Dear Editor,Protein histidine methylation has been found to be a widespread post-translational modification(PTM)in mammalian cells and represents more than 13%of all protein methylation events in the human methylome(Ning et al.,2016;Kapell and Jakobsson,2021).Recently,we and other groups independently identified METTL9 as the first broad-specificity histidine N1 methyltransferase in mammals(Davydova et al.,2021;Lv et al.,2021).METTL9 methylates a variety of substrates with a common x-His-x-His motif(His is for histidine;the second x is preferably small amino acids:A/C/G/S)and catalyzes methyl transfer to the last histidine.

Grasses,including turf and forage,cover most of the earth's surface;predominantly important for land,water,livestock feed,soil,and water conservation,as well as carbon *** production and quality of grasses by modern molecular breeding is gaining more research *** advances in genome-editing technologies are helping to revolutionize plant breeding and also offering smart and efficient acceleration on grass ***,we reviewed all recent researches using(CRISPR)/CRISPR-associated protein(Cas)-mediated genome editing tools to enhance the growth and quality of forage and turf ***,we highlighted emerging approaches aimed at advancing grass breeding *** assessed the CRISPR-Cas effectiveness,discussed the challenges associated with its application,and explored future perspectives primarily focusing on turf and forage *** the promising potential of genome editing in grasses,its current efficiency remains limited due to several bottlenecks,such as the absence of comprehensive reference genomes,the lack of efficient gene delivery tools,unavailability of suitable vector and delivery for grass species,high polyploidization,and multiple homoeoalleles,*** these challenges,the CRISPR-Cas system holds great potential to fully harness its benefits in grass breeding and genetics,aiming to improve and sustain the quantity and quality of turf and forage grasses.

R3-MYB proteins OsTCL1 and OsTCL2 modulate seed germination via dual pathways in rice

Salt and drought stress are common abiotic factors that exert a detrimental influence on seed germination,potentially leading to significantly impaired growth and production in *** a comprehensive understanding of the molecular response of seeds to abiotic stress during the germination is of paramount *** the present study,we identified two R3-MYB genes in rice,namely OsTCL1 and OsTCL2,and characterized their roles in regulating seed germination under salt and drought *** with tcl1 and tcl2 mutant alleles exhibited delayed seed germination,particularly under stress *** tcl1 tcl2 double mutant showed an even more pronounced reduction in germination during initial stages of germination,thereby indicating a redundant regulatory function of OsTCL1 and OsTCL2 in seed germination under abiotic ***,we demonstrated that the transcript levels of several phospholipase D(PLD)genes were downregulated in the tcl1 tcl2 mutant,resulting in a decreased level of the phosphatidic acid(PA)*** of 1-butanol,a competitive substrate inhibitor of PLD-dependent production of PA,attenuated the stress response of the tcl1 tcl2 *** suggests that OsTCL1 and OsTCL2 partially modulate seed germination through the PLD-PA signaling ***,there were alterations in the expression of genes involved in abscisic acid(ABA)biosynthesis,metabolism and signaling transduction in the double *** changes affected the endogenous ABA level and ABA response,thereby influencing seed *** of both 1-butanol and ABA synthesis inhibitor sodium tungstate(Na2WO4)nearly eliminated the differences in stress response between wild type and the tcl1 tcl2 *** indicates that OsTCL1 and OsTCL2 synergistically coordinate seed germination under abiotic stresses through both the PLD-PA signaling and ABA-mediated pathways.

Pumpkin polysaccharides(PPe)have a variety of bioactive effects and our previous research showed the acid hydrolysate(PPe-S,a mixture)from PPe had an antioxidative capacity both in vitro and in *** aim of this study was to purify PPe-S and investigate the antioxidant stress effects of 2 purified components(PPe-S-1 and PPe-S-2)using Caenorhabditis elegans as model *** results showed that PPe-S-2 had a notable antioxidant effect,and could significantly enhance the activities of antioxidant enzymes including superoxide dismutase(SOD)(P<0.01),catalase(CAT)(P<0.01)and glutathione reductase(GR)(P<0.05),and increase the level of glutathione(GSH)(P<0.01),and decreased the content of malondialdehyde(MDA)(P<0.05).PPe-S-2 could significantly extend the survival time of ***(P<0.01),which were stress-induced by hydrogen peroxide and methyl ***-S-2 was a heteropolys accharide composed of glucose,arabinose,rhamnose and galactose with the molar ratio of 1.00:0.03:0.02:*** molecular weight of PPe-S-2 was 0.73 kDa detected by high performance liquid *** studies demonstrated that PPe-S-2 obtained by the acid hydrolysis of PPe had a prominent protective effect to the damage induced by the intracellular free radical generating agents.

Srgap2 suppression ameliorates retinal ganglion cell degeneration in mice

Slit-Robo GTPase-activating protein 2(SRGAP2) plays important roles in axon guidance, neuronal migration, synapse formation, and nerve regeneration. However, the role of SRGAP2 in neuroretinal degenerative disease remains unclear. In this study, we found that SRGAP2 protein was first expressed in the retina of normal mice at the embryonic stage and was mainly located in the mature retinal ganglion cell layer and the inner nuclear layer. SRGAP2 protein in the retina and optic nerve increased after optic nerve crush. Then, we established a heterozygous knockout(Srgap2+/–) mouse model of optic nerve crush and found that Srgap2 suppression increased retinal ganglion cell survival, lowered intraocular pressure, inhibited glial cell activation, and partially restored retinal function. In vitro experiments showed that Srgap2 suppression activated the mammalian target of rapamycin signaling pathway. RNA sequencing results showed that the expression of small heat shock protein genes(Cryaa, Cryba4, and Crygs) related to optic nerve injury were upregulated in the retina of Srgap2+/– mice. These results suggest that Srgap2 suppression reduced the robust activation of glial cells, activated the mammalian target of rapamycin signaling pathway related to nerve protein, increased the expression of small heat shock protein genes, inhibited the degeneration of retinal ganglion cells, and partially restored optic nerve function.

Noise-induced hearing loss(NIHL) is a highly prevalent form of sensorineural hearing damage that has significant negative effects on individuals of all ages and there are no effective drugs approved by the US Food and Drug Administration. In this study, we unveil the potential of superparamagnetic iron oxide nanoparticle assembly(SPIOCA) to reshape the dysbiosis of gut microbiota for treating NIHL. This modulation inhibits intestinal inflammation and oxidative stress responses, protecting the integrity of the intestinal barrier. Consequently, it reduces the transportation of pathogens and inflammatory factors from the bloodstream to the cochlea. Additionally, gut microbiota-modulated SPIOCA-induced metabolic reprogramming in the gut–inner ear axis mainly depends on the regulation of the sphingolipid metabolic pathway, which further contributes to the restoration of hearing function. Our study confirms the role of the microbiota–gut–inner ear axis in NIHL and provides a novel alternative for the treatment of NIHL and other microbiota dysbiosis-related diseases.

The white light interferometry technique can measure the three-dimensional topography of ultra-precise complex surface with sub-nanometer resolution and precision, and has been widely concerned by academia and industry since it was proposed. The measurement range of white light interferometry technology is usually limited by the size of the optical field of view of the system, and the surface of the measured object can only be measured within the limited range of the camera's single imaging. In order to obtain the microscopic surface topography with a larger field of view, it is usually necessary to conduct stitching measurement. In this paper, a measurement method of aperture stitching of white light interferometry based on feature extraction is presented. The first step is subaperture partitioning and data preprocessing. The second step is feature extraction. The feature points are extracted by the Speeded-Up Robust Features (SURF) algorithm. The third step is to identify and filter wrong match points in the feature extraction algorithm using the Random Sample Consensus (RANSAC) algorithm. The fourth step is to use the improved Iterative Closest Point (ICP) algorithm to accurately register the point cloud, and perform data fusion in the overlapping region to complete the reconstruction of the microscopic three-dimensional surface topography. This method is used to measure the microstructure of quartz vibratory beam accelerometer. The experimental results show that the proposed algorithm has a faster speed than the traditional algorithm while maintaining high precision. © COPYRIGHT SPIE.
