metabolic and hormonal irregularities. We investigated the effects of placenta-derived mesenchymal stem cells (PDMSCs) and platelet-rich plasma (PRP), as well as their combination on follicular development, hormonal profile, inflammatory parameters, and insulin resistance in a model of PCOS. In this study, 25 female Wistar rats were randomly allocated into five groups: Sham (given a dose of 1 mL of a 0.5% carboxymethylcellulose (CMC) solution), PCOS (administered 1 mg/kg of letrozole (LTZ) dissolved in CMC for 21 days), PDMSC (treated with a single intraovarian dose of PDMSCs), PRP (treated with a single intraovarian dose of PRP), and a combined PDMSC and PRP-treated group. After two weeks, serum and ovarian samples were collected for biochemical and histological analyses. Our results demonstrated that the simultaneous administration of PDMSCs and PRP had a synergistic effect compared to monotherapy, leading to an increase in estradiol (E2) and follicle-stimulating hormone (FSH) serum levels, a decrease in luteinizing hormone (LH) and testosterone levels, as well as inflammatory factors. Moreover, the combined therapy was associated with significantly lower levels of the homeostatic model of insulin resistance (HOMA-IR), fasting insulin (FINS), and blood glucose (FBG) compared to monotherapy. The combined treatment also caused a significant reduction in cystic follicles and an elevated number of corpus luteum, primordial, primary, secondary, and antral follicles. In conclusion, the combination of PRP and PDMSCs may have an ameliorative effect on modifying metabolic abnormalities and accelerating ovarian regeneration in PCOS.

Exposure of testicular tissue to supraphysiological concentrations of bile acids can lead to infertility. This study aimed to investigate the protective effects of heat-killed Lactobacillus reuteri (L. reuteri) against male reproductive toxicity induced by bile duct ligation (BDL). Thirty-two male Wistar rats were randomly assigned to four groups (N = 8): control normal (CN), sham-control (SC), BDL-control (underwent BDL surgery), and BDL + heat-killed L. reuteri (received probiotic supplementation for 7 days before and 21 days after BDL). At the end of the study, reproductive parameters, including sex hormones, sperm characteristics, oxidative stress markers, antioxidant status, and inflammatory gene expression in testicular tissue, were analyzed. Additionally, histological assessments of the testis and epididymis were performed. The administration of heat-killed L. reuteri significantly improved sperm viability (p <= 0.05) and luteinizing hormone levels (p <= 0.05) while reducing total oxidative status, carbonyl protein, and oxidative stress index (p <= 0.05) compared to the BDL-control group. Moreover, nitric oxide levels were significantly increased (p <= 0.05) in the probiotic-treated group. The expression levels of TNF-alpha and IL-6 were notably downregulated (p <= 0.05), indicating reduced testicular inflammation. Histological analysis demonstrated significant improvements in epithelial height, spermatogenic epithelial area ratio, lumen diameter, and lumen area (p <= 0.05) in the probiotic-treated group. The Effect of auriculotherapy on sleep quality in children with attention deficit hyperactivity disorder: a randomized clinical trial

Introduction Considering the importance of sleep disorders in children with attention-deficit/hyperactivity disorder (ADHD) and effective therapeutic strategies, the present study aimed to investigate the effects of auriculotherapy on sleep quality in children with ADHD. Materials and methods This clinical trial was conducted in children with ADHD in kashan, Iran, 2021-2022. Fifty-two eligible samples were selected using convenience sampling and randomly assigned to intervention and sham groups. The intervention group used Vacaria seeds to apply ear acupressure to Shenmen, Sympathetic, Subcortex, Heart, and Endocrine points for four weeks, while the sham group received adhesives without seeds and pressure. Sleep quality was assessed using children's sleep habits questionnaire at the beginning (T0), at the end of the intervention (T1), and one month later (T2). Data from 45 children (23 and 22 children in the intervention and sham groups, respectively) were analyzed using the per-protocol and intention-to-treat designs using repeated measures analysis of variance. Results The background variables did not significantly differ between two groups. The between-group analysis revealed a significant interaction effect of time and intervention on sleep quality (Effect Size = 0.545, p < 0.0001). The sleep quality score in the intervention group was significantly greater than in the sham group at T1 and T2 (p < 0.0001). The within-group analysis of the intervention group revealed a significant difference in sleep quality scores at three time points (Effect Size = 0.672, p < 0.05). In the sham group, sleep quality score increased significantly over time (Effect Size = 0.511, p < 0.05). Conclusions The findings suggest that auriculotherapy may be a beneficial complementary treatment for improving sleep quality in children with ADHD.

Hospital-acquired pneumonia (HAP) is the second most common cause of nosocomial infections and is responsible for 15% of nosocomial infections, with a high mortality rate, which has led to increased concern and significant costs in healthcare settings. The most significant agents of HAP are Pseudomonas aeruginosa and Klebsiella pneumoniae, which create a biofilm that results in a resistant infection. We aimed to study the synthesis of Cu2Ti2O5 nanoparticles, their effects on the growth and biofilms of Pseudomonas aeruginosa and Klebsiella pneumoniae isolated from respiratory infections, and their anticancer effects. In this study, for the first time, the Pechini method was used to synthesize Cu2Ti2O5 nanostructures. The effects of nanoparticles on the growth and biofilms of Pseudomonas aeruginosa and Klebsiella pneumoniae were evaluated using a microdilution broth and the microtiter plate method, and the cytotoxic effect of the nanoparticles on the A549 cell line was also assessed by MTT. The characteristics of the nanoparticles were confirmed through XRD, FTIR, SEM, and TEM techniques. Cu2Ti2O5 showed a minimum inhibitory effect in concentrations of 156.25 to 625 mu g mL-1 for ten isolates of K. pneumoniae and 625 to 1250 mu g mL-1 for ten isolates of P. aeruginosa and at sub-MIC concentrations as well. It reduced the biofilms of K. pneumoniae and P. aeruginosa strains by 75% and 44.4%. The nanoparticles killed 50% of A549 cancer cells in 48 h at concentrations of 30 to 40 mu g mL-1 and in 24 h at concentrations of 200 to 250 mu g mL-1. The findings of this study show the antibacterial, anti-biofilm, and anti-cancer effects of Cu2Ti2O5 nanoparticles. Therefore, these nanoparticles can be considered potential antimicrobial candidates;however, these effects should be confirmed with more bacterial isolates.

Colorimetric Assessment of Extracellular Electron Transfer in Bacteria: a Novel Approach for Evaluating Antibiotic Efficacy

A need exists for a rapid and straightforward method to measure antibiotic susceptibility in bacteria to optimize antibiotic usage and mitigate the emergence of antimicrobial resistance. In this study, we investigated a novel, facile, and rapid colorimetric method for assessing bacterial antibiotic susceptibility via the extracellular electron transfer (EET) assay. This method relies on the colorimetric detection of EET from bacterial respiratory metabolism to an EET reagent containing an electron transfer mediator (riboflavin) and a detectable electron acceptor (methylene blue). The minimum inhibitory concentration (MIC) values obtained for three real clinical nosocomial pathogens (Staphylococcus aureus, Pseudomonas aeruginosa, and Klebsiella pneumoniae) using our technique align closely with those derived from the gold standard micro-dilution broth assay. Employed riboflavin as an external mediator for electron transfer, enables to screen antibiotic susceptibility of bacterial strains with different capabilities of EET. Our innovative approach offers a rapid assay for identifying EET, and strain-specific effectiveness of antibiotics with diverse mechanisms of action. As we employed simple and cost-effective equipment, there is potential for this method to evolve into a practical approach for simplified antibiotic susceptibility testing.

Stakeholders' experiences, perceptions and satisfaction with an electronic appointment system: a qualitative content analysis

BackgroundOnline appointment scheduling systems have been designed and implemented to address barriers and problems related to in-person appointment scheduling;however, these systems also face challenges and issues that require continuous evaluation and resolution. This research aimed to investigate the experiences, perceptions and satisfaction of stakeholders with an electronic appointment system and identify its *** was a qualitative study conducted at a specialty Clinic in Iran during 2022-2023. A systematic purposive sampling method was used to select the participants. The participants included 10 administrative and executive users working at the Specialty Clinic, 8 physicians working at the clinic and 18 patients and visitors to the clinic. Data was collected through semi-structured face-to-face interviews. The interviews were analyzed using qualitative content *** findings derived from the semi-structured interview data revealed that the problems with the appointment system fell into two main themes: "Problems Related to Planning and Management of the Electronic Appointment System" and "Non-managerial Problems of the Electronic Appointment System". The problems related to planning and management were divided into three categories: national-level system management, university-level system management and clinic-level system management. The non-managerial problems were classified into two groups: functional problems and non-functional *** results of this study showed that the electronic appointment system, despite facilitating appointment processes, can be influenced by various factors and lead to stakeholder dissatisfaction. Preparation of ceramic nanocomposite and determination of its physical properties and cytotoxicity on stem cells from human exfoliated deciduous teeth

Aim: In this research, we made nano hydroxyapatite (HA) particles and combined them with graphene sheets. After determining physical properties of compound we checked its toxicity on stem cells derived from human exfoliated deciduous (SHED) cells obtained from the pulp of deciduous teeth using MTT test. Materials and methods: In this intervention study, nano HA was made using the co-precipitation method, and then a nano HA/graphene nanocomposite was created with the help of an ultrasonic bath. After examining their physical characteristics with a scanning electron microscope, X-ray diffraction, and elemental, analysis of their cytotoxicity were examined on SHED cells and compared with cold ceramics. To determine the cytotoxicity, stem cells were first extracted from the pulp of deciduous teeth and multiplied. Then, the toxicity of the mentioned substances on SHED cells was evaluated by the MTT test, and the percentage of cell viability compared to control cells was reported. Results: The XRD, SEM, and EDS results analysis confirm the structure, shape, and purity of the synthesized materials, respectively. Nano HA and HA/graphene nanocomposite had relatively low cytotoxicity, and both inhibited the growth of 50 % of cells at a concentration of 3 mg/ml. Concentrations below 1 mg/ml caused little cell death and almost no cytotoxicity. Also, cold ceramic at a concentration of 1.6 mg/ml inhibited the growth of 50 % of cells. The statistical comparison of the results showed that the survival percentage of the cells in the vicinity of the nanocomposite was significantly higher than that of the cold ceramic (P = 0.027). Conclusion: The results of this study on the physical characteristics of nano HAand its appropriate biocompatibility lead us to the conclusion that if additional studies are confirmed, ceramic nanocomposite can be used in various fields of dentistry.

Changes in Serum Immunoglobulin Levels Play as Predictors of Treatment Response and Prognosis in Pediatric Idiopathic Nephrotic Syndrome During the Remission Phase

Background Nephrotic syndrome is an immune-mediated renal disorder characterized by T-cell and B-cell dysfunctions with changes in immunoglobulin (Ig) levels and the IgG:IgM ratio. Therefore, this study aimed to determine whether the serum level of Igs can be considered as an index to predict the response to treatment and the prognosis of idiopathic nephrotic syndrome (INS) in children in the remission phase. Methods The study population consisted of 38 children with INS in the remission phase and 38 age- and sex-matched healthy children. Blood samples were collected from participants and serum values of IgG, IgM, IgE, and IgA were measured using EISA KITS from Aptech Services. The IgG:IgM ratio was studied in the participants. Results Patients significantly increased IgM and IgE levels compared with healthy subjects, unlike IgG and IgA values (p < 0.001-0.05). Patients with steroid-resistant nephrotic syndrome (SRNS) had a significant increase in IgM levels compared with those with steroid-sensitive nephrotic syndrome (SSNS) (p < 0.05). While, subjects with SRNS showed significant reductions in IgG and IgA values (p < 0.01). There were significant differences in the levels of IgG and IgM between steroid-sensitive patients with and without a history of relapse (p < 0.01). Furthermore, patients with steroid-independent and frequently relapsing NS showed a significant increase in IgE value compared with that of subjects with steroid-dependent and relapse (p < 0.05). The ratio of IgG/IgM was significantly reduced in patients compared with healthy individuals (p < 0.05). Other results indicated that there was a significant difference between patients with steroid-independent and steroid-dependent who had a history of relapse (p < 0.01). Conclusion Alterations in serum Ig values can be considered as predictors of treatment response and prognosis in pediatric idiopathic nephrotic syndrome during the remission phase.

BackgroundPatient-centered, measurable, and transparent care is essential for improving healthcare outcomes, particularly for patients undergoing percutaneous coronary intervention (PCI) procedures. Electronic follow-up questionnaires offer the potential for efficient and accurate data collection, enhancing the monitoring of patient experiences and outcomes. This study aimed to design and evaluate an electronic follow-up questionnaire tailored for post-PCI patients, focusing on real-time symptom monitoring and data *** developmental study was conducted in 2020 in three phases. In the first phase, a follow-up questionnaire was developed through a needs assessment and expert consultations. Each item's content validity ratio (CVR) and content validity index (CVI) were evaluated to ensure content validity. The finalized questionnaire elements were then reviewed and refined by a panel of ten cardiologists using the Delphi technique. In the second phase, an electronic platform was designed to host the follow-up questionnaire. The tool's effectiveness for post-PCI follow-up was evaluated in the third *** confirmed all items in the Delphi technique's first round, validating the follow-up questionnaire's content. A total of 41 patients undergoing PCI were enrolled in the study. The most frequently reported symptoms included issues at the catheter insertion site, chest discomfort, digestive complications, and shortness of breath. Of these patients, 21 (51.2%) utilized the electronic follow-up tool. The primary reasons for non-participation were busy schedules, forgetfulness, and perceived recovery. Investigating the effect of the orientation program on the performance of academic advisor

BackgroundTeacher-student interactions and academic advisors are critical for student success and have positive effects on academic integrity. On the other hand, studies show that the performance of the academic advisors has not been able to gain the satisfaction of the students. Therefore, this study was conducted to investigate the effect of the orientation program on the performance of academic advisors from the perspective of students and advisor professors. Materials and methodsThis quasi-experimental study was conducted, before and after the intervention in 2022-2023 in kashan School of Nursing and Midwifery. Data were collected from 220 students of different fields who were selected through stratified random sampling and 20 faculty advisors through a questionnaire of individual characteristics and two questionnaires "Evaluation of the academic advisor performance from the perspective of himself and the perspective of the student". The intervention of this research included five 30 to 40-minute training sessions as a group for academic advisors. Data were analyzed using descriptive statistics (mean, standard deviation) and analytical statistics (repeated-measures ANOVA) via SPSS version 22 software. ResultsThe results of the repeated-measures ANOVA showed that the mean score of advisor performance from the perspective of students was 88.31 +/- 46.70 before the intervention, which reached 114.09 +/- 45.79 and 99.76 +/- 37.53, respectively, 6 and 12 weeks after the implementation of the orientation program. Also, the mean score of advisor performance from their perspective before the intervention was 118.13 +/- 35.89 before the intervention, and 6 and 12 weeks after the implementation of the orientation program, it reached 160.38 +/- 17.23 and 168.75 +/- 8.10, respectively.
