The single-detector spectrometers based on 2D layer van der Waals (vdW) heterojunctions offer advantages in spectral reconstruction due to their high sensitivity, tunable optical properties, and the ability to cover a broad spectral range. There exist two principal algorithms dominating spectrum reconstruction for this kind spectrometer: the Tikhonov regularization method combined with the Least Squares Method (LSM) and neural network-based approaches, particularly Deep Learning (DL). However, both of the algorithms exhibit inherent limitations in spectral reconstruction, which constrain the versatility of computational spectrometers that rely solely on a single algorithm for reconstructing diverse spectral profiles. To overcome this limitation, we introduce an artificial neural network (ANN)-based classification model capable of dynamically selecting the optimal algorithm throughout the reconstruction process. This enables highly accurate spectral reconstruction within the 440-700 nm wavelength range, achieving a spectral resolution of 6 nm. By harnessing the complementary strengths of multiple algorithms, our approach proposes a novel strategy for combining techniques to enhance the precision of spectral reconstructions, laying the groundwork for more sophisticated methods in the future. © 1980-2012 IEEE.

Dopamine can be used to treat depression,myocardial infarction,and other ***,few reports are available on the de novo microbial synthesis of dopamine from low-cost *** this study,integrated omics technology was used to explore the dopamine metabolism of a novel marine multi-stress-tolerant aromatic yeast Meyerozyma guilliermondii ***6 was found to have the ability to biosynthesize dopamine when using glucose as the substrate.14 key genes for the biosynthesis of dopamine were identified by whole genome-wide *** and proteomic data showed that the expression levels of gene AAT2 encoding aspartate aminotransferase(regulating dopamine anabolism)were upregulated,while gene AO-I encoding copper amine oxidase(involved in dopamine catabolism)were downregulated under 10%NaCl stress compared with non-NaCl stress,thereby contributing to biosynthesis of ***,the amount of dopamine under 10%NaCl stress was 2.51-fold higher than that of zero NaCl,which was consistent with the multi-omics ***-time fluorescence quantitative PCR(RT-qPCR)and high-performance liquid chromatography(HPLC)results confirmed the metabolic model of ***,by overexpressing AAT2,AST enzyme activity was increased by 24.89%,the expression of genes related to dopamine metabolism was enhanced,and dopamine production was increased by 56.36%in recombinant *** conclusion,Meyerozyma guilliermondii GXDK6 could utilize low-cost carbon source to synthesize dopamine,and NaCl stress promoted the biosynthesis of dopamine.

Highly crystalline organic semiconductors are ideal materials for photocatalytic hydrogen evolution in water ***,the instability and complex synthesis processes of most reported organic molecule-based photocatalysts restrict their *** this study,we introduce benzo[1,2-b:4,5-bʹ]bis[1]benzothiophene-3,9-dicarboxylic acid,5,5,11,11-tetraoxide(FSOCA),a highly crystalline,stable molecular crystal that is easy to synthesize and serves as an efficient photocatalyst for the hydrogen evolution *** exhibits high efficiency in sacrificial hydrogen evolution reaction(760μmol h^(−1),76 mmol g^(−1)h^(−1)at 330 mW cm^(−2);570μmol h^(−1),57 mmol g^(−1)h^(−1)at 250 mW cm^(−2)),and FSOCA remains stable during photocatalysis for up to 400 *** and theoretical studies confirmed the presence of hydrogen bonds between the sulfone group and the sacrificial agent(ascorbic acid).This interaction significantly improved the oxidation reaction kinetics and boosted the photocatalytic *** study presents a scalable and convenient approach to synthesize highly crystalline,active,and stable organic photocatalysts with potential applications in large-scale photocatalysis.

Apoptosis is an important process for organism development that functions to eliminate cell damage,maintain homeostasis,and remove obsolete tissues during *** mammals,apoptosis is accompanied by the release of cytochrome C(Cyt-c)from mitochondria to the ***,whether this process is conserved in the fruit fly,Drosophila melanogaster,remains *** this study,we discovered that during the degradation of Drosophila salivary gland,the transcription of mitochondria apoptosis factors(MAPFs),Cyt-c,and death-associated APAF1-related killer(Dark)encoding genes are all upregulated antecedent to initiator and effector caspases encoding *** proteins Cyt-c and the active caspase 3 appear gradually in the cytoplasm during salivary gland ***,the Cyt-c protein colocates with mito-GFP,the marker indicating cytoplasmic mitochondria,and the change in mitochondrial membrane potential coincides with the appearance of Cyt-c in the ***,impeding or promoting 20E-induced transcription factor E93 suppresses or enhances the staining of Cyt-c and the active caspase 3 in the cytoplasm of salivary gland,and accordingly decreases or increases the mitochondrial membrane potential,*** research provides evidence that cytoplasmic Cyt-c appears before apoptosis during Drosophila salivary gland degradation,shedding light on partial conserved mechanism in apoptosis between insects and mammals.

Dear Editor,Traffic driving is a dynamic and complicated task in which drivers are required to pay close attention to the important targets or regions to maintain safe *** weather conditions make it more challenging with factors such as low visibility,raindrops,pedestrian with umbrellas,wipers,*** showed that rainy condition affects driving safety significantly[1],[2].It is reported that,in raining weather condition,the odds for a fatal accident are 3.340 times higher on highways than on streets[3].An investigation of the relationship between rainfall and fatal crashes in Texas from 1994 to 2018 on fatality analysis reporting system(FARS)database illustrated that rain-related fatal crashes represented about 6.8%of the total fatal crashes on average,moreover,the proportion showed high variability at the annual,monthly,and hourly time scales[4].Therefore,raining is a complex and critical factor for road safety planning and *** fact,the traffic environment is a dynamic scene with multiple sources of information,including important targets that are highly relevant to the current driving task as well as irrelevant targets that may distract the driving task[5].

Plant endophytic fungi exhibit diverse biotransformation pathways of mogrosides and show great potential application in siamenoside I production

Fungal endophytes,as an untapped resource of glycoside hydrolase biocatalysts,need to be further *** V,the primary active compound in Siraitia grosvenorii fruit,can be converted into other various bioactive mogrosides by selective hydrolysis of glucose residues at C3 and C24 *** present study,20 fungal strains were randomly selected from our endophytic fungal strain library to assess their capability for mogroside V *** results revealed that relatively high rate(30%)endophytic fungal strains exhibited transformative *** analysis indicated that endophytic fungi could produce abundant mogrosides,and the pathways for biotransforming mogroside V showed *** the given fungal endophytes,Aspergillus sp.S125 almost completely converted mogroside V into the end-products mogroside II A and aglycone within just 2 days of fermentation;Muyocopron sp.A5 produced rich intermediate products,including siamenoside I,and the end-product mogroside II ***,we optimized the fermentation conditions for Aspergillus sp.S125 and Muyocopron sp.A5 to evaluate the feasibility of large-scale mogroside V *** optimization,Aspergillus sp.S125 converted 10 g/L of mogroside V into 4.5 g/L of mogroside II A and 3.6 g/L of aglycone after 3 days of fermentation,whereas Muyocopron sp.A5 selectively produced 4.88 g/L of siamenoside I from 7.5 g/L of mogroside V after 36 h of *** study not only identifies highly effective biocatalytic candidates for mogrosides transformation,but also strongly suggests the potential of plant endophytic fungi as valuable resources for the biocatalysis of natural compounds.

An automatic annotation tool and reference database for T cell subtypes and states at single-cell resolution

T cells have various subtypes and states with different functions. However, a reference list and automated annotation tool for T cell subtypes and states are lacking, which is critical for analyzing and comparing T cells under various conditions. We constructed the largest human T cell reference, containing 1,348,268 T cells from 35 conditions and 16 tissues. We classified T cells into 33 subtypes and further stratified them into 68 categories according to subtype and state. Based on this reference, we developed a tool named STCAT to automatically annotate T cells from scRNA-seq data by hierarchical models and marker correction. The accuracy of STCAT was 28% higher than that of existing tools validated on six independent datasets, including cancer and healthy samples. Using STCAT, we consistently discovered that CD4+ Th17 cells were enriched in late-stage lung cancer patients in multiple datasets, whereas MAIT cells were prevalent in milder-stage COVID-19 patients. We also confirmed a decrease in Treg cytotoxicity in post-treatment ovarian cancer. Systematic landscape analyses of CD4+ and CD8+ T cell references revealed that CD4+ Treg cells were enriched in tumor samples and that CD8+ naive-related cells were abundant in healthy individuals. Finally, we deposited all the T cell references and annotations into a TCellAtlas (https://***/TCellAtlas) database, which allows users to browse T cell expression profiles and analyze customized scRNA-seq data by STCAT. In conclusion, comprehensive human T cell subtypes and states reference, automated annotation tool, and database will greatly facilitate research on T cell immunity and tumor immunology. © 2025 science China Press

Excessive fructose consumption increases hepatic de novo lipogenesis,resulting in cellular stress,inflammation and liver ***-B is a resident protein of the endoplasmic reticulum that regulates its structure and *** Nogo-B is a key protein in glycolipid metabolism,and inhibition of Nogo-B has protective effects against metabolic syndrome,thus small molecules that inhibit Nogo-B have therapeutic benefits for glycolipid metabolism *** this study we tested 14 flavones/isoflavones in hepatocytes using dual luciferase reporter system based on the Nogo-B transcriptional response system,and found that 6-methyl flavone(6-MF)exerted the strongest inhibition on Nogo-B expression in hepatocytes with an IC50 value of 15.85 μ*** of 6-MF(50 mg·kg-1·d-1,*** 3 weeks)significantly improved insulin resistance along with ameliorated liver injury and hypertriglyceridemia in high fructose diet-fed *** HepG2 cells cultured in a media containing an FA-fructose mixture,6-MF(15 μM)significantly inhibited lipid synthesis,oxidative stress and inflammatory ***,we revealed that 6-MF inhibited Nogo-B/ChREBP-mediated fatty acid synthesis and reduced lipid accumulation in hepatocytes by restoring cellular autophagy and promoting fatty acid oxidation via the AMPKα-mTOR ***,6-MF may serve as a potential Nogo-B inhibitor to treat metabolic syndrome caused by glycolipid metabolism dysregulation.
