There is a considerable amount of work that shows the biomagnetism of organic components without ferromagnetic components at the molecular level,but it is of great challenge to cover the giant gap of biomagnetism between their experimental and theoretical *** we show that the diamagnetism of aromatic peptides is greatly enhanced for about 11 times by self-assembling,reaching two orders of magnitude higher than the mass susceptibility of pure *** self-assembly of aromatic rings in the peptide molecules plays the key role in such a strong diamagnetism.

Voltage-gated sodium channel 1.7(Nav1.7)remains one of the most promising drug targets for pain *** the current study,we conducted a high-throughput screening of natural products in our in-house compound library to discover novel Nav1.7 inhibitors,then characterized their pharmacological *** identified 25 naphthylisoquinoline alkaloids(NIQs)from Ancistrocladus tectorius to be a novel type of Nav1.7 channel *** stereostructures including the linkage modes of the naphthalene group at the isoquinoline core were revealed by a comprehensive analysis of HRESIMS,1D,and 2D NMR spectra as well as ECD spectra and single-crystal X-ray diffraction analysis with Cu Kα *** the NIQs showed inhibitory activities against the Nav1.7 channel stably expressed in HEK293 cells,and the naphthalene ring in the C-7 position displayed a more important role in the inhibitory activity than that in the C-5 *** the NIQs tested,compound 2 was the most potent with an IC50 of 0.73±0.03 μ*** demonstrated that compound 2(3 μM)caused dramatical shift of steady-state slow inactivation toward the hyperpolarizing direction(V1/2 values were changed from-39.54±2.77 mV to-65.53±4.39 mV,which might contribute to the inhibition of compound 2 against the Nav1.7 *** acutely isolated dorsal root ganglion(DRG)neurons,compound 2(10 μM)dramatically suppressed native sodium currents and action potential *** the formalin-induced mouse inflammatory pain model,local intraplantar administration of compound 2(2,20,200 nmol)dose-dependently attenuated the nociceptive *** summary,NIQs represent a new type of Nav1.7 channel inhibitors and may act as structural templates for the following analgesic drug development.

Multigenerational Metastatic Effects of Perfluorooctanoic Acid (PFOA) on Drosophila Melanogaster with Metabolic Reprogramming and Dysrhythmia

Perfluorooctanoic acid (PFOA) exposure correlated with various cancers and their mortality. However, it remained unanswered whether its persistent exposure could influence metastasis which contributes ~90% to cancer mortality. In the present study, long-term effects of PFOA were measured in Drosophila melanogaster over 3 consecutive generations. In the morning-eclosed (AM) adult flies, PFOA significantly promoted tumor invasion rates and distances which increased over generations. Regarding metabolic reprogramming, PFOA disturbed the expressions of Glut1 and Pdk1, activities and contents of FASN1 (fatty acid synthase), ACC (acetyl-CoA carboxylase) and SREBP1 (sterol regulatory element binding protein). Regarding antioxidant responses, PFOA exposure generated provoked oxidative stress via H2O2 and stimulated antioxidants including glutathione (GSH), catalase (CAT), melatonin, serotonin and cortisol, with downregulations on PI3K/AKT pathways and upregulations on MAPK ones. The biochemical and molecular effects altered over generations. In the afternoon-eclosed (PM) adult flies, the metastasis of PFOA was more deteriorated than in AM adults. The significant influences by dysrhythmia were also observed in the multigenerational effects of PFOA on the metabolism reprogramming and antioxidant responses. The effects on rhythm regulating gene expressions and protein levels explained the dysrhythmia and also indicated close interactions among metabolism reprogramming, antioxidant responses and rhythm regulation. © 2023, The Authors. Experimental Study on Surface Integrity and Subsurface Damage of Fused Silica in Ultra-Precision Grinding

To realize low-damage ultra-precision grinding on fused silica, the surface quality and subsurface damage (SSD) distribution with fine-grained grinding wheel under different depth-of-cut and cutting speed are experimentally studied. The material removal mechanism under different grinding parameters is revealed by calculating undeformed chip thickness and observed with the help of transmission electron microscopy. The results show that brittle-ductile surfaces and ductile-like surfaces are generated during grinding. With the decrease of depth-of-cut and the increase of wheel cutting speed, the ultra-precision grinding changes to ductile-regime grinding with plastic flow removal. Besides, the surface roughness (SR) and SSD depth are reduced. The fracture defects such as fractured pits and grinding streaks on brittle-ductile surface gradually decrease. Instead, a ductile-like surface covered with grinding streaks is found. On brittle-ductile surfaces, the nonlinear relationship SSD∝SR4/3 is no longer proper under the influence of plastic flow. Using surface roughness Ra to predict SSD depth is more accurate. When depth-of-cut is 1 μm, cutting speed is 23.4 m/s and the material removal mode is dominated by plastic flow removal, the surface Ra is improved to 2.0 nm and there is no crack but only a 3.4 nm deep plastic flow layer in subsurface after grinding. © 2021, CC BY.

Suspension is the common preparation whose absorption rate is second only to the solution. However, problems like the difficulty in screening excipients and the complicated screening process limit its application to a certain extent. In this study, the Compound sulfachlorpyridazine sodium Powder(CSSP)was used as the research vector to explore the effects of wetting agent(WA), mixing processes between excipients and drug on the preparation stability from the perspectives of the change of the surface free energy between the drug powder and solvent, the reduction of the particle size of the drug, and the interaction between the drug and the excipients, serving the contact angle, sedimentation volume ratio, re-dispersibility, particle size, and appearance as indicators. In addition to that, taking the number density distribution, mean square displacement (MSD), and Van der Waals force (VDW) as indicators, employ molecular dynamics (MD) simulation techniques to explore the differences in the wetting ability of wetting agents (WAs) and the wetting mechanism of WAs on drug molecules. Suitable indicators were screened to provide new ideas for the initial screening of wetting agents from the simulation level and other solutions in tedious and time-consuming problems of experimental excipients screening. © 2023, The Authors. Jasmonate(JA)regulates various aspects of plant growth and development and stress responses,with prominent roles in male reproductive development and defenses against herbivores and necrotrophic ***-ZIM DOMAIN(JAZ)proteins are key regulators in the JA signaling pathway and function to repress the expression of JA-responsive ***,we show that JAZ proteins directly interact with several chromatin-associated Polycomb proteins to mediate repressive chromatin modifications at JA-responsive genes and,thus,their transcriptional repression in *** analyses revealed that the developmental defects,including anther and pollen abnormalities,resulting from loss or block of JA signaling were partially rescued by loss of Polycomb protein-mediated chromatin silencing(Polycomb repression).We further found that JAZ-mediated transcriptional repression during anther and pollen development requires Polycomb proteins at four key Regulatory *** of genome-wide occupancy of a Polycomb factor and transcriptome reprogramming in response to JA revealed that Polycomb repression is involved in the repression of various JA-responsive *** together,our study reveals an important chromatin-based mechanism for JAZ-mediated transcriptional repression and JA signaling in plants.

Social avoidance behavior in male tree shrews and prosocial behavior in male mice toward unfamiliar conspecifics in the laboratory

Adult male tree shrews vigorously defend against intruding male conspecifics. However, the characteristics of social behavior have not been entirely explored in these males. In this study, male wild-type tree shrews(Tupaia belangeri chinensis)and C57 BL/6 J mice were first allowed to familiarize themselves with an open-field apparatus. The tree shrews exhibited a short duration of movement(moving) in the novel environment, whereas the mice exhibited a long duration of movement. In the 30 min social preference-avoidance test, target animals significantly decreased the time spent by the experimental tree shrews in the social interaction(SI)zone, whereas experimental male mice exhibited the opposite. In addition, experimental tree shrews displayed a significantly longer latency to enter the SI zone in the second 15 min session(targetpresent) than in the first 15 min session(targetabsent), which was different from that found in *** behavioral patterns in response to a conspecific male were also observed in male tree shrews and mice in the first, second, and third 5 min periods. Thus, social behaviors in tree shrews and mice appeared to be time dependent. In summary,our study provides results of a modified social preference-avoidance test designed for the assessment of social behavior in tree shrews. Our findings demonstrate the existence of social avoidance behavior in male tree shrews and prosocial behavior in male mice toward unfamiliar conspecifics. The tree shrew may be a new animal model, which differs from mice, for the study of social avoidance and prosocial behaviors.

We collected blood from coronavirus disease 2019(COVID-19)convalescent individuals and investigated SARS-CoV-2-specific humoral and cellular immunity in these discharged ***-up analysis in a cohort of 171 patients at 4-11 months after the onset revealed high levels of IgG antibodies.A total of 78.1%(164/210)of the specimens tested positive for neutralizing antibody(NAb).SARS-CoV-2 antigen peptide pools-stimulated-IL-2 and-IFN-y response can distinguish COVID-19 convalescent individuals from healthy ***,NAb survival was significantly affected by the antigen peptide pools-stimulated-IL-2 response,-IL-8 response,and-IFN-y *** antigen peptide pools-activated CD8+T cell counts were correlated with *** antigen peptide pools-activated natural killer(NK)cell counts in convalescent individuals were correlated with NAb and disease *** data suggested that the development of NAb is associated with the activation of T cells and NK *** work provides a basis for further analysis of the protective immunity to SARS-CoV-2 and for understanding the pathogenesis of *** also has implications for the development of an effective vaccine for SARS-CoV-2 infection.

A Novel Petal-Type Battery Thermal Management System with Dual Phase Change Materials

Phase change material (PCM) cooling is a promising approach to battery thermal management, but its wide application is limited by the narrow phase change temperature range. When the PCM temperature is below the melting point, PCM is equivalent to a purely thermally conductive material. When the ambient temperature exceeds the phase change temperature of PCM, PCM melts completely and cannot play the role of battery thermal management. Therefore, a novel petal-type battery thermal management system (BTMS) with dual PCMs is proposed in this paper to solve the environmental adaptability problem of PCM cooling. The thermal performance of the system at different ambient temperatures and different discharge rates is investigated by numerical simulation methods. The results show that the maximum temperature of battery at 2 C discharge rate is 30.49 ℃, 34.16 ℃ and 43.70 ℃ at ambient temperatures of 20 ℃, 30 ℃ and 40 ℃, respectively. Compared with the BTMS filled with a single PCM, the thermal performance of the proposed BTMS is particularly good at the high operating temperature represented by 40 ℃. At lower ambient temperatures such as 20 ℃ and 30 ℃, the thermal performance is not the best, but it fully meets the requirements of battery thermal management. Furthermore, the above BTMS is optimized using fin-reinforced heat transfer technology in this paper. The solution with the optimal fin arrangement can reduce the maximum temperature difference by 36.2% and 42.8% and the maximum temperature rise by 5.53% and 29.19% at 30 ℃ and 40 ℃, respectively. © 2022, The Authors.
