A highly transparent dopamine-copolymerized hydrogel with enhanced ROS-scavenging and tissue-adhesive properties for chronic diabetic wounds

Chronic diabetic wounds with complex symptoms represent a major challenge in clinical practice, causing a serious threat to human health and life. Excessive oxidative stress and persistent inflammatory responses are the important reasons for the long-term difficult healing of diabetic wounds. Designing wound dressing materials with intrinsic antioxidant performance, high transparency, adhesiveness, and favorable mechanical properties is of great significance for promoting wound healing, especially in movable parts. Here, a dopamine-copolymerized highly transparent antioxidant hydrogel was developed for the treatment of chronic diabetic wounds. The hydrogel was easily prepared via free radical polymerization using acrylated dopamine monomer (ADA), acrylamide (AM), and phenylboronic acid modified dextran (DP). The dynamic phenylborate ester bonds formed between the catechol of polydopamine and phenylboronic acid effectively mitigated the darkening of the hydrogel color caused by the auto-oxidation of catechol, resulting in the PAM/PDA/DP hydrogel (DP3) with durable transparency. In addition, this hydrogel had good adhesiveness and mechanical properties, as well as desirable reactive oxygen species (ROS)-scavenging performance. Furthermore, in vivo results demonstrated that DP3 hydrogel can stimulate the polarization of macrophages toward anti-inflammatory M2 phenotype, increase the secretion of anti-inflammatory factors, so as to smooth the transition of wound healing from the inflammatory phase to the proliferative phase, and accelerate the repair of diabetic wounds by promoting angiogenesis and collagen deposition. Therefore, the DP3 hydrogel holds great potential for remolding the tissue regeneration microenvironment and serving as a promising dressing for chronic diabetic wounds. Effect of modified laparoscopic salpingectomy on ovarian reserve:Changes in the serum antimüllerian hormone levels

Objective:To retrospectively estimate the impact of modified laparoscopic salpingectomy on the ovarian reserve in infertile ***:There were 74 infertile women undergone modified laparoscopic salpingectomy from June 2016 to January 2017,and their levels of serum antimüllerian hormone,basal follicle-stimulating hormone and estradiol were reviewed ***:No significant change was detected in serum antimüllerian hormone at 3 months after surgery compared to preoperative level(p=0.857).Similar changes were observed for the basal follicle-stimulating hormone(p=0.102)and estradiol(p=0.233)***:Our results revealed that modified laparoscopic salpingectomy might be a valuable treatment for hydrosalpinx before in vitro fertilization.

Autophagy can be dynamically induced in response to stresses and is an essential, ubiquitous intracellular recycling system that impacts the fate of damaged resident cells, thereby influencing wound healing. Endometrial fibrosis is a form of abnormal wound healing that causes intrauterine adhesion (IUA) and infertility. We previously demonstrated that overactivated sonic hedgehog (SHH) signaling exacerbated endometrial fibrosis, but the role of autophagy in this process is still unknown. Here, we report that impaired autophagy participates in SHH pathway-induced endometrial fibrosis. Endometrial stroma-myofibroblast transition accompanied by autophagy dysfunction was present in both endometrial biopsies of IUA patients and Amhr2cre/+ R26-SmoM2+/− (AM2) transgenic mouse. Mechanistically, SHH pathway negatively regulated autophagy through pAKT-mTORC1 in a human endometrial stromal cell line (T-HESCs). Furthermore, SHH pathway-mediated fibrosis was partly counteracted by autophagy modulation in both T-HESCs and the murine IUA model. Specifically, the impact of SHH pathway inhibition (GANT61) was reversed by the pharmacological autophagy inhibitor chloroquine (CQ) or RNA interference of autophagy-related gene ATG5 or ATG7. Similar results were obtained from the murine IUA model treated with GANT61 and CQ. Moreover, promoting autophagy with rapamycin reduced fibrosis in the AM2 IUA model to baseline levels. In summary, defective autophagy is involved in SHH pathway-driven endometrial fibrosis, suggesting a potential novel molecular target for IUA treatment.

Objective:To evaluate cervical function and pregnancy outcomes after hysteroscopic resection of the complete uterine septum,duplicate cervix and vaginal septum with Metzenbaum ***:Between January 2010 and December 2016,13 patients admitted to the Department of Obstetrics and Gynecology of Sir Run Run Shaw Hospital with complete uterine septum,duplicate cervix and vaginal septum,were enrolled into this *** cervical and corporal septum was cut by Metzenbaum scissors,and residual septum was cut by micro scissors under *** vaginal septum was cut with the unipolar electric ***:The operation time was about 10±1.31 *** the 13 patients present normal uterine cavities without scar formation under hysteroscopy at 3 months after operation,there was mild adhesions between anterior and posterior intrauterine wall on 2 *** operation,there were 13 pregnancies naturally conceived in 11 patients,10 *** live birth rate was 76.92%,the early miscarriage rate was 23.08%.The cesarean section ratewas 30%,the vaginal delivery ratewas 70%,and all were term ***:The operation was simple,convenient,and fast,without any complications and cervical *** was easy to have vaginal deliveries.

There are few reports of the successful delivery of unicornuate uterus pregnancy after laparoscopic cervical ***,we report a 32-year-old woman,gravida 2 para 1,who had been admitted for unicornuate uterus pregnancy by a thawed frozen blastocyst *** accepted laparoscopic cervical cerclage for cervical insufficiency and delivered successfully via caesarean section at 35^(+3) *** cerclage may be used as an effective method of preventing abortion in unicornuate uterus pregnancy,while surgery by laparoscopy would be a better choice for patients with poor cervical *** should pay due attention to complications such as uterine rupture for these highrisk patients.

Stem cell-based and stem cell-derived exosome-based therapies have shown promising potential for endometrial regeneration and the clinical treatment of intrauterine adhesions(IUAs).Evidence shows that apoptosis occurs in a majority of grafted stem cells,and apoptotic bodies(ABs)play a critical role in compensatory tissue ***,the therapeutic potential of AB-based therapy and its mechanism have not been explored in ***,a cell-free therapeutic strategy was developed by incorporating mesenchymal stem cell-derived ABs into a hyaluronic acid(HA)hydrogel to achieve endometrial regeneration and fertility ***,we found that the ABs could induce macrophage immunomodulation,cell proliferation,and angiogenesis in *** HA hydrogel promoted the retention of ABs and facilitated their continuous *** a murine model of acute endometrial damage and a rat model of IUAs,in situ injection of the AB-laden HA hydrogel could efficiently reduce fibrosis and promote endometrial regeneration,resulting in the fertility ***,ABs show good potential as therapeutic vesicles,and the AB-laden HA hydrogel appears to be a clinically feasible and cell-free alternative for endometrial regeneration and IUA treatment.

NLRP3 inflammasome:a new therapeutic target for high-risk reproductive disorders?
