Background: India accounts for a third of the global dengue disease burden, out of an annual global incidence of 100–400 million infections. The non-linearity and regional heterogeneity inherent in climate-dengue relationship and the lack of consistent data makes it difficult for preparing predictions suited for effective disease prevention. The current study investigates the climate-dengue links in India, and presents a model framework for both near-term and future dengue predictions. Methods: Statistical techniques and machine learning methods are used to evaluate the climate-dengue links and prepare the dengue model framework. Mortality data for a dengue hotspot region, Pune, from 2004 to 2015 is analyzed along with the meteorological conditions, considering the non-linearity and the time-lag between them. The future climate simulations from the Coupled Model Intercomparison Project phase6 models are used as inputs in the dengue model for generating dengue future projections up to 2100, under different future emission pathways. Findings: The current study finds that temperature, rainfall, and relative humidity impact dengue mortality in Pune at a lag of 2–5 months. Moderate rains spread in time leads to an increase in dengue mortality, whereas heavy rains reduce it through the flushing effect. Our dengue model provides a reliable forecast accuracy at a lead time of two months. In a changing climate, dengue mortalities over Pune are projected to increase by 13% in the near-term (2021–2040) and 30–112% in the long-term (2081–2100) under different emission scenarios in response to the changes in rainfall patterns and increase in temperature and relative humidity. We performed a deep proteogenomic analysis of bulk tumor and laser microdissection enriched tumor cell populations from high-grade serous ovarian cancer (HGSOC) tissue specimens spanning a broad spectrum of purity. We identified patients with longer progression-free survival had increased immune-related signatures and validated proteins correlating with tumor-infiltrating lymphocytes in 65 tumors from an independent cohort of HGSOC patients, as well as with overall survival in an additional 126 HGSOC patient cohort. We identified that homologous recombination deficient (HRD) tumors are enriched in pathways associated with metabolism and oxidative phosphorylation that we validated in independent patient cohorts. We further identified that polycomb complex protein BMI-1 is elevated in HR proficient (HRP) tumors, that elevated BMI-1 correlates with poor overall survival in HRP but not HRD HGSOC patients, and that HRP HGSOC cells are uniquely sensitive to BMI-1 inhibition.

Background: Diabetes is one of the leading causes of death and disability worldwide, and affects people regardless of country, age group, or sex. Using the most recent evidentiary and analytical framework from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD), we produced location-specific, age-specific, and sex-specific estimates of diabetes prevalence and burden from 1990 to 2021, the proportion of type 1 and type 2 diabetes in 2021, the proportion of the type 2 diabetes burden attributable to selected risk factors, and projections of diabetes prevalence through 2050. Methods: Estimates of diabetes prevalence and burden were computed in 204 countries and territories, across 25 age groups, for males and females separately and combined;these estimates comprised lost years of healthy life, measured in disability-adjusted life-years (DALYs;defined as the sum of years of life lost [YLLs] and years lived with disability [YLDs]). We used the Cause of Death Ensemble model (CODEm) approach to estimate deaths due to diabetes, incorporating 25 666 location-years of data from vital registration and verbal autopsy reports in separate total (including both type 1 and type 2 diabetes) and type-specific models. Other forms of diabetes, including gestational and monogenic diabetes, were not explicitly modelled. Total and type 1 diabetes prevalence was estimated by use of a Bayesian meta-regression modelling tool, DisMod-MR 2.1, to analyse 1527 location-years of data from the scientific literature, survey microdata, and insurance claims;type 2 diabetes estimates were computed by subtracting type 1 diabetes from total estimates. Dural arteriovenous fistulas (DAVFs) are rare but significant intracranial vascular malformations that are usually idiopathic and can lead to severe complications like venous hypertension and intracranial hemorrhage. We present 2 cases of DAVF occurring after venous sinus stenting (VSS) for presumed idiopathic intracranial hypertension (IIH) in a 51-year-old and a 52-year-old female. In both patients, Cognard type 1 DAVF was detected by Catheter angiography and successfully obliterated with Onyx™ embolization. These cases suggest that VSS may either unmask pre-existing DAVFs by altering intracranial pressure dynamics or induce new fistula formation by angiogenic factors or stent-induced hemodynamic changes. Clinicians should maintain an index of suspicion for DAVF in patients with persistent symptoms post-VSS and catheter angiography might be needed to detect the DAVFs.

Type 2 diabetes is an increasingly prevalent disease and patients do not always manage the disease properly. Therefore, creating tools that help diabetics self-manage their condition over time is of the utmost importance. Technological tools that include features meeting this population's needs, with an integrated personalized feedback system, in an environment of gamified incentives, may be the way for developing a sustained app's usage. This work aims to present a still in development, novel approach to managing diabetes type 2 with a focus on data analysis from user-inputs, gamification, and personalized coaching, with an accessible user interface. Thus, we will present some functionalities, in particular the theoretical and practical concept that is behind this development, namely the Transtheoretical model of behavior change to create different profiles, to provide customized feedback according to the user's behavioral stage, and also to ascertain if it induces any improvement at the behavioral level.

ABSTRACTABSTRACTAll-terrain vehicle (ATV) crashes are one of the leading causes of death and injury in agriculture across the United States. Use of ATVs is highly prevalent in rural areas and has seen an increase in their application for a variety of agricultural work tasks. Empirical research on the use of these vehicles for agricultural tasks and associated injuries is limited, especially in the United States. Moreover, little is known about the risk factors associated with ATV-related injuries while doing farm work. A comprehensive review was conducted to evaluate the current injury burden of ATV use in agriculture, the need for future research, and possible solutions related to agricultural ATV safety. Potential injury prevention approaches are evaluated based on the hierarchy of control, including elimination or substitution (using side-by-side vehicles instead of ATVs or modifying ATV design), engineering control (operator protection devices), administrative authority (regulations and standards in the United States and around the world), training, and use of personal protective equipment. In addition, vehicle nomenclature, risk factors, and crash mechanisms are reviewed. Recommendations to decrease the likelihood of ATV crashes and injuries are provided.
