Alginate is a polysaccharide extracted from cell walls and intracellular matrix of brown seaweed, and has been widely used for industrial purpose due to gelling capabilities in the presence of divalent cations. However, the characteristics of alginate extracted are depend on alginate gel strength and viscosity. The objective of this research was to investigate the effect of extraction time and Na2CO3 concentration on the characteristics of alginates extracted from Sargassum sp. by acid method. Alginate extractions were conducted by varying the extraction time for 1.5, 2.0 and 2.5 hours;and varying the Na2CO3 concentration of 1, 2 and 3 %. The characteristics observed are alginate yield, viscosity, pH, and water content. The alginates extracted using various Na2CO3 concentration and extraction time resulted alginates with low, medium and high viscosity. Furthermore, the yield, pH and water content of alginates extracted in this research had fulfilled the standard of Food Chemical Codex. © 2018 Published under licence by IOP Publishing Ltd.

The potential of Brachionus sp. for Koi fish (Cyprinus carpio) cultivation infected by Myxobolus sp.

The first step to success in Koi fish cultivation is hatchery activity. However, in the process of cultivation often suffer difficulty due to the parasitic outbreak. The type of parasites that often attacks Koi fish is Myxobolus sp. Therefore, the natural feed is needed such as Brachionus sp. as a protein resource for the survival of Koi fish that infected by Myxobolus sp. Purpose of this research was to find out the potential of Brachionus sp. as a natural feed for Koi fish cultivation that infected by Myxobolus sp. The research method used the descriptive method. The sample was taken at two pond sites that infected by Myxobolus sp. and clean pond by filtering pond water using a plankton net and preserved in a bottle then conducted the identification and calculation of the abundance of Brachionus sp. under the microscope. The results show that observation on each pond found Zooplankton with species of Brachionus sp. and obtained a number of Brachionus sp. abundance on site 1 (infected by Myxobolus sp.) got 57ind/ml. while site 2 (clean pond) got 22 ind/l. The hemagglutinin test results showed fish infected with Myxobolus sp. react positively.

We addressed the underrepresentation of non-European populations in genome-wide association studies (GWASs) by building HiGenome, a large-scale genetic resource for the Taiwanese Han population. Using a custom genotyping array, we integrated deidentified electronic medical records (2003 to 2021) with genomic data to enable GWASs, phenome-wide association studies, and polygenic risk score (PRS) analysis. Among 413,000 participants, 323,397 passed ancestry and quality control filtering. GWASs covered 1085 traits, focusing on diseases prevalent in Taiwan such as type 2 diabetes, chronic kidney disease, gout, and alcoholic liver damage. PRSs were calculated for 238 traits, with the strongest associations observed in musculoskeletal disorders. Incorporating PRS into clinical practice supports early risk prediction and personalized prevention. To further expand translational value, we also conducted pharmacogenomic analysis and human leukocyte antigen typing. HiGenome offers a large-scale genetic and clinical dataset from the Taiwanese Han population, supporting population-specific analyses and precision medicine development in East Asia. The hospital-based design enables continuous follow-up and longitudinal data expansion.

Objective: Angiotensin-converting enzyme(ACE) insertion/deletion(I/D) polymorphisms are considered biomarkers of late-onset Alzheimer's disease(AD). However, the results of studies of these polymorphisms have been inconsistent. The ACE protein directly degrades beta-amyloid and thereby slows the progression of AD and its onset. However, it also activates the renin–angiotensin–aldosterone system, which can contribute to hypertension and/or cardiovascular events that increase the risk for developing AD. Methods: In this study, we examined the bidirectional association of the ACE gene and AD in patients with AD with and without hypertension. Results: Patients who were clinically diagnosed with AD(n = 983) underwent ACE I/D genotyping. The distribution of the ACE I/D genotypes(P = 0.355 for I/I vs. D/D; P = 0.888 for I/D vs. D/D) and I alleles(P = 0.895) did not significantly differ between hypertensive patients with AD and patients with AD without hypertension. Conclusions: In contrast to traditional theories, the results of this study suggested that the contribution of the ACE gene to the development of AD was not associated with hypertension and similar cardiovascular effects.

In this paper, we explain how an experiential learning course and study tour to Gansu Province (People’s Republic of China) enabled undergraduates at the Singapore Management University (SMU) to acquire 21st-century competencies and higher-order thinking skills by analyzing and evaluating specific aspects of China’s Belt and Road Initiative (BRI) and China–Singapore (Chongqing) Connectivity Initiative — New International Land–Sea Trade Corridor (CCI-ILSTC) with emphasis on developing viable Go-To-Market (GTM) strategies aimed at selling Gansu produce in four Southeast Asian markets. We share how the course was designed to support the attainment of key learning goals and discuss how we turned pedagogical aspirations into concrete learning outcomes. We introduce key aspects of the so-called “SMU-XO” project that the students conducted in partnership with an industry partner, Pacific International Lines (PIL), and discuss how the project work helped learners to gain global competency by (i) examining critical issues related to BRI such as multi-modal infrastructure connectivity, (ii) appreciating the local perspectives of project stakeholders in Lanzhou and Shanghai and (iii) successfully interacting with people from different cultures, namely China, Indonesia, Vietnam, Malaysia and Thailand.

The ACE （angiotensin converting enzyme） inhibitors are not only drugs widely prescribed drugs in cardiovascular diseases, but also potentially therapeutic agents in dementia. Based on the findings that the ACE inhibitors could activate the c-Jun N-terminal kinase signal to increase the ACE gene expression and that the Alu element of the human ACE gene involved in regulating ACE promoter activity, we aimed to investigate whether there are different pharmacogenetic responses of ACE I/D polymorphism to the ACE inhibitors in neurons. The three reporter vectors, pACEpro（0-SEAP, p-I-ACEpro-SEAP, and p-D-ACEpro-SEAP were used to examine the transcriptional activity of the vectors responding to the lisinopril treatment using a transient-transfection method in SH-SY5Y cells. Our results showed that lisinopril increased the promoter activity of an ACE gene by 16.7%. Additionally, we found the lisinopril enhanced the ACE promoter activity of the I-form vector by 17.2%, but adversely reduced that of the D-form vector by 16.8%, as compared with the respective control without the lisinopril treatment. Firstly, our findings had proved that the UD polymorphism of ACE gene contrarily responds to the ACE inhibitors in regulating the ACE expression in neurons, which provide a novel insight suggesting genetic testing to tailor the treatment regimens in AD （Alzheimer＇s disease） patients.

IgA nephropathy (IgAN) is the most common cause of primary glomerular *** responses have been shown to correlate with the progression of ***3 inflammasome, a multiprotein complex, positively regulates caspase-1 activity and the maturation and release of IL-1β.NLRP3 inflammasome plays an important role in inflammatory response and controls the pathogenesis of various inflammatory diseases including pathogen infection, diabetes, atherosclerosis, Alzheimer's disease, and ***, the potential pathogenic role of NLRP3 inflammasome on IgAN is *** this study we demonstrated that IgA immune complexes induce caspase-1 activation and IL-1β secretion in mice J774A.1 macrophages and these effects were reduced significantly by knockdown of *** of total intracellular reactive oxygen species (ROS) and mitochondrial ROS were increased by IgA immune complexes treatment, while antioxidant N-acetyl-cysteine and mitochondrial ROS scavenger Mito-TEMPO reduced IL-1β secretion in IgA immune complexes stimulated J774A.1 *** results indicated that IgA immune complexes activate NLRP3 inflammasome;therefore, we hypothesized that blocking of NLRP3 inflammasome might prevent the development of *** demonstrated that blockade of renal NLRP3 expression by ultrasound-mediated microbubble shRNA gene transfer significantly (1) reduced the IL-1β expression and caspase-1 activation in the kidney, (2) prevented renal injury, and (3) attenuated proteinuria and improved renal ***, these effects were associated with greatly inhibited infiltration of T cells/macrophages and suppressed NF-B activation in the *** results indicate that activation NLRP3 inflammasome is involved in the development of IgAN, and a kidney-specific knockdown of NLRP3 may have therapeutic potential for IgAN.

Amyotrophic lateral sclerosis (ALS) is a complicated and devastating neurodegenerative disease. To date, its diagnosis is still mainly based on clinical symptoms and electromyographic findings. High rates of misdiagnosis and delayed diagnosis are the major hurdles in ALS treatment. Thus, searching for biomarkers to improve clinical diagnosis of ALS is a highly desirable goal. Here we review current potential biomarkers derived from the various pathogenic mechanisms of ALS, including those involved in oxidative stress, synaptic excitotoxicity, neuroinflammation and the autoimmune response. Oxidative stress results from genetic mutation or an increase in protein aggregation, synaptic excitotoxicity arising from elevated levels of glutamate and D-serine, and the neuroinflammation occurring from elevated levels of inflammatory molecules and cytokine receptors. Some of these biomarkers could be used for monitoring the disease progression and to assess effectiveness of treatment for ALS. We conclude that neuroinflammation plays a crucial role in ALS, which may lead to a better understanding of this devastating disease and ultimately to a cure. In addition, the identification of new biomarkers would undoubtedly provide critical insights into the pathogenesis of ALS. © 2013 Bentham science Publishers.
