NQO1 Mediates Lenvatinib Resistance by Regulating ROS-induced Apoptosis in Hepatocellular Carcinoma

Objective Hepatocellular carcinoma(HCC)is the third leading cause of cancer-associated death *** a first-line drug for advanced HCC treatment,lenvatinib faces a significant hurdle due to the development of both intrinsic and acquired resistance among patients,and the underlying mechanism remains largely *** present study aims to identify the pivotal gene responsible for lenvatinib resistance in HCC,explore the potential molecular mechanism,and propose combinatorial therapeutic targets for HCC *** Cell viability and colony formation assays were conducted to evaluate the sensitivity of cells to lenvatinib and ***-Seq was used to determine the differences in transcriptome between parental cells and lenvatinib-resistant(LR)*** upregulated genes were analyzed by GO and KEGG ***,qPCR and Western blotting were employed to determine the relative gene expression ***,the intracellular reactive oxygen species(ROS)and apoptosis were detected by flow *** PLC-LR and Hep3B-LR were *** was a total of 116 significantly upregulated genes common to both LR cell *** GO and KEGG analyses indicated that these genes were involved in oxidoreductase and dehydrogenase activities,and reactive oxygen species ***,NAD(P)H:quinone oxidoreductase 1(NQO1)was highly expressed in LR cells,and was involved in the lenvatinib *** high expression of NQO1 decreased the production of ROS induced by lenvatinib,and subsequently suppressed the *** combination of lenvatinib and NQO1 inhibitor,dicoumarol,reversed the resistance of LR *** The high NQO1 expression in HCC cells impedes the lenvatinib-induced apoptosis by regulating the ROS levels,thereby promoting lenvatinib resistance in HCC cells.

Grasping is a critical motor skill essential for daily activities, but it is often compromised in individuals with neural impairments. Functional Electrical Stimulation (FES) has emerged as a promising intervention, utilizing electrical pulses to stimulate muscles and thereby restore impaired motor functions. However, existing closed-loop FES systems depend on pre-calibrated angles or forces specific to individual objects, which limits their practicality in dynamic, real-world environments with varying object properties. This paper presents a novel closed-loop FES (CLFES) system with visual feedback, designed to dynamically adjust stimulation parameters based on real-time interaction states without requiring object-specific calibration. The system employs a finite state machine to manage sequential grasp-release tasks and integrates a visual perception module for slip detection and intent recognition. The system was tested with two individuals with disabilities on five common household objects. Experimental results demonstrate significant improvements, including a 42.6% increase in success rate and a 45.9% reduction in task completion time compared to tasks performed without the system. These results underscore the system's potential to improve daily task performance for individuals with neural impairments. © 2018 IEEE.

Immune checkpoint inhibitors(ICIs)therapy targeting programmed cell death ligand 1(PD-L1)and programmed death protein 1(PD-1)had exhibited significant clinical benefits for cancer treatment such as triple negative breast cancer(TNBC).However,the relatively low anti-tumor immune response rate and ICIs drug resistance highlight the necessity of developing ICIs combination therapy strategies to improve the anti-tumor effect of ***,the immunomodulator epigallocatechin gallate palmitate(PEGCG)and the immunoadjuvant metformin(MET)self-assembled into tumor-targeted micelles via hydrogen bond and electrostatic interaction,which encapsulated the therapeutic agents doxorubicin(DOX)-loaded PEGCG-MET micelles(PMD)and combined with ICIs(anti-PD-1 antibody)as therapeutic strategy to reduce the endogenous expression of PD-L1 and improve the tumor immunosuppressive *** results presented that PMD integrated chemotherapy and immunotherapy to enhance antitumor efficacy in vitro and in vivo,compared with DOX or anti-PD-1 antibody for the therapy of *** micelles might be a potential candidate,which could remedy the shortcomings of antibody-based ICIs and provide synergistic effect to enhance the antitumor effects of ICIs in tumor therapy.

Biochar as an adsorbent is paid more attention for treating uranium-containing wastewater. In this work, a novel microalgae-derived biochar(A-HTC) was synthesized by dual-template one-step carbonization for uranium adsorption. This biochar exhibited a maximum adsorption capacity of 472.3 mg·g−1. Especially, A-HTC demonstrated excellent selectivity for uranium over 11 competing cations and remarkable reusability after 10 adsorption-desorption cycles. The adsorption behavior with time and uranium concentration followed the pseudo-second-order kinetic model and the Langmuir isotherm model, respectively. Moreover, the adsorption process was spontaneous and endothermic. Zeta potential, XPS, and FTIR analyses revealed that A-HTC adsorbs U(VI) mainly through electrostatic attraction and complexation. The present study offers an innovative adsorbent and a novel method to produce biochar for uranium recovery from aquatic environments. © 2025 The Korean Society of Industrial and Engineering Chemistry

The human-derived novel gut commensal Luoshenia tenuis regulates body weight and food intake in mice

Xenopus has proven to be a remarkably versatile model organism in the realm of biological research for numerous years, owing to its straightforward maintenance in laboratory settings and its abundant provision of ample-sized oocytes, eggs, and embryos. The cell cycle of these oocytes, eggs, and early embryos exhibits synchrony, and extracts derived from these cells serve various research purposes. Many fundamental concepts in biochemistry, cell biology, and development have been elucidated through the use of cell-free extracts derived from Xenopus cells. Over the past few decades, a wide array of cell-free extracts has been prepared from oocytes, eggs, and early embryos of different Xenopus species at varying cell cycle stages. Each of these extracts possesses distinct characteristics. This review provides a concise overview of the Xenopus species employed in laboratory research, the diverse types of cell-free extracts available, and their respective properties. Furthermore, this review delves into the extensive investigation of spindle assembly in Xenopus egg extracts, underscoring the versatility and potency of these cell-free systems in the realm of cell biology.

Enhancing biofilm formation in the hydrogen-based membrane biofilm reactor through bacterial Acyl-homoserine lactones

The slow growth rate of autotrophic bacteria and regulation of biofilm thickness are critical factors that limit the development of a hydrogen-based membrane biofilm reactor(H_(2)-MBfR).The acylhomoserine lactone(AHL)mediated quorum sensing(QS)system is a crucial mechanism regulating biofilm ***,the AHLs that promote biofilm formation in autotrophic denitrification systems and their underlying mechanisms,remain *** study explored the impact of AHLmediated QS signaling molecules on biofilm development in H_(2)-*** study revealed that C_(14)-HSL and C_(4)-HSL are potential signaling molecules that enhanced biofilm formation in long-term stable operating H_(2)-*** short-term experiments with C_(14)-HSL and C_(4)-HSL confirmed their ability to increase bacterial adhesion to carrier surfaces by promoting the production of extracellular polymeric substances(EPS).Functional gene annotation indicated that exogenous C_(14)-HSL and C_(4)-HSL increased the abundance of signal transduction(increased by 0.250%–0.375%),strengthening the inter bacterial QS response while enhancing cell motility(increased by 0.24%and 0.21%,respectively)and biological adhesion(increased by 0.044%and 0.020%,respectively),thereby accelerating the initial bacterial attachment to hollow fiber membranes and facilitating biofilm *** findings contribute to the understanding of microbial community interactions in H_(2)-MBfRs and provide novel approaches for biofilm management in wastewater treatment systems.
