Across many scientific and engineering disciplines, it is important to consider how much the output of a given system changes due to perturbations of the input. Here, we investigate the glassy phase of ±J spin glasses at zero temperature by calculating the robustness of the ground states to flips in the sign of single interactions. For random graphs and the Sherrington-Kirkpatrick model, we find relatively large sets of bond configurations that generate the same ground state. These sets can themselves be analyzed as subgraphs of the interaction domain, and we compute many of their topological properties. In particular, we find that the robustness, equivalent to the average degree, of these subgraphs is much higher than one would expect from a random model. Most notably, it scales in the same logarithmic way with the size of the subgraph as has been found in genotype-phenotype maps for RNA secondary structure folding, protein quaternary structure, gene regulatory networks, as well as for models for genetic programming. The similarity between these disparate systems suggests that this scaling may have a more universal origin. © 2020, CC BY.

Objectives: To evaluate adherence to lifestyle intervention activities in the SINgapore GERiatric Intervention Study to Reduce Cognitive Decline and Physical Frailty (SINGER) study's intensive intervention arm over one year, as part of ongoing process evaluation. The analysis identifies sociodemographic, psychosocial, and vascular risk factors associated with adherence to optimize intervention strategies and improve participant engagement. Methods: Adherence was categorised as non-adherent, low, moderate and high adherence based on participants' attendance. Total adherence was assessed in two ways, i) a composite of all attendance and ii) Latent Class Analysis (LCA) to identify distinct participant subgroups based on adherence to the SINGER study activities. Regression models assessed associations between individual factors with adherence to each type of intervention component and adherence profiles derived from LCA. Results: More than 70 % of participants achieving moderate to high adherence. LCA identified three distinct adherence profiles: Low, Moderate and High adherence. Participants showed higher adherence to vascular and diet components, followed by exercise components, and lower adherence to cognitive components. Factors such as being female and having higher education were associated with higher adherence while living alone was associated with lower adherence. Participants with hypertension had higher overall adherence but lower adherence to vascular management sessions. The complexity, delivery mode and frequency of the activities influenced adherence, with more complex and frequent activities showing lower adherence. Conclusion: The intensive intervention arm showed satisfactory adherence. A comprehensive analysis highlighted the relationship between adherence, sociodemographic factors, and vascular risks, emphasizing the need for targeted strategies to support subgroups with lower adherence and optimize engagement. Chronic pain following peripheral nerve injury is accompanied by increased neuronal activity in the somatosensory cortex. However, whether and how cortical microglia contribute to these changes is less understood. To this end, we applied an optogenetic strategy to specifically target cortical microglia and investigate their function in behavioral pain sensitization. We found that optogenetic activation of microglia in the primary somatosensory cortex (S1) via red-activated channelrhodopsin (ReaChR) triggered pain hypersensitivity and affective-motivational responses in mice. Remarkably, S1-targeted optogenetic stimulation increased microglial landscape changes and ATP release. In addition, optogenetic stimulation altered the microglial proteomic profile, upregulated neuronal c-Fos expression, and enhanced neuronal Ca²⁺ signaling in the S1. Our results provide mechanistic evidence linking cortical microglia with neuronal hyperactivity and chronic pain behaviors. © 2025 The Author(s)

Nontraumatic subperiosteal orbital hemorrhage in a laboring patient with gestational immune thrombocytopenic purpura

Purpose: To describe a case of nontraumatic subperiosteal orbital hemorrhage (NTSOH) in a laboring patient with gestational immune thrombocytopenic purpura. Observations: A 28-year-old G3P2 laboring patient was urgently evaluated by our ophthalmology unit after she developed sudden onset left eye proptosis, headache, and diplopia in the final hour of pushing. The patient's platelet count was markedly decreased at 45,000 and subsequent work-up established a diagnosis of gestational immune thrombocytopenic purpura. On examination, visual acuity was 20/25 bilaterally and there was a minus two restriction in upgaze in the left eye and a left hypotropia in primary position. Computed tomography demonstrated an elliptical, hyperdense collection at the left orbital roof consistent with NTSOH. The patient was deemed clinically stable through serial examinations and symptoms resolved with conservative management. Conclusions and Importance: NTSOH is a rare, potentially sight-threatening condition that requires timely ophthalmological evaluation. To our knowledge, this is the first report in the literature of NTSOH in a laboring patient with gestational immune thrombocytopenic purpura. Consideration of the possibility of NTSOH as a complication in this population may allow for appropriate diagnosis, monitoring, and treatment when indicated. © 2020

The Ireland - Northern Ireland - National Cancer Institute Cancer Consortium was established to improve clinical cancer services and patient care on the island of Ireland, and to foster joint collaboration in cancer research and development. An important part of this Consortium was to develop telemedicine systems to facilitate collaboration between participating partners. We have implemented a telemedicine environment for the Consortium based on the TELESYNERGY/spl reg/ Medical Consultation Workstation developed at the U.S. National Institutes of health. The collaborative use of the system at the three sites has led to the improvement in quality of care in that each patient, regardless of location, can receive an expert assessment and given optimal therapy. The system will be a platform for collaborative clinical and translational research. In the future, this technology will be implemented at additional sites and applied to other medical specialties including cardiology, radiology, and infectious diseases.
