The single-detector spectrometers based on 2D layer van der Waals (vdW) heterojunctions offer advantages in spectral reconstruction due to their high sensitivity, tunable optical properties, and the ability to cover a broad spectral range. There exist two principal algorithms dominating spectrum reconstruction for this kind spectrometer: the Tikhonov regularization method combined with the Least Squares Method (LSM) and neural network-based approaches, particularly Deep Learning (DL). However, both of the algorithms exhibit inherent limitations in spectral reconstruction, which constrain the versatility of computational spectrometers that rely solely on a single algorithm for reconstructing diverse spectral profiles. To overcome this limitation, we introduce an artificial neural network (ANN)-based classification model capable of dynamically selecting the optimal algorithm throughout the reconstruction process. This enables highly accurate spectral reconstruction within the 440-700 nm wavelength range, achieving a spectral resolution of 6 nm. By harnessing the complementary strengths of multiple algorithms, our approach proposes a novel strategy for combining techniques to enhance the precision of spectral reconstructions, laying the groundwork for more sophisticated methods in the future. © 1980-2012 IEEE.

Scene flow estimation (SFE) and motion segmentation (MOS) using 4D radar are emerging yet challenging tasks in robotics and autonomous driving applications. Existing LiDAR- or RGB-D-based point cloud processing methods often deliver suboptimal performance on radar data due to radar signals' highly sparse, noisy, and artifact-prone nature. Furthermore, for radar-based SFE and MOS, the lack of annotated datasets further aggravates these challenges. To address these issues, we propose a novel self-supervised framework that exploits denoising diffusion models to effectively handle radar noise inputs and predict point-wise scene flow and motion status simultaneously. To extract key features from the raw input, we design a transformer-based feature encoder tailored to address the sparsity of 4D radar data. Additionally, we generate self-supervised segmentation signals by exploiting the discrepancy between robust rigid ego-motion estimates and scene flow predictions, thereby eliminating the need for manual annotations. Experimental evaluations on the View-of-Delft (VoD) dataset and TJ4DRadSet demonstrate that our method achieves state-of-the-art performance for both radar-based SFE and MOS. © 2016 IEEE.

An automatic annotation tool and reference database for T cell subtypes and states at single-cell resolution

T cells have various subtypes and states with different functions. However, a reference list and automated annotation tool for T cell subtypes and states are lacking, which is critical for analyzing and comparing T cells under various conditions. We constructed the largest human T cell reference, containing 1,348,268 T cells from 35 conditions and 16 tissues. We classified T cells into 33 subtypes and further stratified them into 68 categories according to subtype and state. Based on this reference, we developed a tool named STCAT to automatically annotate T cells from scRNA-seq data by hierarchical models and marker correction. The accuracy of STCAT was 28% higher than that of existing tools validated on six independent datasets, including cancer and healthy samples. Using STCAT, we consistently discovered that CD4+ Th17 cells were enriched in late-stage lung cancer patients in multiple datasets, whereas MAIT cells were prevalent in milder-stage COVID-19 patients. We also confirmed a decrease in Treg cytotoxicity in post-treatment ovarian cancer. Systematic landscape analyses of CD4+ and CD8+ T cell references revealed that CD4+ Treg cells were enriched in tumor samples and that CD8+ naive-related cells were abundant in healthy individuals. Finally, we deposited all the T cell references and annotations into a TCellAtlas (https://***/TCellAtlas) database, which allows users to browse T cell expression profiles and analyze customized scRNA-seq data by STCAT. In conclusion, comprehensive human T cell subtypes and states reference, automated annotation tool, and database will greatly facilitate research on T cell immunity and tumor immunology. © 2025 science China Press

Nanopore sequencing has become the primary technology for modern DNA sequencing. Solid nanopores are favored for their cost-effectiveness, stability, and scalability, particularly for portable applications. This paper presents a new design for a portable solid-state nanopore sequencing device based on the Linux platform. It employs a cutting-edge biological sequencing chip with a nanopore channel to capture through-hole data. Electrical signals from the chip are processed to create DNA sequence data files. The Linux-based Raspbian OS serves as the control terminal, facilitating seamless communication between components. Data is then transmitted to a cloud platform via a 5G unit for further analysis. This study integrates software and hardware for instrument validation. © The Author(s) 2025.

Traditional cell imaging is limited to 3D surface imaging and has a relatively low resolution. A phase microscopy technique based on transmission matrix has been proposed for high-resolution tomographic imaging within cells. This method requires only a single measurement with the phase microscope to obtain the phase information from the cell and construct a transmission matrix. By analyzing the singular values of the transmission matrix, this method enables the extraction of structural information at different depths. The effectiveness of the technique was validated through Monte Carlo simulations. Experimental results with red blood cells revealed intracellular structures, achieving an axial resolution of 5 nm. Additionally, the application of this technique in distinguishing white blood cell types demonstrates its ability to effectively image and identify unique structural characteristics of cells. Combining the white light diffraction phase microscope with singular value decomposition based on the transmission matrix, lateral resolution can be achieved within 100 nm. Experimental results highlight the potential of this method in distinguishing subtle cellular components and structural variations, making it a valuable tool for cell biology research and medical diagnostics. This innovative technique provides a highly efficient and non-invasive approach for high-resolution cell imaging, with promising applications in medical diagnostics and biological research. © 2025 Optica Publishing Group under the terms of the Optica Open Access Publishing Agreement.

Covert communication is considered a promising technology for hiding transmission processes and activities from malicious eavesdropping. With the development of detection technology, the traditional point-to-point covert communication method cannot effectively guarantee the reliability of covert communication. In order to solve this problem, unmanned aerial vehicle (UAV) are widely adopted in covert communication as common relay equipment. However, the UAV maximizes its benefits by taking advantage of the information asymmetry between itself and the covert communication sender. Therefore, it is necessary to encourage UAV-assisted potential users (UPU) to transmit covert data honestly by designing an information asymmetry-based incentive mechanism. In this paper, we propose a UAV-assisted contract-theoretic (UACT) model to improve the covert communication performance, where UPU honestly transmits covert data and the throughput of UACT can be increased. Specifically, we first divide UPU into finite types and model the data transfer contract between the sender, UPU, and receiver. Then, the contract feasibility of UACT is derived with sufficient and necessary conditions. Finally, we analyze the contract optimization in the continuous case of UPUs. Numerical simulations show that the UACT can incentivize UPUs to transmit covert data through UAV assistance and make covert communication performance close to the information symmetry case. © 1967-2012 IEEE.

Administration of Bifidobacterium animalis Subsp. lactis BLa80 and Lactobacillus acidophilus LA85 Improved Hyperglycemia and Modulated Gut Microbiota in Type 2 Diabetic Mice

Type 2 diabetes mellitus (T2DM) is a prevalent metabolic disorder and constitutes a significant threat to global public health. Increasing evidence has shown the therapeutic potential of probiotics in the management of T2DM. This study established a T2DM mouse model through high-fat diet combined with streptozotocin injection (HFD/STZ) and investigated the preventive effects of two probiotic strains: Bifidobacterium animalis subsp. lactis BLa80 and Lactobacillus acidophilus LA85. The results indicated that both probiotic strains significantly improved glucose homeostasis by reducing fasting blood glucose (FBG) levels, enhancing insulin sensitivity, and increasing glucagon-like peptide-1 (GLP-1) levels. Moreover, probiotics decreased blood lipid and pro-inflammatory mediator levels, enhanced the production of anti-inflammatory cytokines, and mitigated pathological alterations in ileal, hepatic, pancreatic, and renal tissues. Subsequent 16S rRNA amplicon sequencing analysis revealed that BLa80 and LA85 interventions effectively modulated gut microbiota composition, particularly by increasing the relative abundance of short-chain fatty acids (SCFAs)-producing bacterial taxa. Notably, the mechanisms of action were strain-specific: BLa80 primarily impacted glycemic control and promoted the proliferation of Bifidobacterium and Limosilactobacillus, whereas LA85 exhibited superior efficacy in regulating lipid metabolism and promoted the growth of Lactobacillus and Alistipes populations. These findings indicate that BLa80 and LA85 can ameliorate symptoms related to T2DM despite their distinct regulatory pathways, suggesting their potential as therapeutic agents in diabetes management. © The Author(s), under exclusive licence to Springer science+Business Media, LLC, part of Springer Nature 2025.
