Contrast-enhanced orbital MRI for activity assessment and treatment response prediction in thyroid eye disease

Objective: To investigate the performance of contrast-enhanced orbital MRI (T1-weighted contrast-enhanced [T1CE] and dynamic contrast-enhanced MRI [DCE-MRI]) for activity assessment and prediction of treatment response to intravenous glucocorticoid (IVGC) treatment in thyroid eye disease (TED). Methods: A retrospective study of 59 active moderate-to-severe TED patients (36 responsive and 23 unresponsive) treated with IVGC, matched with 31 inactive TED patients were included. T1CE and DCE-MRI were collected from each subject. Univariate and multivariate logistic regression analyses, as well as receiver operating characteristic (ROC) curves (evaluated by area under the curve, AUC) were used to evaluate the diagnostic and predictive models. Results: Active patients showed a higher signal intensity ratio of lateral rectus (LR-SIR) on T1CE and a higher wash-in rate of inferior rectus (IR-WIR) on DCE-MRI compared to inactive patients (p < 0.05). The AUC for T1CE and DCE-MRI based models were 0.866, and 0.844, respectively. The combined model (LR-SIR and IR-WIR) significantly enhanced model performance (AUC = 0.914, 95 % CI: 0.858–0.970). Responsive patients demonstrated a longer time to peak of lateral rectus (LR-TTP) than unresponsive patients (p < 0.05). The cutoff value of prediction probability was 0.678, and the AUC based on this model was 0.739 (95 % CI: 0.608–0.807). Conclusions: T1CE and DCE-MRI could effectively assist in TED activity assessment, and DCE-MRI could help predict IVGC treatment response. Specifically, LR-SIR and IR-WIR were indicators of TED activity, and IR-TTP was predictive of IVGC treatment response. Combining both contrast-enhanced sequences could further enhance the efficiency of TED activity assessment. © 2025 Elsevier B.V.

Introduction: Developmental competence of oocytes matured in vitro is limited due to a lack of complete understanding of metabolism and metabolic gene expression during oocyte maturation and embryo development. Conventional metabolic analysis requires a large number of samples and is not efficiently applicable in oocytes and early embryos, thereby posing challenges in identifying key metabolites and regulating their in vitro culture system. Objectives: To enhance the developmental competence of sheep oocytes, this study aimed to identify and supplement essential metabolites that were deficient in the culture systems. Methods: The metabolic characteristics of oocytes and embryos were determined using ultrasensitive metabolomics analysis on trace samples and single-cell RNA-seq. By conducting integrated analyses of metabolites in cells (oocytes and embryos) and their developmental microenvironment (follicular fluid, oviductal fluid, and in vitro culture systems), we identified key missing metabolites in the in vitro culture systems. In order to assess the impact of these key missing metabolites on oocyte development competence, we performed in vitro culture experiments. Furthermore, omics analyses were employed to elucidate the underlying mechanisms. Results: Our findings demonstrated that betaine, carnitine and creatine were the key missing metabolites in vitro culture systems and supplementation of betaine and L-carnitine significantly improved the blastocyst formation rate (67.48% and 48.61%). Through in vitro culture experiments and omics analyses, we have discovered that L-carnitine had the potential to promote fatty acid oxidation, reduce lipid content and lipid peroxidation level, and regulate spindle morphological grade through fatty acid degradation pathway. Additionally, betaine may participate in methylation modification and osmotic pressure regulation, thereby potentially improving oocyte maturation and early embryo development in sheep. Generalist foundation models (GFMs) are renowned for their exceptional capability and flexibility in effectively generalizing across diverse tasks and modalities. In the field of medicine, while GFMs exhibit superior generalizability based on their extensive intrinsic knowledge as well as proficiency in instruction following and in-context learning, specialist models excel in precision due to their in-depth domain-specific knowledge. In this work, for the first time, we explore the synergy between the GFM and specialist models, to enable precise medical image analysis on a broader scope. Specifically, we propose a novel cooperative framework, Generalist-Specialist Collaboration (GSCo), which consists of two stages, namely the construction of GFM and specialists, and collaborative inference on downstream tasks. In the construction stage, we develop MedDr, the largest open-source GFM tailored for medicine, showcasing exceptional instruction-following and in-context learning capabilities. Meanwhile, a series of lightweight specialists are crafted for specific downstream tasks with low computational overhead. In the collaborative inference stage, we introduce two cooperative mechanisms, Mixture-of-Expert Diagnosis (MoED) and Retrieval-Augmented Diagnosis (RAD), to harvest the generalist’s in-context learning abilities alongside the specialists’ domain expertise. Concretely, MoED incorporates predictions from specialists as references, while RAD employs specialists to retrieve similar cases, collectively providing MedDr with much more contextual guidance. For a comprehensive evaluation, we curate a large-scale benchmark featuring 28 datasets and about 250,000 images across a wide range of medical modalities, including radiology, pathology, dermatology, ophthalmology, and gastroenterology. The origination of new genes contributes to the biological diversity of life. New genes may quickly build their network, exert important functions, and generate novel phenotypes. Dating gene age and inferring the origination mechanisms of new genes, like primate-specific genes, is the basis for the functional study of the genes. However, no comprehensive resource of gene age estimates across species is available. Here,we systematically date the age of 9,102,113 protein-coding genes from 565 species in the Ensembl and Ensembl Genomes databases, including 82 bacteria, 57 protists, 134 fungi, 58 plants, 56 metazoa, and 178 vertebrates, using a protein-family-based pipeline with Wagner parsimony algorithm. We also collect gene age estimate data from other studies and uniformly distribute the gene age estimates to time ranges in a million years for comparison across studies. All the data are cataloged into Gen Origin(http://***/), a user-friendly new database of gene age estimates, where users can browse gene age estimates by species, age, and gene ontology. In Gen Origin, the information such as gene age estimates,annotation, gene ontology, ortholog, and paralog, as well as detailed gene presence/absence views for gene age inference based on the species tree with evolutionary timescale, is provided to researchers for exploring gene functions.

With the gradual development of the Marine field, the underwater robot has gradually entered people's perspective. However, the structure and control of the current amphibious robot is too complex, and most of them need to switch two sets of structures to adapt to the amphibious environment. Therefore, a simple and reliable robot that can adapt to two environments with seven sets of structures is urgently needed. Based on the undulation motion principle of ray's pectoral fin, the undulation mechanism driven by 6 groups of crank rocker mechanism on both sides is designed, and the rocker mechanism is connected by rubber material to form the fin driven by rocker. The structure modeling and kinematics simulation of the robot are carried out, and it is confirmed that the mechanism of the ray can move effectively and the fin can realize regular fluctuation, which verifies the feasibility of the bionic ray underwater robot.

The wind power industry is grappling with the sustainable disposal of a growing volume of retired wind turbine blades. While these blades contain recyclable composite materials (e.g., glass fiber/epoxy resin), effective recycling methods remain limited. This study characterized the influence of the N2 and CO2 atmospheres on the thermal decomposition of glass fiber/epoxy resin composite through thermogravimetry (TG), TG-Fourier transform infrared spectroscopy (TG-FTIR), and pyrolysis-gas chromatography-mass spectrometry (Py-GC/MS). Both atmospheres exhibited similar mass loss profiles, with devolatilization occurring between 237.80−578.30 °C (N2) and 236.62−553.32 °C (CO2). CO2 inhibited pyrolytic gas formation compared with N2. Pyrolytic oils identified primarily consisted of phenols, alcohols/ethers, and aliphatics, with compositional changes with 400 °C, 500 °C, and 600 °C. At the elevated temperatures, phenolic compounds underwent deoxidation to form aromatic structures, while pyrolytic carbon surface fragments contracted and melted. Not only were residual mass, decomposition rate, and five volatile products jointly optimized, but also their sensitivities to the temperature and atmosphere were quantified using artificial neural networks. This study provides a theoretical and scalable basis for developing sustainable strategies of thermal circularity for waste wind turbine blades. © 2024, The Authors. All rights reserved.

Interspecific Competition Between Levanderina fissa(Dinophyceae) and the Other Three Microalgal Species in Co-Cultures

Growth interactions between the dinoflagellate Levanderina fissa (=Gyrodinium instriatum) and the other three algal bloom species Skeletonema costatum (Bacillariophyceae), Prorocentrum micans (Dinophyceae), and Chattonella marina (Raphido- phyceae) were studied using bi- or tri-algal co-cultures under laboratory conditions. The Lotka-Volterra model was applied to esti- mate the interspecific competition between them. Results showed that the growth of L. fissa was significantly suppressed in both bi-algal and tri-algal cultures, and the maximum cell densities in co-cultures were only between 21% and 69% of that in mono-cul- ture. L. fissa outcompeted P. micans;however, its effects on S. costatum and C. marina were generally less than the effects of their intraspecific competitions based on the competition coefficients. Contrarily, the inhibitory effects of S. costatum and C. marina on L. fissa were much greater. S. costatum outcompeted the other three species in all co-cultures, and its competition coefficients on the other species were more than 10000 times higher than the opposite. The growth of C. marina was suppressed by S. costatum;how- ever, it outcompeted L. fissa and P. micans. The growth of P. micans was most strongly inhibited in co-cultures, and the maximum cell densities in co-cultures were less than 15% of that in mono-species culture. The results suggested that L. fissa had few advan- tages in the interspecific competition. The adaptation to low salinity and the utilization of organic phosphorus might be the important reasons for L. fissa to take the competitive predominance in brackish estuarine environments.
