CRISPR-based gene editing technology and its application in microbial engineering

Gene editing technology involves the modification of a specific target gene to obtain a new function or *** advances in clustered regularly interspaced short palindromic repeats(CRISPR)-Cas-mediated technolo-gies have provided an efficient tool for genetic engineering of cells and ***,we review the three emerging gene editing tools(ZFNs,TALENs,and CRISPR-Cas)and briefly introduce the principle,classification,and mechanisms of the CRISPR-Cas *** for gene editing based on endogenous and exogenous CRISPR-Cas systems,as well as the novel base editor(BE),prime editor(PE),and CRISPR-associated transposase(CAST)technologies,are described in *** addition,we summarize recent developments in the application of CRISPR-based gene editing tools for industrial microorganism and probiotics ***,the potential challenges and future perspectives of CRISPR-based gene editing tools are discussed.

Limited by the theoretical output power limitation of single-mode fiber laser, achieving higher laser output power requires us to turn to using laser beam combination technology. The current beam combining methods can mainly be divided into spectral beam combining and aperture beam combining. The former method has the advantages of better optical quality, but the beam combining grating bears too much laser energy density and is limited by wavelength selection and scalability. The collimated beam combining method performs better at scalability and shows more uniform energy density, while the beam quality is seriously affected by decrease of optical axis uniformity caused by the thermal deformation of the mechanical cylinder shell, so the problem of laser heat deposition should be solved urgently. Considering that the laser is a Gaussian beam, there is still a small amount of energy beyond the waist radius, So an attempt was made to design a collimating beam scheme with the laser emitting end face as a whole continuous compound eye lens to replace the traditional discrete collimating unit. Zernike polynomial is a classical polynomial to describe optical surface figure. It is consistent with the description form of optical geometric aberration, and its unit orthogonality ensures that changing the number of polynomial terms will not change the original coefficients, which shows its excellent characteristics in otpical surface fitting. Continuous compound eye lens is a freeform lens contains several aspheric collimating elements. Dear Editor, Histone deacetylases(HDACs)are evolutionally conserved enzymes that remove acetyl modifications from histones and play a central role in epigenetic gene silencing.1 Class Ⅰ HDACs are promising targets for epigenetic therapies for a range of diseases such as cancers,inflammations,infections,and neurological diseases.2 Yeast Rpd3 is the founding member of class Ⅰ HDACs,which forms two distinct complexes:the～1.2MDa Rpd3L deacetylating histones at promoter regions,and the～0.6MDa Rpd3S targeting transcribed regions to suppress intragenic transcription initiation.3,4 Rpd3S consists of three core proteins:Rpd3,Sin3,and Ume15 along with two dedicated chromatin binding subunits:Eaf3 and Rco 1.6 The structures of the yeast Rpd3S complex and its human homolog Sin3B complex have been recently reported.7,8 Here,we report the cryo-electron microscopy(cryo-EM)structure of the Rpd3S holoenzyme binding a nucleo-some at 3.7 Å resolution(Supplementary information,Table S1).The structure illuminates an intact H3 tail(1-24 aa)binding to the Rpd3S complex in a catalysis-competent conformation,with the H3K18 residue poised for catalysis.

Candidate bacterial phylum CSP1-3 has not been cultivated and is poorly understood. Here, we analyzed 112 CSP1-3 metagenome-assembled genomes and showed they are likely facultative anaerobes, with 3 of 5 families encoding autotrophy through the reductive glycine pathway(RGP), Wood–Ljungdahl pathway(WLP) or Calvin-Benson-Bassham(CBB), with hydrogen or sulfide as electron donors. Chemoautotrophic enrichments from hot spring sediments and fluorescence in situ hybridization revealed enrichment of six CSP1-3 genera, and both transcribed genes and DNA-stable isotope probing were consistent with proposed chemoautotrophic metabolisms. Ancestral state reconstructions showed that the ancestors of phylum CSP1-3 may have been acetogens that were autotrophic via the RGP, whereas the WLP and CBB were acquired by horizontal gene transfer. Our results reveal that CSP1-3 is a widely distributed phylum with the potential to contribute to the cycling of carbon, sulfur and nitrogen. The name Sysuimicrobiota phy. nov. is proposed.

Most plasmalemmal proteins are organized into clusters to modulate various cellular ***,the machineries that regulate protein clustering remain largely ***,with EGFR as an example,we directly and in detail visualized the entire process of EGFR from synthesis to secretion onto the plasma membrane(PM)using a high-speed,high-resolution spinning-disk confocal ***,colocalization imaging revealed that EGFR secretory vesicles underwent transport from the ER to the Golgi to the PM,eventually forming different distribution forms on the apical and basal membranes;that is,most EGFR formed larger clusters on the apical membrane than the basal membrane.A dynamic tracking image and further siRNA interference experiment confirmed that fusion of secretory vesicles with the plasma membrane led to EGFR clusters,and we showed that EGFR PM clustering may be intimately related to EGFR signaling and cell ***,we found that the size and origin of the secretory vesicles themselves may determine the difference in the distribution patterns of EGFR on the *** importantly,we showed that actin influenced the EGFR distribution by controlling the fusion of secretory vesicles with the ***,a comprehensive understanding of the EGFR secretion process helps us to unravel the EGFR clustering process and elucidate the key factors determining the differences in the spatial distribution of EGFR PM,highlighting the correlation between EGFR secretion and its PM distribution pattern.

In modern optical detection, the vertical resolution of three-dimensional(3-D) topography obtained by white light interferometer (WLI) can reach sub-nanometer level. Under the same detection environment, optical system and scanning sampling system, the quality of coherent peak extraction algorithm directly determines the measurement accuracy. After decomposing and analyzing the representation form of WLI signal, this paper proposes a surface topography reconstruction method based on the modulation function curve of interference signal for gaussian center positioning. Under the influence of typical error sources, the measurement accuracy of this method is simulated and analyzed. Finally, based on the method proposed in this paper, a comparative measurement experiment of 1.8µm standard step is carried out with ZYGO commercial interferometer. The simulation and experimental results show that the algorithm has good robustness and effectiveness. © 2023 SPIE.

Plasma-activated water(PAW),as an extended form of cold atmospheric-pressure plasma,greatly expands the application of plasma-based *** biological effects of PAW are closely related to the aqueous reactive species,which can be regulated by the activation *** this study,surface plasma-activated air(SAA)and a He+O_(2)plasma jet(Jet)were parallelly combined(the SAA+Jet combination)or sequentially combined(the SAA→Jet combination and the Jet→SAA combination)to prepare plasma-activated saline(PAS).The PAS activated by the combinations exhibited stronger bactericidal effects than that activated by the SAA or the Jet *** concentrations of H_(2)O_(2)and NO_(2)^(-)were higher in the PAS activated by the Jet→SAA combination,while ONOO^(-)concentrations were close in the three kinds of PAS and^(1)O_(2)concentrations were higher in the PAS activated by the SAA+Jet *** analysis of scavengers also demonstrated that H_(2)O_(2),^(1)O_(2),and ONOO^(-)in the PAS activated by the SAA+Jet combination,and^(1)O_(2)in the PAS activated by the Jet→SAA combination played critical roles in bactericidal ***,the effective placement time of the three PAS varied,and the PAS activated by the Jet→SAA combination could also inactivate 2.6-log_(10)of MRSA cells after placement for more than 60 *** regulation of reactive species in plasma-activated water via different combinations of plasma devices could improve the directional application of plasma-activated water in the biomedical field.

Calcium ions(Ca2+) play a crucial role as secondary messengers in both excitable and non-excitable cells.A complex system of proteins and molecules involved in calcium handling allows Ca2+signals to be *** cancer cells,mutations,aberrant expression,and dysregulation of these calcium handling toolkit proteins disrupt the normal Ca2+flux between extracellular space,cytosol,endoplasmic reticulum and mitochondria,as well as the spatio-temporal patterns of Ca2+*** leads to the dysregulation of calcium-dependent effectors that control key signaling pathways involved in cancer cell proliferation,survival and *** there has been progressing in understanding the remodelling of calcium homeostasis in cancer cells and identifying key calcium transport molecules that promote malignant phenotypes,much work remains to be done to translate these fundamental findings into new tools for diagnosing and treating cancer by targeting Ca2+homeostasis.

The coronavirus disease 2019(COVID-19)pandemic,caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),has claimedmillions of lives and caused innumerable economic losses ***,state-of-the-art treatments still lag behind the continual emergence of new *** to resolving this issue is developing antivirals to deactivate coronaviruses regardless of their structural ***,we report an innovative antiviral strategy involving extracellular disintegration of viral proteins with hyperanion-grafted enediyne(EDY)*** core EDY generates reactive radical species and causes significant damage to the spike protein of coronavirus,while the hyperanion groups ensure negligible cytotoxicity of the *** EDYs exhibit antiviral activity down to nanomolar concentrations,and the selectivity index of up to 20,000 against four kinds of human coronavirus,including the SARS-CoV-2 Omicron variant,suggesting the high potential of this new strategy in combating the COVID-19 pandemic and a future“disease X.”
