Indonesia is located along the archipelago which has abundance of volcanoes along the arc. Therefore, Indonesia has a very high geothermal energy potential, such as the Rendingan-Ulubelu-Waypanas Geothermal Field in Tanggamus Regency, Lampung Province. Geophysical exploration is required to characterize the subsurface geothermal system. One of the methods that can be used is gravity method. The data used in this study is an analog Rendingan-Ulubelu-Waypanas Geothermal Field Complete Bouguer Anomaly Map of 2005. The analog map was then converted into digital map for further data processing. Separation of regional and residual Bouguer anomalies has been performed using moving average and spectral filter methods, in which the spectral analysis was conducted in 1D and 2D. The value of Bouguer background density used for 2.5D subsurface modeling is 2.67 g/cc. The gravity anomalies obtained in this study can be interpreted as follow. The existence of high anomalies under Mount Waypanas was interpreted as a Tertiary intrusion of granite with a density contrast of 0.22 g/cc. While low anomalies in the northern part was interpreted as a Quaternary intrusion of diorite with a density contrast of -0.22 g/cc, which acts as the heat source of the system. The reservoir of the system is the Quaternary young volcanics exposed on the surface with averaged thickness of 1500 m. We concluded that the fluid flow in this geothermal system is the meteoric water from Mount Rendingan and Mount Tanggamus, which has a higher topography, flows to a lower topography area in the Mount Waypanas, The fluid flow also creates an upflow under Mount Rendingan. © 2019 IOP Publishing Ltd. The Southern Coast of West Java Province in the Java Islands is one of prone and exposed area to tsunami hazard. It has about 428 kilometers coastline length, covering 5 administrational regencies (i.e. Sukabumi, Cianjur, Garut, Tasikmalaya, and Pangandaran). It is a strategic area that support many economic activities including tourism, fisheries, electricity power plants, agricultures, markets as well as social factors such as coastal villages, schools and other public facilities. The existence of this strategic area is threatened by the potential for a tsunami disaster mainly from megathrust along the Southern Coast of Java Island. On 2006, a tsunami earthquake of M7.7 occurred off Pangandaran Regency with tsunami height of 5-8 meters and inundated about 500 meters along southern coast of West and Central Java Province. The tsunami caused more than 600 casualties and damage to buildings, public facilities and insfrastructure. Yet, the Indonesia National Earthquake Source and Hazard Map suggest a bigger threat of a plausible M8.7 - M9.2 megathrust earthquake off Java island. This research aims to analyze and map potential areas affected by the tsunami in the South Coast of West Java Province based on numerical modelling carried out with Cornell Multi-grid Coupled Tsunami Model (COMCOT) v1.7 based on the several megathrust earthquake scenarios in southern off West Java. The results of tsunami modelling show that the highest impact is generated in the Sunda Straits & West Java - Central Java Megathrust Scenarios with highest tsunami inundation and the fastest travel time occurred in Cianjur Regency with 26,7 meters height and estimated arrival time of tsunami wave approximately 10 minutes after the mainshock, while the farthest inundation distance is estimated in Sukabumi Regency about 5.8 kilometers from the coast due to existence of river. © Published under licence by IOP Publishing Ltd.

The CRISPR/Cas9-mediated base editing design of HBV specific gRNA: A computational approach

Therapy to cure a persistent infection of the Hepatitis B Virus (HBV) is needed to suppress viral replication. Current therapy with nucleoside analogues does not destroy HBV covalently closed circular DNA (cccDNA). A new strategy using Base Editing is required to tackle this viral latent infection. We designed guide RNAs (gRNAs) to target the pre-S and S genes of HBV for the development of antiviral, based on the dominant HBV genotype in Indonesia. The conserved regions of HBV, pre-S and S genes were used to determine a selected target of the Cas9-mediated base editing system. Base Editing (BE)-Designer web-based software to designed gRNA and SnapGene to visualized the expression vector. We identified 5 candidate sequences, including 3 sequences in the pre-S and 2 sequences in the S gene. These target sequences show no mismatches and GC contains between 45-60%. In addition, the gRNA sequences were checked in Nucleotide BLAST tools (NCBI) to prevent the off-target effect and ensure the differences with the human genome. This study provides the candidate target sequences to be developed in antiviral therapy. The utility and efficiency of the candidate sequences need to be evaluated in viral gene expression by in vitro HBV infection systems.

The Sumatran fault zone has a high seismicity and seismic hazard threat levels due to this fracture through densely populated areas. The fault zone of Sumatera has 19 main segments which each segment has the possibility to generate a strong earthquake. Therefore, in this study we attempt to improve the accuracy of hypocenter locations which is needed as basic analysis in advance seismology and in seismic hazard analysis. The final objective of this study is to analyze the subsurface seismic velocity structure of the Sumatran fault by applying seismic tomography method. For the preliminary result, we have been conducting double-difference method to relocate 967 events recorded by 49 stations compiled by Meteorological, Climatological, and Geophysical Agency of Indonesia (MCGA) from 2009 to 2017 along Sumatran fault. Our preliminary results from the relocation proces indicate prominent features of event cluster around Sumatra fault system. The next step, we will improve the onset of P-and S-wave arrival time by using waveform cross-correlation in order to get more reliable hypocenter location. © 2019 IOP Publishing Ltd. The Complete Bouguer Anomaly (CBA) map from 1997 gravity survey at Karaha-Telaga Bodas, West Java, Indonesia is used to make a model of the subsurface geothermal system. A two-dimensional cross-section of the system is produced using a 2.5-D forward modeling. Initially, the print CBA maps of the Karaha-Telaga Bodas geothermal system is digitized. Next, the digital map is separated by using the moving average and spectral filtering methods, so a regional and residual anomaly maps are obtained. The subsurface modelling is made based on geological information of the area to produce a model that can represents the real condition as similar as possible. The line chosen for modelling has a North - South orientation, passing through Karaha and Telaga Bodas to foresee both of the geothermal system in one cross-section. Subsurface model interpretation shows that there are two separate granodiorite intrusion body with density contrast of 0.5 g/cc below Karaha - Telaga Bodas. This conclusion coincides with the intrusion body found during drilling operation. The intrusion body is possibly the heat source of the system. The reservoir of this system is interpreted as volcanic breccia with thickness of two kilometres in Telaga Bodas and keeps thinning towards Karaha. The fluids of the Karaha-Telaga Bodas Geothermal System are estimated to come from acid sulphate water and meteoric water entering from Telaga Bodas, as well as meteoric water entering from Karaha. A future networking design called "software-defined networking" combines network programmability with centralized administration (SDN). Network administration is currently handled by SDN, which, at the regulator, which acts as the cable network processor and divides the data plane into a control plane and a data aircraft. Recent breakthroughs in artificial intelligence (Al) have shown a stronger inclination for the science community to benefit from their ability to give learning capabilities and enhance Indicator. The author of this research provides a comprehensive analysis of initiatives underway to incorporate Al with SDN. The study concluded that the three primary Al thread where scientific research was centered were computer science, conceptual, and fuzzy reasoning systems. The authors of this paper explore the several fields in which these approaches may be used, their potential future applications, and the innovations made possible by the integration of AI-based methods into the SDN paradigm. By choosing the right SDN controller, any large organization may lower network complexity, implementation expenses, and maintenance costs. The focus of this article is software defined networking's benefits and downsides (SDN). Following is a basic explanation of artificial intelligence and a list of some of its most significant applications in SDN.

Tempe is a traditional food that is popular in Indonesia. Tempe consists of soaking, boiling, grinding, transferring, zymosis, packaging, and finally, the storage process. The packaging process is the process that causes the most musculoskeletal disorders. Complaints about the packaging process occur because of the wrong sitting position, repeated within 6-7 hours per day. Identification using Nordic Body Maps scored 71. This condition indicates a high level of risk for musculoskeletal disorders. The packaging process includes 88.89% complaints of pain in the buttocks, 77.78% in the hips, 66.78% in the waist, 55.56% on the back, and 44,44% on the right arm, 33.33% on the right wrist. This research develops a packaging process tool using the Kansei engineering method. The tool’s design is made according to the consumer’s wishes based on "Kansei words," and the manufacture of dimensions is based on anthropometry. Based on the validity test using a significance level of 5% obtained a marginal homogeneity value > 0.05 which indicates the design follows the wishes of consumers. Meanwhile, for the convenience attribute test using the Wilcoxon Signed Rank Test, the *** (2 tailed) value was obtained at 0.040<0.05, which indicates a significant difference between the packaging process before and after using the tool. The user satisfaction test using the User Experience Questionnaire (UEQ) shows an average value of > 0.8. A score of 0.8 on the UEQ indicates that the user is satisfied with the design.

In the present study, the polyvinylidene fluoride (PVDF) was incorporated with polyvinylpyrrolidone (PVP) and titanium dioxide (TiO₂) nanoparticles to prepare PVDF-TiO₂-PVP composite membrane for Bovine Serum Albumin (BSA) rejection from protein contaminated wastewater. The preparation of PVDF-TiO₂-PVP composite membrane was conducted by a non-solvent phase inversion (NIPS) process with varying PVDF weight percentages ranging from 16 to 19 wt%. The polymer concentration plays a critical role in the formation of composite membranes that is commonly overlooked. The primary goal of this study is to examine how the concentration of polymers affects the process of membrane creation, the ensuing structure, and the properties of these membranes. Presently, there is a restricted comprehension concerning the impact of PVDF polymer concentration in the fabrication of membranes using the NIPS approach, particularly for PVDF-TiO₂-PVP composite membranes. FE-SEM analysis revealed the lowest skin layer thickness and the number of macro-void formations in a PVDF composite membrane were at 16 wt% PVDF, compared to composite membranes over higher PVDF concentrations from 17 to 19 wt%. There were only slight variations in the percentages of individual elements across all PVDF-TiO₂-PVP membranes, yet the values were identical for C, O, F, and Ti. The viscosity was lowest at 16 wt percent PVDF (576.5 MPa s) and greatest at 19 wt percent PVDF (2054.3 MPa s). It showed that 19%wt PVDF viscosity increased fourfold higher than 16%wt. As the concentration of PVDF was increased, a related decrease in the surface porosity of the membrane was observed. The highest hydrophobicity with the contact angle of 79.62° of the composite membranes were detected at 19 wt% of PVDF loading. The maximum water flux (265.43 L/m²h) and BSA flux (250.31 L/m²h) were obtained at 19 wt% of PVDF loading. The lowest PVDF concentration of 16

The successful implementation of smart grids depends on energy efficiency, mostly relying on the operation of electricity source infrastructure. The combination of energy sources undoubtedly provides complexity in real applications. Some of the challenges for a smart grids comes from the intermittent renewable energy characteristics such as bioenergy, geothermal, hydropower, solar, ocean power, and wind energy to the base load energy such as natural gas, coal, and nuclear. This paper aims to review the implementation of current advances of smart grids from combined energy mix sources. The review includes major components of smart grids, i.e. power plants, monitoring systems, substations, power transmission and distribution, controllers and smart meters. The review also investigates the state of the art contribution of power electronics, artificial intelligence and Internet of Things to more significant optimization, safety and efficiency in these areas. The results are expected to provide a further gap analysis of applicable technologies and best practices in mixed electricity generation sources. The final result will also consider its application in an archipelago country such as Indonesia.

The Unified Power Quality Conditioner (UPQC) is a combination of a series active filter (SeAF) and a shunt active filter (ShAF) connected in parallel by a DC link capacitor. This device can mitigate power quality (PQ) problems i.e. sag/swell, harmonics, and unbalance on the source and load bus of threephase three-wire (3P3W) on low voltage distribution systems simultaneously. The disadvantage of UPQC is that it is unable to overcome the voltage interruption so that the source can not deliver power to the load. This paper proposes a dual UPQC model to overcome the voltage interruption on the source bus so that the load bus continues to get power supply. There are six disturbance cases i.e. sinusoidal supply-sag-non-linear load (SSag-NL-L), sinusoidal supply-swell-NL-L (S-Swell-NL-L), sinusoidal-interruption-NL-L (S-Inter-NL-L), distorted supplysag-NL-L load (D-Sag-NL-L), distorted supply-swell-NL-L (DSwell-NL-L), and distorted supply-interruption-NL-LL (DInter-NL-L). The proportional Integral (PI) method is used to control the SeAF and the ShAF in the dual UPQC circuit model. The simulation results show that in the D-Inter-NL-LL case, a Dual UPQC model can maintain a load voltage magnitude of 266.60 V (voltage drop only of 14%), higher compared to a Single UPQC model of 173.97 V (voltage drop of 43.88%). In the same case, a dual UPQC model is capable of resulting in an average total harmonics distortion (THD) of load voltage of 10.10%, lower compared to a single UPQC model of 26.70%.
