Decoding transcriptional regulation via a human gene expression predictor

Transcription factors(TFs)regulate cellular activities by controlling gene expression,but a predictive model describing how TFs quantitatively modulate human transcriptomes is *** construct a universal human gene expression predictor named EXPLICIT-Human and utilize it to decode transcriptional *** the expression of 1613 TFs,the predictor reconstitutes highly accurate transcriptomes for samples derived from a wide range of tissues and *** broad applicability of the predictor indicates that it recapitulates the quantitative relationships between TFs and target genes ubiquitous across *** interacting TF-target gene pairs are extracted from the predictor and enable downstream inference of TF regulators for diverse pathways involved in development,immunity,metabolism,and stress response.A detailed analysis of the hematopoiesis process reveals an atlas of key TFs regulating the development of different hematopoietic cell lineages,and a portion of these TFs are conserved between humans and *** results demonstrate that our method is capable of delineating the TFs responsible for fate *** to other existing tools,EXPLICIT-Human shows a better performance in recovering the correct TF regulators.

As a hydrocarbon-rich sedimentary basin in China,the Ordos Basin has enormous potential for shale gas *** shale of the Upper Carboniferous Benxi Formation is rich in organic matter,however,its palaeoenvironment and organic matter enrichment mode are yet to be *** this study,the geochemical characteristics of the shale of the Benxi Formation in the east-central part of the Ordos Basin were analyzed to investigate its *** the same time,the organic matter enrichment modes in different sedimentary facies were compared and *** results indicate that:1)the shale of the Benxi Formation was mainly deposited on the continental margin and strong terrestrial clastic input;2)the deposition period of the Benxi Formation shale had a hot and humid climate with high palaeoproductivity and local volcanic hydrothermal fluid,and a high sedimentation rate with the strong stagnant *** bottom water was in dysoxic conditions and a semi-saline deposition environment;3)multiple factors,such as palaeoproductivity,volcanic hydrothermal,redox conditions,and palaeosalinity interact to influence the enrichment of shale organic matter in Benxi Formation;4)the organic matter enrichment modes of continental,marine-continental transitional,and marine shales can be classified into three types:“production mode”,“hybrid mode of preservation and production”,and“preservation mode”,*** study provides a reference for the organic matter enrichment mode,shale gas formation conditions,and core area evaluation in these marine-continental transitional shales,and also offers new guidance for exploration ideas for shale gas in different sedimentary facies.

To address issues of insufficient sensitivity and weak anti-noise characteristics in existing image sharpness evaluation algorithms, we propose a method combining local variance and gradient analysis. Traditional methods like the Tenengrad function use horizontal and vertical gradient values, while multi-directional gradients can increase noise sensitivity and false detections. Our approach integrates the Tenengrad function with gray variance processing, analyzing the deviation of each pixel's brightness from the average to distinguish content from background. Normalizing these values helps reduce background and impurity pixel influence. Preprocessing with an anisotropic diffusion filter smooths the image while preserving edges. Experimental results show this method offers higher sensitivity and better noise resistance than traditional functions, suitable for autofocus systems. © 2024 IEEE.

The combinational chemo-immunotherapy as a novel treatment strategy has been widely studied and applied in clinic to enhance antitumor therapeutic efficacy and relieve side *** interference(RNAi)targeting PD-L1 via inhibiting novo production of PD-L1 will overcome the innate and adaptive PD-L1 expression during chemotherapy,thus enable sustained and efficient immune checkpoint blockade(ICB)to active antitumor immune ***,we designed a glutathione(GSH)-responsive camptothecin(CPT)prodrug-based hybrid micellar nanoparticles(siPD-L1@HM-CPT)to achieve synergistic antitumor chemoimmunotherapy by PD-L1 ***-L1@HM-CPT derived from the one-step loading PD-L1 siRNA(siPD-L1)into the CPT prodrug-based hybrid micelles(HM-CPT)which were co-assembled from biodegradable polyphosphoesters-based prodrug CPT-ss-PAEEP15 and stabilizer DSPE-PEG,showed high loading efficiency,GSH-responsive drug release,and excellent stability and ***-L1@HM-CPT achieved simultaneously the co-delivery of CPT and siPD-L1 in vitro and in vivo,high accumulation at the tumor sites,and rapid intracellular release to promote antitumor efficacy via sensitizing CPT chemotherapy,inducing strong immunogenic cell death(ICD)and sustained ICB to improve intratumoral CD8+T cells *** addition,the antitumor immunity response limited by the differentiated immunogenicity,intrinsic PD-L1 expression,and intracellular GSH level was facilitated by efficient ICD and ICB from silencing PD-L1 and synergistic CPT chemosensitization in our experimental B16-F10 and 4T1 tumor *** study might offer a perspective on designing novel co-delivery nanoparticles by convenient and controllable preparation for antitumor chemo-immunotherapy.

Designing cathode possessing crystalline@amorphous core-shell structure with both active core and shell is a meaningful work for resolving the low speciﬁc capacity,unstable cycling performance and sluggish reaction kinetics issues of rechargeable magnesium batteries(RMBs)by providing more active sites as well as releasing inner stress during ***,WO_(3)@WO_(3-x)S_(x) owning crystalline@amorphous core-shell structure containing both active core and active shell is constructed successfully by introducing S into metastable WO3 structure under temperatureﬁeld *** such structure,amorphous shell would provide continuous Mg^(2+)diffusion channels due to its isotropy property for most Mg^(2+)migrating rapidly to interface and then adsorb at ions reservoir formed by interfacial electricﬁeld for increasing speciﬁc *** also makes security for stable structure of WO_(3)@WO_(3-x)S_(x) by alleviating volume expansion of crystalline core WO_(3) during cycling to prolong cycling ***,“softer”ions S^(2-)would weaken interaction between hard acid Mg^(2+) and ionic lattice to enhance Mg^(2+)storage ***,WO_(3)@WO_(3-x)S_(x) delivers the superior cycling performance(1000 cycles with 83.3%),rate capability(88.5 mAh g^(-1) at 1000 mA g^(-1))and speciﬁc capacity(about 150 mAh g^(-1) at 50 mA g^(-1)),which is near 2 times higher than that of *** is believed that the crystalline@amorphous core-shell structure with both active core and shell designing via doping strategy is enlightening for the development of high-performance RMBs,and such design can be extended to other energy storage devices for better electrochemical performance.

Anchorene,identified as an endogenous bioactive carotenoid-derived dialdehyde and diapocarotenoid,affects root development by modulating auxin ***,the precise interaction between anchorene and auxin,as well as the mechanisms by which anchorene modulates auxin levels,remain largely *** this study,we conducted a comparative analysis of anchorene's bioactivities alongside auxin and observed that anchorene induces multifaceted auxin-like *** genetic and pharmacological examinations,we revealed that anchorene's auxin-like activities depend on the indole-3-pyruvate-dependent auxin biosynthesis pathway,as well as the auxin inactivation pathway mediated by Group II Gretchen Hagen 3(GH3)proteins that mainly facilitate the conjugation of indole-3-acetic acid(IAA)to amino acids,leading to the formation of inactivated storage *** measurements indicated that anchorene treatment elevates IAA levels while reducing the quantities of inactivated IAA–amino acid conjugates and ox *** sequencing further revealed that anchorene triggers the expression of numerous auxin-responsive genes in a manner reliant on Group II ***,our in vitro enzymatic assays and biolayer interferometry(BLI)assay demonstrated anchorene's robust suppression of GH3.17-mediated IAA conjugation with ***,our findings highlight the significant role of carotenoid-derived metabolite anchorene in modulating auxin homeostasis,primarily through the repression of GH3-mediated IAA conjugation and inactivation pathways,offering novel insights into the regulatory mechanisms of plant bioactive apocarotenoids.

Recombinant keratins possess strong hemostatic and wound healing properties but suffer from poor water solubility that restricts their bioactivities in biomedical ***,we report the rational design and synthesis of water-soluble keratins using a simple methodology named the QTY *** vitro biophysical analyses and molecular dynamic simulation demonstrated a 200-fold increase in the water solubility of QTY variant keratins without apparent structural changes compared to native *** self-assembly was observed for the first time in recombinant keratins in an aqueous environment,without urea and after QTY *** and animal experiments showed the in situ gel-forming capability of QTY variant keratins with superior hemostatic and wound healing activities at the wound sites compared to native recombinant *** work not only presented a simple and feasible pathway to produce large amounts of water-soluble keratins using QTY modification but also validated the enhanced self-assembly,hemostasis,and wound healing properties of these novel keratin species that may open up new venues for biomedical applications.
