Although the polarization converter utilizing silicon photonics technology has been reported several years ago, either the performance (conversion efficiency and operation window) or the manufacture is always limited by the cross-polarization coupling theory. We discovered that an asymmetric nanowire waveguide changing along the wave propagation direction produced a gradient effective mode index. A TE-to-TM polarization converter consisted of an asymmetric nanowire waveguide and a uniform nanowire waveguide constructed on the SOI platform was demonstrated. Gradient effective mode index effect and mode coupling theory promoted an ultra-high polarization conversion efficiency of 99.997% and broad wavelength operation window of 100 nm. Compact configuration presents high integration density, which benefits the on-chip mode multiplexing technology. Simple structure with moderate critical dimension promotes the fabrication fast and cheap. © 2022, The Authors. All rights reserved.

Accurate counting of RNA in tissues with highly denoising amplified imaging

Characterization of the distribution and accurate counting of RNA molecules in the context of tissues is necessary to understand their complexity and heterogeneity. Single-molecule fluorescence in situ hybridization reveals the abundance and distribution of RNA and resolves different cell types in complex tissues. Especially, off-target binding and nonspecific adsorption of probes are prone to producing nonspecific amplification. Herein, we present highly de-noising amplified imaging, which leverages a site-specific cleavage-amplifying design to achieve accurate counting of RNA in tissues. Our method avoids adding probe as primer, decreases nonspecific spots of single cells from 7 to nearly zero, and enables RNA imaging in uncleared tissue sections with nearly zero noise. We demonstrate the efficacy of this method on various thickness of mouse tissue sections. We envision this approach will serve as a tool to revealing the information content from patient samples for biomedical purpose.

Effects of formulation composition and CeO2 nanoparticles doping on the morphologies of polymer spacer columns and electro-optical properties of PDLC

Polymer dispersed liquid crystal (PDLC) with polymer spacer columns were prepared by ultraviolet (UV) stepwise polymerization induced phase separation (PIPS) process. The influences of the formulation composition, such as the internal proportion of monomers and the liquid crystal (LC) content, on the morphologies of polymer spacer columns and electro-optical properties of PDLC films, were researched. Due to the difference in cross-linking density and polymerization rate, PDLC films exhibit different morphologies of polymer spacer columns and electro-optic properties. Based on the above, the CeO2 nanoparticles were doped into PDLC films. The influences of the doping content of CeO2 nanoparticles on the polymer network and electro-optical properties of PDLC were studied, and the EDS spectra were used to characterize the extent of dispersion of CeO2 nanoparticles in the polymer matrix. The results of the study showed that the contrast ratio (CR) of PDLC film increases from 10.9 to 27.3 at 0.8% CeO2 nanoparticles doping, while the saturation voltage (Vsat) decreases from 71.3 V to 47.9 V. Therefore, the work offers a new approach to the preparation of PDLC films with excellent comprehensive performance. © 2022 Elsevier Ltd

Plant Functional Genomics Based on High-Throughput CRISPR Library Knockout Screening: A Perspective

Plant biology studies in the post-genome era have been focused on annotating genome sequences' functions. The established plant mutant collections have greatly accelerated functional genomics research in the past few decades. However, most plant genome sequences' roles and the underlying regulatory networks remain substantially unknown. Clustered, regularly interspaced short palindromic repeat (CRISPR)-associated systems are robust, versatile tools for manipulating plant genomes with various targeted DNA perturbations, providing an excellent opportunity for high-throughput interrogation of DNA elements' roles. This study compares methods frequently used for plant functional genomics and then discusses different DNA multi-targeted strategies to overcome gene redundancy using the CRISPR-Cas9 system. Next, this work summarizes recent reports using CRISPR libraries for high-throughput gene knockout and function discoveries in plants. Finally, this work envisions the future perspective of optimizing and leveraging CRISPR library screening in plant genomes' other uncharacterized DNA sequences.

Influence of N, P, Fe Nutrients Availability on Nitrogen Metabolism-Relevant Genes Expression in Skeletonema marinoi

Nitrogen(N), Phosphorus(P), and Iron(Fe) are essential elements for cellular structure and metabolism. In addition to dissolved inorganic nitrogen(DIN), phytoplankton is able to utilize dissolved organic nitrogen(DON). There is general consensus that both bacteria and higher plants nitrogen metabolism is affected by phosphate availability; this was also found to be true in coccolithophorid. Iron affects the structure and function of ecosystems through its effects on nitrogen metabolism. However, it is unclear how these nutrients affect Skeletonema marinoi's nitrogen metabolism. Here, using RT-qPCR, we investigate effects of N, P, and Fe on S. marinoi's nitrogen metabolism and nitrate reductase activity. These results illuminate that in S. marinoi, various nutrients have direct regulation on these genes expression at the molecular level. The varying degree of responses for these genes expression with differing N sources may act to increase the efficiency of nutrient capture when nitrate is limited. Suitable gene expression occurs at a N/P ratio of 16, which represents the atomic N/P ratio of phytoplankton cells and N/P concentrations in ocean; thus, nitrogen metabolism gene expression should be regulated by the existing N/P ratios in the phytoplankton's internal and external environment. Fe concentration has a direct and significant effect on nitrogen metabolism by regulating gene expression and nitrate reductase activity. Gene expression profiles identified in S. marinoi provide a foundation for understanding molecular mechanisms behind diatom nitrogen metabolism with changing N, P, and Fe nutrients allowing a basic understanding of how diatom growth is affected by nutrient utilization.
