Effects of aqueous leaf extract of Tridax procumbens on contractile activity of corpus cavernosum in N-nitro-L-arginine methyl ester-induced hypertensive male rats

Objective： This study investigated the effects of aqueous leaf extract of Tridax procumbens （ALETP） on con- tractile activity of corpus cavernosum in N-nitro-L-arginine methyl ester （L-NAME）-induced hypertensive male rats. Methods： Twenty normal, adult male rats （130-150 g） were divided into four groups of five rats each. Group I （control） was given normal saline （0.6 mL/kg） and group II was given L-NAME （40 mg/kg） for 6 weeks. Groups Ill and IV also received L-NAME （40 mg/kg） for 6 weeks but were further co-treated with 100 and 200 mg/kg of ALETP, respectively, from week 4 to week 6. All treatments were given orally. Strips of corpus cavernosum from each of the four groups were exposed to increasing concentrations of acetyl~ choline （ACh） and sodium nitroprusside （SNP）（10^-9-10^-5 tool/L） after contraction with phenylephrine （10^-7 mol/L） to test for a dose-response effect, Response to potassium and calcium was also measured after cumulatively adding potassium and calcium （10-50 mmol/L） to potassium- and calcium-free organ chamber. Isometric contractions were recorded through an Ugo Basile data capsule acquisition system. Results： Mean arterial blood pressure was significantly reduced in the ALETP co-treated group compared to the control and L-NAME-only groups （P 〈 0.05）. Cavernosa strips from ALETP co-treated rats exhibited significant inhibition of contraction in response to phenylephrine, potassium chloride, and calcium chlo- ride （P 〈 0.05）. Relaxation in response to Ach and SNP was also significantly impaired in cavernosa strips from the L-NAME-only treated group （P 〈 0.05）, while ALETP co-treated groups showed enhanced per- centage relaxation. Conclusion： ALETP treatment of L-NAME-induced hypertensive rats promotes a relaxant effect on isolated cavernosa strips. ALETP shows potential in correcting erectile dysfunction in hypertension.

Maternal treatment with dexamethasone during lactation delays male puberty and disrupts reproductive functions via hypothalamic-pituitary-gonadal axis alterations

The effects of maternal treatment with dexamethasone during lactation on pubertal timing, serum hormonal profile and sperm indices in the male offspring were assessed. Twenty lactating dams were divided into 4 groups (n = 5). Group 1 was administered subcutaneously 0.02 ml/100 g/day normal saline at lactation days 1-21. Groups 2-4 were administered subcutaneously 100 μg/kg/day dexamethasone (Dex) at lactation days 1-7, 1-14, and 1-21 respectively. Results showed that there was significant reduction in serum testosterone in the DexLD 1-7 (p < 0.05), DexLD 1-14 (p < 0.01) and DexLD 1-21 (p < 0.001) relative to control. In addition there was a significant reduction in serum FSH and LH in the DexLD 1-7 (p < 0.01), DexLD 1-14 (p < 0.001) and DexLD 1-21 (p < 0.001) when compared with the control. Treatment with dexamethasone during lactation significantly increased the days of preputial separation in the DexLD 1-7 (p < 0.05), DexLD 1-14 (p < 0.05) and DexLD 1-21 (p < 0.001) relative to control. Maternal treatment with dexamethasone throughout lactation period also significantly reduced sperm counts (p < 0.001), motility (p < 0.01) and increased percentage abnormal sperm (p < 0.001) in the offspring when compared with the control. In conclusion, maternal treatment with dexamethasone during lactation may induce delayed puberty and disrupt reproductive functions by altering activities at hypothalamic-pituitary-gonadal axis in the male offspring. © 2015 Elsevier Ireland Ltd.

Maternal treatment with dexamethasone during lactation alters reproductive functions and increases serum corticosterone in the male offspring. Excess corticosterone may induce oxidative stress. This study was designed to evaluate the effects of maternal treatment with dexamethasone during lactation on oxidative stress indices in the testis and epididymis of a male offspring. Twenty lactating dams were divided into 4 groups (n = 5). Group 1 was administered 0.02 ml/100 g/day normal saline subcutaneously at lactation days 1–21. Groups 2, 3, and 4 were administered 100 μg/kg/day dexamethasone (Dex) subcutaneously at lactation days (LD) 1–7, 1–14, and 1–21 respectively. Testis and epididymis malondialdehyde (MDA), catalase and superoxide dismutase (SOD) activities were measured as markers of oxidative stress. The mean testis and epididymis MDA were significantly raised (p < 0.05) in the dexamethasone-treated groups when compared with control. This was accompanied with a significant reduction (p < 0.05) in SOD and catalase activities in these tissues in the DexLD 1–21, when compared with control. The mean total protein level of the epididymis was significantly reduced (p < 0.05) in all the dexamethasone treated groups when compared with control. In conclusion, maternal treatment with dexamethasone during the first two weeks of lactation and throughout lactation may lead to increase in oxidative stress in the testis and epididymis of the male offspring of Wistar rats. © 2017 Elsevier B.V.

Background: Polycystic ovary syndrome (PCOS), also known as the Stein-Leventhal syndrome is one of the most common causes of anovulation, infertility and hyperandrogenism in women, affecting between 5–10 % of women of reproductive age (12–35 years) worldwide. Despite substantial effort to define the cause of PCOS, its pathophysiology remains poorly understood. Consequently, determining the mechanisms of PCOS and the possible treatment is the major goal of medical research in endocrine and reproductive physiology. Aim: To investigate the mechanism of ovarian metabolic changes in dehydroepiandrosterone (DHEA)-induced polycystic ovary in Wistar rats treated with vitamin C. Methods: Twenty-eight immature female Wistar rats weighing (16–21 g) were randomly divided into four groups (n = 7/group): group I served as control and was given water, group II were injected with DHEA (6 mg/100 g in 0.2 ml corn oil subcutaneously to induce PCOS condition), group III received 150 mg/kg BW of Vitamin C orally, group IV were co-administered with 6 mg/kg BW DHEA in 0.2 ml of corn oil subcutaneously and 150 mg/kg BW of Vitamin C orally. All treatments lasted for 15 days. Twenty-four hours after the last administration, the rats were sacrificed by cervical dislocation. Blood samples and ovaries were collected for reproductive hormonal analysis, biochemical and histopathological analysis. The expressions of mRNA androgen receptor gene in the ovary were determined by real-time reverse transcriptase polymerase chain reaction. All data were analysed using one-way ANOVA. Results: There was a significant decrease (p < 0.05) in the antioxidant and metabolic enzyme activity in the DHEA treated group compared with the control group. DHEA co-administration with Vitamin C showed a significant decrease in Malondialdehyde, cytokines and Estrogen and a significant increase (p < 0.05) in antioxidant and metabolic enzymes compared with DHEA treated group only. Objective:To investigate the protective effect of vitamin E in dehydroepiandrosterone(DHEA)-induced polycystic ovary in ***:Premature female Wistar rats were randomly allocated into four groups,with 7 rats in each ***栺received corn oil(vehicle)and served as the control group;group栻received 0.2 mL of 0.06 mg/g DHEA in corn oil;group栿received 200 mg/kg vitamin E;group桇received DHEA plus vitamin *** treatments lasted for 15 days,with DHEA administered subcutaneously,while vitamin E and corn oil were administered *** the experiment,serum samples and ovaries were harvested for biochemical,immunohistochemical,hormonal,and histological *** ovarian mRNA expression of androgen receptor was analyzed by reverse transcriptase quantitative polymerase chain reaction(qPCR).Results:The antioxidant and metabolic enzyme activity significantly decreased in the DHEA-treated rats compared to the control rats(P<0.05).Administration of vitamin E to DHEAtreated rats significantly decreased cytokines and malondialdehyde compared to the DHEA-treated *** histological analysis showed reduced atretic and cystic ovaries,increased E-cadherin and Bcl-2 expression,and reduced expression of Bax in the DHEAtreated rats co-treated with vitamin *** mRNA expression of androgen receptor was upregulated in the DHEA-treated rats compared to the control ***:Vitamin E ameliorates the hyperandrogenic effect of DHEA-induced polycystic ovaries via metabolic,antioxidant,and anti-apoptotic pathways.

Quantity of water is a major concern, but the quality is of more urgent concern due to heavy metal pollution. In this study, the focus was to develop adsorbents (tannin resin (TR) and iron-doped tannin resin (Fe-TR)) from invasive Acacia mearnsii for Pb²⁺ and Ni²⁺ in solution. The developed TR and Fe-TR were prepared directly by crosslinking and iron-grafting;respectively with thermal stable at ≤ 350 °C. Surface functionality analysis revealed the presence of chelating groups at ≈ 1688 and 3661 cm⁻¹. The crystallite sizes of the adsorbents were smaller compared to tannin;however, the % carbon and nitrogen were significantly higher. Also, the Brunauer–Emmet–Teller study revealed that iron-grafting of tannin after crosslinking increases the surface area, pore volume and pore size. Furthermore, the surface morphology of adsorbents indicated a well-defined structure with the formation of a whitish deposit and rough lump-like after ion uptake. Both adsorbents showed a type II isotherm with adsorption capacities of 40.650 mg/g and 13.763 mg/g for Pb²⁺and 120.63 mg/g and 110.74 for Ni²⁺ ion by TR and Fe-TR;respectively. The absorbate-adsorbent interactions were observed to be endothermic with negative and positive Gibb's free energy values for the adsorption of Pb²⁺ and Ni²⁺;respectively. The adsorption kinetics of Ni²⁺ is governed by several models;however, Pb²⁺ interaction is regulated by Pseudo-second-order model. Furthermore, the data generated offered the possibility of both chemisorption and physisorption processes;highlighting the flexibility and multifunctionality of tannin traps for the erasure of ion impurities in wastewater. © 2021 The Author(s)
