In this paper, the ALE fluid-solid coupling algorithm is used to realize the simulation of the dynamic opening process of a certain type of airplane drag parachute. Based on the drag parachute working principle and folding process, a finite element model of the main parachute in the installed state is established. In the LS-DYNA software, the parachute opening characteristics of the drag parachute were simulated and analyzed under no crosswind conditions and Stable crosswind conditions at speed 10m/s. The results show that, under the condition of no crosswind, the error of the important parameters such as parachute straightening force, parachute opening dynamic load, parachute deflection angle and test data is less than 15%, and the simulation accuracy meets the engineering development requirements. On this basis, the parameters such as the unfolding shape, deflection angle, and stress state of the parachute parachute under stable crosswind conditions, which are difficult to test and simulate, were studied. This will provide a theoretical basis and technical approach for drag chute design and other types of parachute product research.

The Sand Martin (Riparia riparia) belongs to Hirundinidae. In this study, the complete mitochondrial genome ofR. ripariawas sequenced and characterized. The genome was 17,963 bases in length (GenBank accession no. OK537984) including 13 protein-coding genes, two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and two control regions. The overall base composition ofR. ripariamitogenome was 30.5% for A, 31.8% for C, 14.5% for G, and 23.2% for T. Phylogenetic analysis revealed thatR. ripariawas genetically closest to the species of genusTachycineta.R. ripariamitogenome could contribute to our understanding of the phylogeny and evolution of this species.

Background: Ponatinib (Iclusig) is an oral tyrosine kinase BCR-ABL inhibitor for treating patients with Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph⁺ ALL) and chronic myeloid leukemia (CML) who are resistant to the therapies with other tyrosine kinase inhibitors. However, adverse cardiovascular events caused by Ponatinib are a serious issue that affects patients' survival rates. Thus, it is necessary to search for candidate drugs to reduce the cardiovascular toxicity of Ponatinib. Purpose: To investigate the effects of Aspirin on Ponatinib-induced cardiovascular toxicity in zebrafish. Methods: AB strain of wild type zebrafish (Danio rerio), Tg (cmlc2: GFP) transgenic zebrafish, and Tg (gata1: dsRed) transgenic zebrafish were used as in vivo models to assess survival, blood flow, cardiac morphology, and function. Thrombus formation was detected using O-dianisidine staining. The transcriptome of zebrafish larvae treated with Ponatinib was assessed using RNA sequencing. Results: Ponatinib not only reduced survival rate but also caused cardiovascular toxic events such as pericardial edema, abnormal heart structure, low heart rate, and thrombosis. In addition, whole-body transcriptome analysis showed that Ponatinib up-regulated the expression of cyclooxygenase-1 (COX-1). Compared with other antithrombotic drugs, a COX-1 inhibitor Aspirin more effectively reduced ponatinib-induced cardiovascular toxicity events and improved the survival rate of zebrafish larvae. Conclusion: Our findings suggest that Aspirin exhibits the potential to reduce Ponatinib-induced cardiovascular toxicity. © 2024 The Authors

To effectively make antibiotic-resistance bacteria (ARB) apoptosis and enable extracellular antibiotic-resistance genes (eARGs) loss of infectiousness, copper-doped zinc ferrite (20CuZFO) photoanode was successfully prepared in this study, and coupled with conductive carbon (C) paper to form a 20CuZFO/C two-electrode system. Besides the hydroxyl radical (·OH) and singlet oxygen (1O2), it also produced hydrogen peroxide (H2O2) under the photoelectrocatalytic (PEC) condition, and H2O2 would react with the dissociated bare Cu atoms to trigger a homogenous fenton rection in the 20CuZFO/C two-electrode system. When the applied voltage was 0.1 V, 20CuZFO/C two-electrode system enabled 5.12 ± 0.05-Log10 CFU/mL of kanamycin-resistance (KanR) E. coli apoptosis in 40 min. Label-free differential proteomics analysis results revealed that the glycolysis process, tricarboxylic acid cycle and oxidation respiratory chain of E. coli (KanR) were impeded, which led to low yields of adenosine triphosphate (ATP). Without sufficient ATP to drive their general secretory pathway (Sec channel), it resulted in their apoptosis. The transformation and regrowth experimental results showed that the KanR gene encoded pKMV plasmids (3.39-Log10 copies/mL) with 3 h of PEC treatment absolutely lost their infectiousness. In summary, this work provides a new route for efficient removal of ARB and eARGs. © 2025 Elsevier B.V.

NeuroExplainer: Fine-Grained Attention Decoding to Uncover Cortical Development Patterns of Preterm Infants

Deploying reliable deep learning techniques in interdisciplinary applications needs learned models to output accurate and (even more importantly) explainable predictions. Existing approaches typically explicate network outputs in a post-hoc fashion, under an implicit assumption that faithful explanations come from accurate predictions/classifications. We have an opposite claim that explanations boost (or even determine) classification. That is, end-to-end learning of explanation factors to augment discriminative representation extraction could be a more intuitive strategy to inversely assure fine-grained explainability, e.g., in those neuroimaging and neuroscience studies with high-dimensional data containing noisy, redundant, and task-irrelevant information. In this paper, we propose such an explainable geometric deep network dubbed as NeuroExplainer, with applications to uncover altered infant cortical development patterns associated with preterm birth. Given fundamental cortical attributes as network input, our NeuroExplainer adopts a hierarchical attention-decoding framework to learn fine-grained attentions and respective discriminative representations to accurately recognize preterm infants from term-born infants at term-equivalent age. NeuroExplainer learns the hierarchical attention-decoding modules under subject-level weak supervision coupled with targeted regularizers deduced from domain knowledge regarding brain development. These prior-guided constraints implicitly maximizes the explainability metrics (i.e., fidelity, sparsity, and stability) in network training, driving the learned network to output detailed explanations and accurate classifications. Experimental results on the public dHCP benchmark suggest that NeuroExplainer led to quantitatively reliable explanation results that are qualitatively consistent with representative neuroimaging studies. © 2023, CC BY-NC-SA.

Unresolved excess accumulation of myelin-derived cholesterol contributes to scar formation after spinal cord injury

Background: Spinal cord injury triggers complex pathological cascades, resulting in destructive tissue damage and incomplete tissue repair. Scar formation is generally considered as a barrier for regeneration in central nervous system (CNS), while the intrinsic mechanism of scar-forming after spinal cord injury has not been completed deciphered. Methods: We assessed cholesterol hemostasis in spinal cord lesions and injured peripheral nerves using confocal reflection microscopy and real-time PCR analyses. The involvement of the proteins, which were predicted to promote cholesterol efflux in spinal cord lesions, were assessed with Liver X receptor (LXR) agonist and Apolipoprotein E (APOE) deficiency. The role of reverse cholesterol transport (RCT) in cholesterol clearance was examined in APOE KO mice injured sciatic nerves and myelin-overloaded macrophages in vitro. Finally, we determined the consequence of excess cholesterol accumulation in CNS by transplantation of myelin into neonatal spinal cord lesions. Results: We found that excess cholesterol accumulates in phagocytes and is inefficiently removed in spinal cord lesions in young-adult mice. Interestingly, we observed that excessive cholesterol also accumulates in injured peripheral nerves, but is subsequently removed by RCT. Meanwhile, preventing RCT led to macrophage accumulation and fibrosis in injured peripheral nerves. Furthermore, the neonatal mouse spinal cord lesions are devoid of myelin-derived lipids, and able to heal without excess cholesterol accumulation. We found that transplantation of myelin into neonatal lesions disrupts healing with excessive cholesterol accumulation, persistent macrophage activation and fibrosis, indicating myelin-derived cholesterol plays a critical role in impaired wound healing. Conclusion: Altogether, our data suggest that the central nervous system (CNS) lacks an efficient approach for cholesterol clearance, resulting in excessive accumulation of myelin-derived cholesterol

UOD: Universal One-shot Detection of Anatomical Landmarks

One-shot medical landmark detection gains much attention and achieves great success for its label-efficient training process. However, existing one-shot learning methods are highly specialized in a single domain and suffer domain preference heavily in the situation of multi-domain unlabeled data. Moreover, one-shot learning is not robust that it faces performance drop when annotating a sub-optimal image. To tackle these issues, we resort to developing a domain-adaptive one-shot landmark detection framework for handling multi-domain medical images, named Universal One-shot Detection (UOD). UOD consists of two stages and two corresponding universal models which are designed as combinations of domain-specific modules and domain-shared modules. In the first stage, a domain-adaptive convolution model is self-supervised learned to generate pseudo landmark labels. In the second stage, we design a domain-adaptive transformer to eliminate domain preference and build the global context for multi-domain data. Even though only one annotated sample from each domain is available for training, the domain-shared modules help UOD aggregate all one-shot samples to detect more robust and accurate landmarks. We investigated both qualitatively and quantitatively the proposed UOD on three widely-used public X-ray datasets in different anatomical domains (i.e., head, hand, chest) and obtained state-of-the-art performances in each domain. Idiopathic pulmonary fibrosis (IPF) is the most common type of idiopathic interstitial pneumonia and has one of the poorest prognosis. However, the molecular mechanisms underlying IPF progression remain largely unknown. In this study, we determined that IL-24, an IL-20 subfamily cytokine member, was increased both in the serum of IPF patients and the bronchoalveolar lavage fluid (BALF) of mice following bleomycin (BLM)-induced pulmonary fibrosis. As a result, IL-24 deficiency protected mice from BLM-induced lung injury and fibrosis. Specifically, loss of IL-24 significantly attenuated transforming growth factor β1 (TGF-β1) production and reduced M2 macrophage infiltration in the lung of BLM-induced mice. Mechanistically, IL-24 alone did not show a perceptible impact on the induction of M2 macrophages, but it synergized with IL-4 to promote M2 program in macrophages. IL-24 suppressed IL-4-induced expression of suppressor of cytokine signaling 1 (SOCS1) and SOCS3, through which it enhanced signal transducer and activator of transcription 6/peroxisome proliferator-activated receptor gamma (STAT6/PPARγ) signaling, thereby promoting IL-4-induced production of M2 macrophages. Collectively, our data support that IL-24 synergizes with IL-4 to promote macrophage M2 program contributing to the development of pulmonary fibrosis.

Nanoscale hydrated zirconium oxide (HZO) holds great potential in groundwater purification due to its ability to form inner-sphere coordination with arsenate. Despite being frequently encapsulated inside bulky hosts, the adsorption mechanism of arsenate onto HZO, including the intrinsic coordination constants (Kintr) and the effects of nanoconfinement, remains scantily explored. In this study, we investigated the Zr-As coordination configuration and corresponding Kintr values using surface complexation modeling (SCM), and distinguished the nanoconfinement effects by comparing bare HZO with the HZO nanoparticles (NPs) encapsulated inside the strongly basic anion exchanger D201. Both samples share a similar crystalline amorphous structure and surface site density (~6 mmol/g-Zr). Potentiometric titration suggests that the protonated form (≡ZrOH2+) is the dominant surface Zr species, and D201 enhances the protonation degree by reducing electrostatic repulsion to H+. Batch adsorption experiments demonstrate that the nanocomposite (HZO@D201) has a higher adsorption capacity in a wide range of pH (3-10) and ionic strength (0.001-0.1 M NaNO3) than bare HZO. SCM simulations reveal that the coordination configuration of diprotonated monodentate mononuclear (MM-H2) dominates at pH 3-6, while deprotonated bidentate binuclear (BB-H0) dominates at pH>7. For each configuration, the Kintr values of HZO@D201 (10−0.66 and 10−16.10, respectively) are significantly higher than those of bare HZO (10−12.24 and 10−44.42, respectively), indicating a superior chemical bonding affinity caused by nanoconfinement. The obtained Kintr values are used to predict arsenate adsorption isotherms in pH 3 and 9, and the results align with the SCM simulation outcomes, verifying their reliability. © 2023, The Authors.
