The available prophylactic vaccines for human papillomavirus (HPV) in the market are only effective against specific types of HPV, rendering them ineffective for other types of HPV infections. The objective of this research is to investigate the stability of the recombinant protein constructed, namely chimeric L1/L2 protein from HPV type 52, with improved cross-neutralization ability. The 3D model, predicted using Alphafold, Robetta, I-Tasser, and refined with Galaxy Refinement, is validated using Ramachandran plot analysis. The stability is verified through molecular dynamics simulations, considering parameters such as RMSD, RMSF, Rg, and SASA, where stable conditions are observed. The chimeric L1/L2 protein from HPV type 52 is purified using affinity chromatography, and the His-tag is cleaved using SUMO protease to obtain pure chimeric protein with the size of ~ 55 kDa. Western blot analysis confirms binding to anti-L1 HPV type 52 polyclonal antibody. The obtained vaccine candidate can be utilized as an effective prophylactic vaccine against HPV. © 2024 Taylor & Francis.

Bacterial infections and multidrug-resistant bacteria are major health burdens in wound *** antimicrobial agents,e.g.,ε-polylysine(ε-PL),provide a broad spectrum of antibacterial properties and support dermal cell ***,ε-PL was incorporated into polycaprolactone(PCL)/gelatin electrospun scaffolds collected at varying rotation ***,the samples were crosslinked using dopamine hydrochloride to provide highly proliferative dressings with broad antimicrobial *** morphological study showed that the electrospun wound dressings were smooth,continuous,and bead-free,with a mean diameter ranging from 267±7 to 331±8 nm for all random and aligned *** fiber alignment of the electrospun PCL/gelatin scaffolds improved their tensile strength and ***,nanofiber mats are highly hydro-philic,which is crucial for an efficient wound *** samples also demonstrated high antimicrobial properties against common wound bacterial strains,including methicillin-resistant Staphylococcus aureus(MRSA),Staphylococcus aureus(SA),Escherichia coli(EC),Acinetobacter baumannii(AB),and Pseudomonas aeruginosa(PA).Mammalian cell prolifera-tion and morphology assays involving primary human dermal fibroblasts(hDFs)and immortalized keratinocytes(HaCaT)showed excellent biocompatibility of the electrospun mats and remarkably aligned ***,aligned mats showed more cell migration than randomly oriented mats,which is desirable for more efficient wound ***,it can be concluded that aligned PCL/gelatin mats containingε-PL are promising for potential use in wound dressings.

Mining water is one of the main problems that has a great importance on mine productivity. In mining implementation, it is necessary to study the Mining Water (Hydrogeology) system to support a good mine design. The Mining Water drainage system is a method for making work sites in the mining area always dry. Prior to mining, hydrogeological analysis and rainwater management methods are carried out. Calculation of rainfall intensity using Gumbel Probability estimation and water capture area analysis obtained CA-1 Q = 915, 35, m3 / day CA-2 Q = 1149, CA-3 Q = 141116, 01CA-4 Q = 1407.89, the form of rainwater trapping channels in the form of trapezium and pump requirements in each catchment area of 1 (one) pump each. © 2020 Published under licence by IOP Publishing Ltd.

A mid-level labor preparation having the qualification and competency appropriate with working world needs is a task from vocational secondary school (VSS). Now days, VSS become an attention by wide community, as a result of the existence of government policy which is trying to empowering vocational secondary education. The policy taken is by performing the revitalization of vocational school which has the aim to improve the quality of labor competitiveness. One of the programs is teacher quality improvement through various teacher training. However, relating to this case, it is suspected that the previous vocational school teacher training is less effective, so that it needs to be conducted the analysis about the teacher training implementation. Therefore, this article will be elaborated the ways needed to be conducted in improving the educational quality and vocational school teacher development training. The ways should be considered when it is planned and implemented the effective training program, such as: determining the needs, setting the goal, determining the content of learning, selecting the participants, determining the best schedule, selecting the appropriate facilities, selecting the proper instructor, selecting and preparing the audiovisual equipment, coordinating the program and evaluating the program. © 2021 Published under licence by IOP Publishing Ltd.

This study aims to determine the influence of industrial work practice and workshop facilities on work readiness in class XI mechanical engineering. The research subjects were 101 students of class XI mechanical engineering, using the ex post facto research method using statistical analysis using the SPSS program. The results of the study are as follows: (1) Industrial work practices have a positive effect on job readiness with a significance of 0.000, a determination coefficient of 30% has an effect;(2) Workshop infrastructure has a positive influence on work readiness with a significance value of 0.000, 28.6% determination efficiency has an effect;(3) Industrial work practices and infrastructure together have an influence on job readiness indicated by a significance value of 0.000 having a coefficient of determination 58,6% in influencing job readiness. © Published under licence by IOP Publishing Ltd.

Database lectures in the informatics engineering study program teach the life cycle of database system development, both in terms of concepts, utilization, administration to database application systems to be studied, practiced and understood by students. Database teaching in the informatics engineering department of a private university with very diverse student participants, the ability to master computer and information technology and skills requires teaching material, lecturing strategies, lecturer quality, and facilities must also be able to adjust toward challenges, concept development, and database technology. This database design research model used Hevner's information systems research framework, starting with reference research, database lecture analysis, rigor and research relevance. This research covered the steps of constructing artifacts in the form of constructs, models, methods, instantiation, and evaluation of artifacts. The use of the framework intended to support business strategies and processes in study programs and ensure the relevance of research. The benefits of this research will produce a database lecture model that can be used as a reference and lecture development in the informatics engineering study program. Reference to produce graduates with understanding and skills in database design, the use of Database Management System (DBMS), the ability in administration and professional capability in the database field. It is very important to reintroduce waste to the economy in the most efficient way and to reduce their amount both from the point of view of economic and environmental problems. However, when waste is evaluated, not only its elimination but also its use in areas with high added value should be considered. This study is aimed to create a different usage area for the slag wastes that cause problems to the environment and factories. For this reason, two different coatings;the low-cost BFS and commercial 80Ni-20Cr coatings have been successfully deposited on 316L stainless steel substrates by atmospheric plasma spraying (APS) and high-velocity oxy-fuel (HVOF) spraying techniques, respectively. The coatings’ microstructural properties and wear performance were studied under dry sliding conditions in ball-on-flat reciprocating tests using an Al2O3 ball as the counterpart. Reciprocating wear tests were comparatively evaluated under various dry sliding conditions. Scanning electron microscopy (SEM) and energy dispersive spectroscopy was used to examine the surface morphology and elemental analysis of coatings (EDS). The wear traces obtained from the tests were examined using a 3D optical profilometer. The phases in the samples were determined with the X-ray diffraction (XRD) technique. This study was carried out to determine the applicability of BFS waste material in material coating technologies, to investigate its usability in industrial applications, and to determine its performance for commercially used coatings against wear-oriented damage mechanisms. As a result of the study results, it has been seen that BFS material can be produced by the thermal spray coating method in material coating technologies and has similar properties to Ni-Cr coatings in commercial use in terms of wear performance. © 2023, The Authors. The main road that connects the city of Sidoarjo to Surabaya is the intersection of Gedangan. By 2018 it has reached saturation by having a poor service level. More severe conditions will occur in the next 10-20 years. One of the government's efforts to overcome the problem of congestion, damage structure is the construction of flyover or other alternatives by widening the road. The purpose of this study is to determine the characteristics of the traffic and to save vehicle operating costs before and after the construction of a flyover at the Gedangan intersection. The method for predicting Vehicle Operating Costs (BOK) is to use PCI method. This study predicts Vehicle Operating Costs (BOK) to determine cost savings. The scenario for predicting BOK is that 80% of the vehicle volume will pass the Fly Over, and 20% still use the existing road. The results showed that the cost savings of Vehicle Operational Costs (BOK) of existing roads compared to the use of flyover in 2019 was IDR 15 Billion. In 2039 the savings are IDR 77 Billion. Another discussion is that if the road is only widened without flyover, then the BOK cost savings in 2019 will be IDR 4 Billion and in 2039 it will be IDR 27 Billion. © 2020 Published under licence by IOP Publishing Ltd.

Generating hydrogen from renewable energy sources is widely recognized as an effective strategy for addressing critical challenges in the energy sector while contributing to the reduction of greenhouse gas emissions. This study aims to assess the wind energy potential for electricity and hydrogen production in five locations on Sumba Island using 10 years (2011–2020) of data from the NASA's POWER Project. A wind variability assessment was performed on five wind turbines ranging from 500 kW to 1.5 MW and using electrolysis technology. The results indicated a hydrogen production potential of 25–57 tons per year. Economic analysis of hydrogen production for Haharu reveals an attractive internal rate of return (IRR) of 43% with a payback period (PBP) of 2.25 years and a net present value (NPV) of 1.3 MM USD, the levelized cost of electricity (LCOE) of 0.0241 USD/kWh, and the levelized cost of Hydrogen (LCOH) of 1.01 USD/kg. Sensitivity analysis underscores the critical influence of investment and revenue on project profitability, highlighting their substantial impact on determining a project's success or failure. These findings emphasize the potential of Sumba Island as a hub for sustainable energy production to meet the growing demand for clean and renewable energy sources. © 2024 Hydrogen Energy Publications LLC

Organic-inorganic halide perovskites have become notable for their efficiency in light harvesting. Nonetheless, the incorporation of organic cations like $\mathbf{M A}^{+}$and $\mathrm{FA}^{+}$presents stability challenges when exposed to environmental factors. This study explores the potential of replacing organic components with inorganic $\mathrm{Cs}^{+}$to enhance stability and efficiency in PSCs, focusing on CsPbBr 3 as the light harvester. We compare spin-coating and spray-coating techniques for depositing SnO 2 as the electron transport layer, revealing that spray-coating 7 cycles achieves higher PCE. Through detailed characterization, including UV-Vis, FTIR, SEM, I-V, and IPCE analyses, we optimized the manufacture of $\mathrm{SnO}_{2}, \mathrm{CsPbBr}_{3}$, and carbon deposition at room temperature, resulting in a PCE of $\mathbf{2. 2 1 \%}$ using the spray-coating process, demonstrating significant improvements over the spin-coating method.
