Computational-guided discovery of UDP-glycosyltransferases for lauryl glucoside production using engineered ***

Defining suitable enzymes for reaction steps in novel synthetic pathways is crucial for developing microbial cell factories for non-natural ***,we developed a computational workflow to identify C12 alcohol-active *** workflow involved three steps:(1)assembling initial candidates of putative UDP-glycosyltransferases,(2)refining selection by examining conserved regions,and(3)3D structure prediction and molecular *** sequences from Candida,Pichia,Rhizopus,and Thermotoga,known for lauryl glucoside synthesis via whole-cell biocatalysis,were *** of 240 predicted glycosyltransferases,8 candidates annotated as glycosyltransferases were selected after filtering out those with signal peptides and identifying conserved UDP-glycosyltransferase *** proteins underwent 3D structure prediction and molecular docking with ***3G,a candidate from Rhizopus delemar RA 99-880 with a relatively high ChemPLP fitness score,was selected and expressed in Escherichia coli BL21(DE3).It was further characterized using a feeding experiment with *** confirmed that the RO3G-expressing strain could convert 1-dodecanol to lauryl glucoside,as quantified by HPLC and identified by targeted *** the growth and fermentation profiles of the engineered strain revealed that RO3G expression did not affect cell ***,acetate,a major fermentation product,was reduced in the RO3G-expressing strain compared to the GFP-expressing strain,suggesting a redirection of flux from acetate to other ***,this work presents a successful workflow for discovering UDP-glycosyltransferase enzymes with confirmed activity toward 1-dodecanol for lauryl glucoside production.

Exploring various phenomena and issues related to leaf images is paramount, particularly in segmentation and classification of such images. This study employs bibliometric analysis to delve into two overarching themes: The trends in publication and the evolution of publications, along with a keyword-based analysis. The research methodology unfolds in two stages: (1) data collection and (2) analysis. The dataset comprises 1,248 articles sourced from the Scopus database, covering the period from 1988 to 2023. The research findings unveil a noteworthy surge in publication trends, peaking at 231 documents in 2023. An in-depth examination of journal names demonstrates that studies on the segmentation and classification of leaf images are predominantly featured in computer science journals, exemplified by the publication of 589 documents. Furthermore, an analysis of frequently used keywords highlights 'Extraction' as the predominant term, employed a total of 364 times. This underscores that the research focus on leaf image segmentation and classification presents ample opportunities for researchers to delve more profoundly into the subject. © 2024 IEEE.

Flexible job shop scheduling (FJSP) has garnered enormous attention within the realm of smart manufacturing, where, beyond job sequencing, the selection of machines holds considerable importance. As smart factories progress with the Internet of things (IoT) and cyber-physical systems (CPS), scheduling methodologies are advancing towards intelligent decentralization. However, with the expansion of factories, conventional cloud computing struggles to manage the substantial influx of data. To tackle this issue, this work incorporates a fog computing and edge computing framework into the distributed FJSP workstations. In this framework, the workstations each of which consists of multiple machines are categorized based on the different nature of the accommodated machines, and operate independently to reduce unnecessary information transmission, in which each machine is equipped with edge computing capacity. The fusion of fog computing and edge computing allows for the offloading of computational tasks from cloud computing, effectively reducing latency. While previous solutions for FJSP have predominantly relied on linear programming or metaheuristic algorithms, this work proposed a novel distributed approach based on a dual deep Q networks (dual DQN) architecture, integrating deep learning (DL) with reinforcement learning (RL). Within the cloud center, the initial neural network determines the machine selection rules for fog computing, while the secondary neural network decides the job dispatching rules for edge computing devices. Edge computing devices execute the schedule and provide feedback to the cloud, which refines the results through an iterative training process, so that to minimize the makespan. The experimental findings indicate that employing dual DQNs outperforms the methods of utilizing only one single machine selection rule. © The Author(s), under exclusive licence to Springer science+Business Media, LLC, part of Springer Nature 2024.

Coral reefs, vital marine ecosystems, are highly vulnerable to thermal stress-induced bleaching. Regional-scale bleaching events are often triggered by elevated sea surface temperature (SST) associated with global warming. While sustained temperature anomalies exceeding ± 1 °C above the long-term maximum monthly mean can induce bleaching, extreme short-term warming and cooling events, known as marine heatwaves (MHW) and marine cold spells (MCS), respectively, pose significant threats. These events, characterized by prolonged periods of anomalous temperature extremes, can have far-reaching ecological and economic consequences. This study investigated the spatial distribution of MHW and MCS in the Lombok Sea and their relationship with coral bleaching. Using daily SST data from 1998 to 2022, we identified MHW and MCS events and analyzed their frequency, intensity, and duration. Additionally, Landsat imagery (Landsat 5, 6, and 7) and field surveys conducted in 2016 were employed to assess changes in coral cover over time. Our results show that the effect of MCS and MHW cause coral bleaching, particularly in years with high frequency, intensity, and duration of these events. Extreme daily intensity (anomaly of SST > 1.5 °C) of MCS in 2000 and MHW in 1998, 2016, and 2022 led to a significant decrease in coral reef cover of approximately 30%. Furthermore, we observed that large-scale climate patterns such as the El Niño-Southern Oscillation (ENSO) can exacerbate MHW (in 1998 and 2016) conditions and contribute to coral decline. These results underscore the urgent need for effective coral reef management strategies to mitigate climate change and ocean warming impacts. © The Author(s), under exclusive licence to Sociedade Brasileira de Engenharia Naval 2025.

Lithium is the cornerstone of energy transition for Indonesia to achieve Net Zero Emissions by 2060 or earlier. This energy transition will drive the Indonesia demand projection for Electric Vehicle Battery and Energy Storage System and in turn will demand much more lithium for this transition effort. Many studies have been conducted to find the sources of lithium, one of them is geothermal brine. Indonesia has enormous geothermal resources, some fields have lithium content that can potentially be extracted. Various methods in the extraction process of lithium from the geothermal brine have been developed, both on laboratory and pilot projects. The geothermal brine is typically pumped to the surface by a geothermal power plant, used for energy generation, and then returned to the underground reservoir via an injection well. Prior to reinjection the geothermal brines contain in the temporary ponds whereas the lithium extraction points may occur here or at any point suitable for the extraction technology. Typically there are three phases in the lithium extraction, like end- to-end process technology solutions, namely 1) Lithium Brine Extraction-depending on the geothermal brines field compositions, pre-treatment may be employed to remove any contaminants or unwanted constituents from the brines, then selective absorbents are used to separate lithium from other elements 2) Concentration with membrane system to concentrate lithium at a desired concentration 3) Deposition using chemical precipitation and purification for saleable lithium production. This paper reviews traditional brine extraction and direct lithium extraction (DLE) in general and specifically in geothermal brine. The study is based on a desktop study and aims to summarize the knowledge, method, technology, and techniques of lithium extraction from geothermal brine that has already been used and to find out which extraction method is suitable for the Indonesian geothermal field. This paper presents an advanced method for addressing the inverse kinematics and optimal path planning challenges in robot manipulators. The inverse kinematics problem involves determining the joint angles for a given position and orientation of the end-effector. Furthermore, the path planning problem seeks a trajectory between two points. Traditional approaches in computer algebra have utilized Gröbner basis computations to solve these problems, offering a global solution but at a high computational cost. To overcome the issue, the present authors have proposed a novel approach that employs the Comprehensive Gröbner System (CGS) and CGS-based quantifier elimination (CGS-QE) methods to efficiently solve the inverse kinematics problem and certify the existence of solutions for trajectory planning. This paper extends these methods by incorporating smooth curves via cubic spline interpolation for path planning and optimizing joint configurations using shortest path algorithms to minimize the sum of joint configurations along a trajectory. This approach significantly enhances the manipulator's ability to navigate complex paths and optimize movement sequences. © 2024 CEUR-WS. All rights reserved.

We propose a method for solving the speech direction estimation problem by computer algebra. The method is based on the function approximation using the minimax polynomial. The minimax polynomial is obtained by an iterative method called the Remez exchange algorithm, in which Gröbner bases computation is employed. This paper discusses the trajectory planning of a robot manipulator with computer algebra. In the operation of robot manipulators, it is important to make the trajectory of the end effector so that it does not collide with obstacles. For this purpose, we have proposed a method of generating a method of trajectory planning using cubic spline curves. However, the method has the disadvantage that the trajectory may not be included in the feasible region of the manipulator, thus an extra test for the inclusion of the curve is needed. In this paper, we propose a new method of generating a trajectory using Bézier curves, which is guaranteed to be included in the feasible region. © 2024 CEUR-WS. All rights reserved.

Occurrence and risk assessment of glycidyl and 3-monochloropropanediol esters in infant formulas marketed in Taiwan

Glycidyl esters (GEs) and 3-monochloropropanediol esters (3-MCPDEs) are process contaminants commonly found in refined edible oils which are often added to infant formulas. The Taiwan Food and Drug Administration (TFDA) launched regulations for GEs in infant formulas that went into effect on 1 July 2021. To investigate levels of GEs and 3-MCPDEs in infant formula powder, 45 products were sampled and analysed during 2020–2021. The contents of GEs and 3-MCPDEs in formulas of different brands significantly varied, but their concentrations in all of the formulas complied with European Union (EU) regulations. Infant formulas containing palm oil had significantly higher 3-MCPDE levels in both extracted oils and milk powder than those without palm oil. Concentrations of GEs and 3-MCPDEs in infant formula powder and extracted oils were significantly lower in products from Europe than those from Australia and New Zealand. Infants aged 0–1 years in Taiwan who consumed only infant formula showed a margin of exposure (MoE) exceeding 25,000. Mean consumer exposures to 3-MCPDEs stayed below the tolerable daily intake (TDI), while high exposures at the 95th percentile (P95) exceeded the TDI by 1.7-fold. Herein, we present the changing trends in the risk assessment results of infant formula across various countries in the decade. Implementation of regulations and mitigation strategy effectively reduced the risk of infants being exposed to GEs and 3-MCPDEs through infant formula. © 2024 Taipei Medical University.
