Calibrated Langevin-dynamics simulations of intrinsically disordered proteins

We perform extensive coarse-grained (CG) Langevin dynamics simulations of intrinsically disordered proteins (IDPs), which possess fluctuating conformational statistics between that for excluded volume random walks and collapsed globules. Our CG model includes repulsive steric, attractive hydrophobic, and electrostatic interactions between residues and is calibrated to a large collection of single-molecule fluorescence resonance energy transfer data on the interresidue separations for 36 pairs of residues in five IDPs: α−, β−, and γ-synuclein, the microtubule-associated protein τ, and prothymosin α. We find that our CG model is able to recapitulate the average interresidue separations regardless of the choice of the hydrophobicity scale, which shows that our calibrated model can robustly capture the conformational dynamics of IDPs. We then employ our model to study the scaling of the radius of gyration with chemical distance in 11 known IDPs. We identify a strong correlation between the distance to the dividing line between folded proteins and IDPs in the mean charge and hydrophobicity space and the scaling exponent of the radius of gyration with chemical distance along the protein.

This study uses a three dimensional model of the knee joint to determine which knee joint motion schemes lead to ACL injury, while exploring the possible types of concomitant injuries associated with each motion scheme. The physical elements of the knee joint including bones, articular cartilage, meniscus and ligaments were obtained by reconstructing magnetic resonance images. A failure locus representing the relationship between motion schemes and knee joint injury was created from finite element simulations considering knee flexion, femoral axial rotation and both valgus and varus motion. The relationships between knee joint orientation and various tissue failures were examined and susceptibility spectrums for knee injuries were obtained. The posterolateral bundle demonstrated higher rupture susceptibility than the anteromedial bundle. The average varus angular displacement for ACL failure was 46.6\% lower than the average valgus angular displacement. Femoral external rotation decreases overall ACL valgus/varus failure angle by 27.5\% compared to internal rotation. Articular cartilage injury was shown to occur prior to ACL failure in all valgus simulations. A simplified and computationally efficient version of the full 3D model showed close correlation to the complete model with respect to ligament failure and shows that if only ligament failure is of interest, a simple model can be used. The results of this study highlight several detrimental joint motions and can aid in improved clinical diagnoses and improved training programs for athletes.

The field of transcranial electrical stimulation (tES) has experienced significant growth in the past 15 years. One of the tES techniques leading this increased interest is transcranial direct current stimulation (tDCS). Significant research efforts have been devoted to determining the clinical potential of tDCS in humans. Despite the promising results obtained with tDCS in basic and clinical neuroscience, further progress has been impeded by a lack of clarity on international regulatory pathways. Therefore, a group of research and clinician experts on tDCS were convened to review the research and clinical use of tDCS. This report reviews the regulatory status of tDCS and summarizes the results according to research, off-label, and compassionate use of tDCS in the following countries: Australia, Brazil, France, Germany, India, Iran, Italy, Portugal, South Korea, Taiwan, and the US. Research use, off label treatment, and compassionate use of tDCS are employed in most of the countries reviewed in this study. It is critical that a global or local effort is organized to pursue definite evidence to either approve and regulate or restrict the use of tDCS in clinical practice on the basis of adequate randomized controlled treatment trials. © 2014 Informa Healthcare USA, Inc. In this study, slurry formulations in the presence of self-assembled surfactant structures were investigated for Ge/SiO2 CMP applications in the absence and presence of oxidizers. Both anionic (sodium dodecyl sulfate-SDS) and cationic (cetyl trimethyl ammonium bromide-C12TAB) micelles were used in the slurry formulations as a function of pH and oxidizer concentration. CMP performances of Ge and SiO2 wafers were evaluated in terms of material removal rates, selectivity and surface quality. The material removal rate responses were also assessed through AFM wear rate tests to obtain a faster response for preliminary analyses. The surfactant adsorption characteristics were studied through surface wettability responses of the Ge and SiO2 wafers through contact angle measurements. It was observed that the self-assembled surfactant structures can help obtain selectivity on the silica/germanium system at low concentrations of the oxidizer in the slurry. © 2013 Materials Research Society.

DC faults may cause severe disruptions in continuity of service to vital loads in a shipboard integrated power system, hence detection, isolation, and protection against such faults must be incorporated in both medium-voltage DC (MVDC) and low-voltage DC (LVDC) systems. Here we consider the effectiveness of existing z-source breakers and propose several new designs more appropriate for fault detection in MVDC and LVDC systems. In particular, we perform an optimization study that aims to minimize dissipation and weight and we identify the key parameters for use in MVDC and LVDC systems. Preliminary verification and validation studies are also included. © 2013 IEEE.

Patterning ultrathin MoS2 layers with regular edges or controllable shapes is appealing since the properties of MoS2 sheets are sensitive to the edge structures. In this work, we have introduced a simple, effective and well-controlled technique to etch layered MoS2 sheets with well-oriented equilateral triangular pits by simply heating the samples in air. The anisotropic oxidative etching is greatly affected by the surrounding temperature and the number of MoS2 layers, whereby the pit sizes increase with the increase of surrounding temperature and the number of MoS2 layers. First-principles computations have been performed to explain the formation mechanism of the triangular pits. This technique offers an alternative avenue to engineering the structure of MoS2 sheets.
