Dear Editor,Learning-induced expression of activity-dependent genes,especially immediate-early genes(IEGs),is required for the conversion of experience-induced neuronal activity into long-term memory[1].Neuronal Per-Arnt-Sim domain protein 4(Npas4)is one of the most rapidly induced IEGs,functioning as a transcription factor to regulate the activity-dependent expression of other IEGs,such as activityregulated cytoskeleton associated protein(Arc),Fos protooncogene,and early growth response 1(Egrl)[1].

Designing cathode possessing crystalline@amorphous core-shell structure with both active core and shell is a meaningful work for resolving the low speciﬁc capacity,unstable cycling performance and sluggish reaction kinetics issues of rechargeable magnesium batteries(RMBs)by providing more active sites as well as releasing inner stress during ***,WO_(3)@WO_(3-x)S_(x) owning crystalline@amorphous core-shell structure containing both active core and active shell is constructed successfully by introducing S into metastable WO3 structure under temperatureﬁeld *** such structure,amorphous shell would provide continuous Mg^(2+)diffusion channels due to its isotropy property for most Mg^(2+)migrating rapidly to interface and then adsorb at ions reservoir formed by interfacial electricﬁeld for increasing speciﬁc *** also makes security for stable structure of WO_(3)@WO_(3-x)S_(x) by alleviating volume expansion of crystalline core WO_(3) during cycling to prolong cycling ***,“softer”ions S^(2-)would weaken interaction between hard acid Mg^(2+) and ionic lattice to enhance Mg^(2+)storage ***,WO_(3)@WO_(3-x)S_(x) delivers the superior cycling performance(1000 cycles with 83.3%),rate capability(88.5 mAh g^(-1) at 1000 mA g^(-1))and speciﬁc capacity(about 150 mAh g^(-1) at 50 mA g^(-1)),which is near 2 times higher than that of *** is believed that the crystalline@amorphous core-shell structure with both active core and shell designing via doping strategy is enlightening for the development of high-performance RMBs,and such design can be extended to other energy storage devices for better electrochemical performance.

Anchorene,identified as an endogenous bioactive carotenoid-derived dialdehyde and diapocarotenoid,affects root development by modulating auxin ***,the precise interaction between anchorene and auxin,as well as the mechanisms by which anchorene modulates auxin levels,remain largely *** this study,we conducted a comparative analysis of anchorene's bioactivities alongside auxin and observed that anchorene induces multifaceted auxin-like *** genetic and pharmacological examinations,we revealed that anchorene's auxin-like activities depend on the indole-3-pyruvate-dependent auxin biosynthesis pathway,as well as the auxin inactivation pathway mediated by Group II Gretchen Hagen 3(GH3)proteins that mainly facilitate the conjugation of indole-3-acetic acid(IAA)to amino acids,leading to the formation of inactivated storage *** measurements indicated that anchorene treatment elevates IAA levels while reducing the quantities of inactivated IAA–amino acid conjugates and ox *** sequencing further revealed that anchorene triggers the expression of numerous auxin-responsive genes in a manner reliant on Group II ***,our in vitro enzymatic assays and biolayer interferometry(BLI)assay demonstrated anchorene's robust suppression of GH3.17-mediated IAA conjugation with ***,our findings highlight the significant role of carotenoid-derived metabolite anchorene in modulating auxin homeostasis,primarily through the repression of GH3-mediated IAA conjugation and inactivation pathways,offering novel insights into the regulatory mechanisms of plant bioactive apocarotenoids.

Recombinant keratins possess strong hemostatic and wound healing properties but suffer from poor water solubility that restricts their bioactivities in biomedical ***,we report the rational design and synthesis of water-soluble keratins using a simple methodology named the QTY *** vitro biophysical analyses and molecular dynamic simulation demonstrated a 200-fold increase in the water solubility of QTY variant keratins without apparent structural changes compared to native *** self-assembly was observed for the first time in recombinant keratins in an aqueous environment,without urea and after QTY *** and animal experiments showed the in situ gel-forming capability of QTY variant keratins with superior hemostatic and wound healing activities at the wound sites compared to native recombinant *** work not only presented a simple and feasible pathway to produce large amounts of water-soluble keratins using QTY modification but also validated the enhanced self-assembly,hemostasis,and wound healing properties of these novel keratin species that may open up new venues for biomedical applications.

Alternative polyadenylation(APA)is a post-transcriptional process that typically determines the length of mature mRNAs'3'untranslated region(3'UTR).The global shortened 3'UTR alters mRNA stability,localization,and translation and contributes to cancer progression.1 The APA characteristics and its effects on acute myeloid leukemia(AML)remain to be comprehensively ***,we identified churchill domain containing 1(CHURC1)as an upregulated gene with a lengthened 3'UTR in CD34+enriched AML cells compared with healthy hematopoietic stem and progenitor *** for APA regulation of CHURC1,we found that rs6745 and RNA binding protein TIA1 potentially contribute to the distal poly(A)site usage in cis-and transways,***,a lengthened CHURC13'UTR affected the apoptosis of AML cells by disrupting the expression of DICER1 by sponging *** chemotherapeutic sensitivity of 8 drugs was associated with CHURC1 poly(A)site ***,our analysis indicated a previously undetected gene,CHURC1,that affected AML progression through 3'UTR length change resulting from *** collection and detailed analysis methods were described in the supplementary material.

The phytohormone auxin plays central roles in many growth and developmental processes in *** of chemical tools targeting the auxin pathway is useful for both plant biology and *** we reveal that naproxen,a synthetic compound with anti-inflammatory activity in humans,acts as an auxin transport inhibitor targeting PIN-FORMED(PIN)transporters in *** experiments indicate that exogenous naproxen treatment affects pleiotropic auxin-regulated developmental *** cellular and biochemical evidence indicates that naproxen suppresses auxin transport,specifically PIN-mediated auxin ***,biochemical and structural analyses confirm that naproxen binds directly to PIN1 protein via the same binding cavity as the indole-3-acetic acid ***,by combining cellular,biochemical,and structural approaches,this study clearly establishes that naproxen is a PIN inhibitor and elucidates the underlying *** use of this compound may advance our understanding of the molecular mechanisms of PIN-mediated auxin transport and expand our toolkit in auxin biology and agriculture.

Migration pattern of a population of Barn Swallows(Hirundo rustica)breeding in East Asian tropical region

Birds exhibit a high degree of migratory diversity,which is influenced by various ecological factors and life history *** studies on tropical bird migration,of which research is scarce,and comparing it with temperate birds can enhance our understanding of bird migration behaviour and its underlying *** this study,we explored the migration behaviour of a breeding population of the Barn Swallow(Hirundo rustica)in Zhanjiang,southern China,a region located in the northern tropics,using light-level *** 2021 to 2023,we deployed geolocators on 92 breeding swallows and retrieved geolocators successfully from 23 *** swallows all exhibited migratory behaviour,and wintering on various islands in Southeast *** displayed sex differences in their wintering *** males concentrated in Borneo,while females primarily chose Borneo but also dispersed to the Philippines,South China Sea,and Vietnam for *** studied swallow population adopted a seasonal migration pattern of“indirect in autumn,direct in spring”,bypassing the ecological barrier of the South China Sea in autumn and tending to directly cross it in spring ***,the distance and duration of autumn migration was significantly longer than those of the spring *** to temperate Barn Swallows,the Barn Swallow population breeding in Zhanjiang adopts a pattern of“intra-tropical migration”and initiates autumn migration *** formation of their migration pattern may be limited by ecological and physiological factors.

Mechanisms of PiT2-loop7 Missense Mutations Induced Pi Dyshomeostasis

PiT2 is an inorganic phosphate(Pi)transporter whose mutations are linked to primary familial brain calcification(PFBC).PiT2 mainly consists of two ProDom(PD)domains and a large intracellular loop region(loop7).The PD domains are crucial for the Pi transport,but the role of PiT2-loop7 remains *** PFBC patients,mutations in PiT2-loop7 are mainly nonsense or frameshift mutations that probably cause PFBC due to C-PD1131 *** date,six missense mutations have been identified in PiT2-loop7;however,the mechanisms by which these mutations cause PFBC are poorly ***,we found that the p.T390A and p.S434W mutations in PiT2-loop7 decreased the Pi transport activity and cell surface levels of ***,we showed that these two mutations attenuated its membrane localization by affecting adenosine monophosphate-activated protein kinase(AMPK)-or protein kinase B(AKT)-mediated PiT2 *** contrast,the p.S121C and p.S601W mutations in the PD domains did not affect PiT2 phosphorylation but rather impaired its substrate-binding *** results suggested that missense mutations in PiT2-loop7 can cause Pi dyshomeostasis by affecting the phosphorylation-regulated cell-surface localization of *** study helps understand the pathogenesis of PFBC caused by PiT2-loop7 missense mutations and indicates that increasing the phosphorylation levels of PiT2-loop7 could be a promising strategy for developing PFBC therapies.
