General expert consensus on the application of network pharmacology in the research and development of new traditional Chinese medicine drugs

The research and development of new traditional Chinese medicine(TCM)drugs have progressively established a novel system founded on the integration of TCM theory,human experience,and clinical trials(termed the“Three Combinations”).However,considering TCM's distinctive features of“syndrome differentiation and treatment”and“multicomponent formulations and complex mechanisms”,current TCM drug development faces challenges such as insufficient understanding of the material basis and the overall mechanism of action and an incomplete evidence chain ***,significant obstacles persist in gathering human experience data,evaluating clinical efficacy,and controlling the quality of active ingredients,which impede the innovation process in TCM drug *** pharmacology,centered on the“network targets”theory,transcends the limitations of the conventional“single target”reductionist research *** emphasizes the comprehensive effects of disease or syndrome biological networks as targets to elucidate the overall regulatory mechanism of TCM *** approach aligns with the holistic perspective of TCM,offering a novel method consistent with TCM's holistic view for investigating the complex mechanisms of TCM and developing new TCM *** is internationally recognized as a“next-generation drug research model”.To advance the research of new tools,methods,and standards for TCM evaluation and to overcome fundamental,critical,and cutting-edge technical challenges in TCM regulation,this consensus aims to explore the characteristics,progress,challenges,applicable pathways,and specific applications of network pharmacology as a new theory,method,and tool in TCM drug *** goal is to enhance the quality of TCM drug research and development and accelerate the efficiency of developing new TCM products.

This article discusses the problems of developing rational technologies and remote control systems for the processes of water injection into the reservoir (pumping stations to maintain reservoir pressure) and oil production using mechanized methods (rod pumping units, electric centrifugal pumps, etc.) in the late stage of oil field operation, which is characterized by difficult oil *** main problems of the process of pumping water into the formation to maintain the pressure necessary to displace the oil-water mixture to production wells, and ways to extract them from production wells, are explored. For this purpose, the concentration of water and oil in the total amount of produced fluid is determined depending on the flow rate of the well, and the function of the dependence of the concentration on the flow rate for selected wells is formulated. The problem of optimizing the well flow rate and methods for solving them are *** issue of technical and computer software for the optimization problem is considered. A block diagram of the system for rational control and management of oil production processes in the late stage of operation is presented, which allows the implementation of the created technologies, algorithms and software for pumping water into the reservoir through pumping stations for injection and production from wells using deep-well pumps. oil production by achieving an increase in the oil/water ratio when extracting oil using mechanized methods. © 2024 IEEE.

We propose an intelligent correlation system for control and initiation of malfunctions of sucker-rod pumping units (SRPU). It is shown that by memorizing the reference force curve of dynamometer charts of characteristic malfunctions and alternately comparing them with the force curves of the current dynamometer charts by the number of the reference dynamometer chart, at which the estimate of the correlation coefficient takes a maximum value, it is possible to identify the current malfunction of SRPU. Implementation of this technology in oil production with the help of inexpensive technical means allows at the expense of significant reduction of control period and early troubleshooting, as a result of increase of overhaul period, to considerably increase the profitability of oil wells. © 2023 IEEE.

Alzheimer’s disease(AD)is a neurodegenerative disease with clinical hallmarks of progressive cognitive *** effects of the Ab-Tau cascade reaction are tightly implicated in AD pathology,and microglial NLRP3 inflammasome activation drives neuronal ***,the underlying mechanism of how Ab mediates NLRP3 inflammasome remains ***,we determined that oligomeric Ab(o-Ab)bound to microglial Kv2.1 and promoted Kv2.1-dependent potassium efflux to activate NLRP3 inflammasome resulting in neuronal tauopathy by using Kv2.1 inhibitor drofenine(Dfe)as a *** underlying mechanism has been intensively investigated by assays with Kv2.1 knockdown in vitro(si-Kv2.1)and in vivo(AAV-ePHP-si-Kv2.1).Dfe deprived o-Ab of its capability to promote microglial NLRP3 inflammasome activation and neuronal Tau hyperphosphorylation by inhibiting the Kv2.1/JNK/NF-kB pathway while improving the cognitive impairment of 5FAD-AD model *** results have highly addressed that the Kv2.1 channel is required for o-Ab-driven microglial NLRP3 inflammasome activation and neuronal tauopathy in AD model mice and highlighted that Dfe as a Kv2.1 inhibitor shows potential in the treatment of AD.

Diabetic peripheral neuropathy(DPN)is a common complication of diabetes,which has yet no curable *** and mitochondrial dysfunction are tightly linked to DPN pathology.G-protein-coupled receptor 40(GPR40)is predominantly expressed in pancreatic β-cells,but also in spinal dorsal horn and dorsal root ganglion(DRG)neurons,regulating neuropathic *** previously have reported that vincamine(Vin),a monoterpenoid indole alkaloid extracted from Madagascar periwinkle,is a GPR40 *** this study,we evaluated the therapeutic potential of Vin in ameliorating the DPN-like pathology in diabetic *** STZ-induced type 1(T1 DM)and db/db type 2 diabetic(T2DM)mice were used to establish late-stage DPN model(DPN mice),which were administered Vin(30 mg·kg-1·d-1,i.p.)for 4 *** showed that Vin administration did not lower blood glucose levels,but significantly ameliorated neurological dysfunctions in DPN *** administration improved the blood flow velocities and blood perfusion areas of foot pads and sciatic nerve tissues in DPN *** demonstrated that Vin administration protected against sciatic nerve myelin sheath injury and ameliorated foot skin intraepidermal nerve fiber(IENF)density impairment in DPN ***,Vin suppressed NLRP3 inflammasome activation through either β-Arrestin2 or β-Arrestin2/IκBα/NF-κB signaling,improved mitochondrial dysfunction through CaMKKβ/AMPK/SIRT1/PGC-1 α signaling and alleviated oxidative stress through Nrf2 signaling in the sciatic nerve tissues of DPN mice and LPS/ATP-treated RSC96 *** the above-mentioned beneficial effects of Vin were abolished by GPR40-specific knockdown in dorsal root ganglia and sciatic nerve ***,these results support that pharmacological activation of GPR40 as a promising therapeutic strategy for DPN and highlight the potential of Vin in the treatment of this disease.

Ischemic stroke(IS)is a globally life-threatening ***,few therapeutic medicines are available for treating IS,and rt-PA is the only drug approved by the US Food and Drug Administration(FDA)in the *** fact,many agents showing excellent neuroprotection but no blood flow-improving activity in animals have not achieved ideal clinical efficacy,while thrombolytic drugs only improving blood flow without neuroprotection have limited their wider *** address these challenges and meet the huge unmet clinical need,we have designed and identified a novel compound AAPB with dual effects of neuroprotection and cerebral blood flow *** significantly reduced cerebral infarction and neural function deficit in tMCAO rats,pMCAO rats,and IS rhesus monkeys,as well as displayed exceptional safety profiles and excellent pharmacokinetic properties in rats and *** has now entered phase I of clinical trials fighting IS in China.
