Background: Racial and ethnic minorities have borne a disproportionate burden from COVID-19. Certain essential occupations, including food processing and farm work, employ large numbers of Hispanic migrant workers and have been shown to carry an especially high risk of ***: We conducted an observational cohort study measuring the seroprevalence of SARS-CoV-2 and assessing risk factors for seropositivity among North Carolina (NC) food processing and farm workers and members of their households. Participants completed questionnaires and we collected blood samples and used an enzyme-linked immunosorbent assay to assess SARS-CoV-2 seropositivity. Univariate and multivariate analyses were carried out to identify risk factors associated with seropositivity, using generalized estimating equations to account for household ***: Among the 218 participants, 94.5% were Hispanic, and SARS-CoV-2 seropositivity was 50.0%. Most seropositive individuals did not report a history of illness compatible with COVID-19. Attending church, having a prior history of COVID-19, having a seropositive household member, and speaking Spanish as one’s primary language were associated with SARS-CoV-2 seropositivity, while preventive behaviors were ***: These findings underscore the substantial burden of COVID-19 among a population of mostly Hispanic essential workers and their households in rural NC. Our study contributes to a large body of evidence showing that Hispanic Americans have suffered a disproportionate COVID-19 burden. Introduction: The risk of developing Multiple Myeloma (MM) is 2-4 fold higher in first-degree relatives of patients with MM compared to the general population, suggesting genetic predisposition to this cancer. Indeed, recent genome-wide association studies have identified common risk alleles that predispose for MM. Yet, the impact of these variants on MM risk is too low to explain familial aggregation of MM. High-impact alleles have been identified for other cancers such as ovarian and breast cancer ( BRCA1,-2 ) and melanoma ( CDKN2A ) but the search for such alleles in MM is still in its infancy. In order to identify high-impact alleles in MM we have performed whole genome/exon sequencing (WGS/WES) in members of MM high risk families. Methods: We included 21 families with multiple cases of MM/MGUS. Whole genome/exome sequencing was performed on a total of 46 affected and 20 unaffected family members. Filtering and prioritization of the variants were performed in accordance with the criteria of our in-house familial cancer variant prioritization pipeline version 2 (FCVPPv2). Loss-of-function variants were further screened using MutPred-LOF, Translate tool and IntOGen/c-BioPortal in order to discriminate pathogenic and neutral variants, to translate a nucleotide sequence to a protein sequence and to visualize the domain affected by the variant and the portion of the protein lost after the newly formed stop codon. Variants were analyzed for predicted effects on splicing by using Human Splicing Finder. Results: We found a total of 148 potentially pathogenic variants, 109 non-synonymous and 39 LOF, in 18 out of 21 MM families. Among our genes, many affect protein metabolism, immune system, and other have known links to carcinogenesis. Additionally, some of them are known to interact with key signaling pathways in MM, including PI3K/Akt/mTOR, Ras/Raf/MEK/MAPK, JAK/STAT, NF-κB, Wnt/β-catenin, and RANK/RANKL/OPG, showing congruency with previously reported literature. Exposure to androgen deprivation therapy and risk of anastomotic leakage after colorectal cancer surgery

Aim: The risk of anastomotic leakage after colorectal cancer surgery is higher in men, regardless of the anatomical location. Previous studies suggest that this might be due to hormonal differences. The aim of this work was to investigate whether androgen deprivation therapy influenced the incidence of anastomotic leakage. Method: This is a nationwide registry-based study of men operated on between 2007 and 2021 for colorectal cancer with an anastomosis. Exposure to androgen deprivation therapy (prescribed drugs or surgical castration) was related to anastomotic leakage using mixed-effects logistic regression models. Two control groups were formed: one without and one with prostate cancer but without androgen deprivation. To study the potential target for androgen effect in intestinal tissue, androgen receptor expression was evaluated using immunohistochemistry in a smaller independent cohort to compare receptor expression in relation to leakage. Results: Some 24 611 men were included in the registry study, of whom 2.4% were exposed to androgen deprivation therapy. In this exposed group, compared with unexposed men with and without prostate cancer, respectively, leak rates were 3.7%, 5.6% and 5.8%, respectively. After adjustment, a nonsignificant reduction of anastomotic leakage in the exposed group was detected (OR 0.70, 95% CI 0.45–1.09) compared with men without prostate cancer. Tissue expression of androgen receptor was very low among patients with and without leakage, albeit with a trend of higher expression among the latter. Conclusion: Anastomotic leakage rates might be lower in men exposed to androgen deprivation therapy before surgery for colorectal cancer, although this finding must be interpreted cautiously. Effects on anastomotic healing do not seem to be mediated through classical androgen receptor signalling in the intestine. © 2025 The Author(s). Introduction: The coronavirus disease 2019 (COVID-19) pandemic has increased the use of technology for communication including departmental conferences, working remotely, and distance teaching. Methods to enable these activities should be developed and promulgated. Objective: To repurpose a preexisting educational website to enable the development of a COVID-19 autopsy biorepository to support distance teaching and COVID-19 research. Methods: After consent was obtained, autopsies were performed on patients with a confirmed positive severe acute respiratory syndrome coronavirus-2 reverse-transcriptase-polymerase-chain reaction test. Autopsies were performed according to a COVID-19 protocol, and all patients underwent both gross and microscopic examination. The H and E histology slides were scanned using a Leica Biosystems Aperio CS ScanScope whole slide scanner and the digital slide files were converted to deep zoom images that could be uploaded to the University of Alabama at Birmingham (UAB) Pathology Educational Instructional Resource website where virtual microscopy of the slides is available. Results: A total of 551 autopsy slides from 24 UAB COVID-19 cases, 1 influenza H1N1 case and 1 tuberculosis case were scanned and uploaded. Five separate COVID-19 research teams used the digital slides remotely with or without a pathologist on a Zoom call. The scanned slides were used to produce one published case report and one published research project. The digital COVID-19 autopsy biorepository was routinely used for educational conferences and research meetings locally, nationally and internationally. Conclusion: The repurposing of a pre-existing website enabled telepathology consultation for research and education purposes. Combined with other communication technology (Zoom) this achievement highlights what is possible using pre-existing technologies during a global pandemic. © 2021 Wolters Kluwer Medknow Publications. Background HPV-associated oral epithelial dysplasia (HPVOED) is characterized by oral epithelial dysplasia linked to the human papillomavirus (HPV). These lesions exhibit distinctive cytopathologic changes resulting from HPV infection, carrying an associated risk of progressing to oral squamous cell carcinoma. The unique histopathologic features of these lesions are outlined in the WHO classification. The use of toluidine blue staining (TBS) is an effective and noninvasive technique that has been employed for the early detection of premalignant and malignant lesions. However, it has not yet been utilized in the evaluation of HPVOED. Objective Assessing the effectiveness of TBS as a tool for the detection of HPVOED. Methods Thirty-four patients with HPV who attended dental clinics at UdeG, UJED, and UAEM were evaluated. We performed clinical evaluations, TBS, and biopsies. Expert pathologists determined HPVOED using the binary classification. We also correlated the TBS results with the histopathological grade. Results The average age was 35 years, with a predominance of female patients. In 4 cases, positive TBS cases were found. The most common changes in the histopathology were papillary and warty architecture and a higher nucleus-cytoplasm ratio. Mild dysplasia was the sole diagnosis, with no cases recurring during the 6 to 24 months of follow-up. Conclusions This is the first study to evaluate HPVOD with TBS. Our results were consistently negative for mild oral dysplasia. We hypothesize that TBS will be positive in cases of severe HPVOD. Therefore, it is necessary to study these cases to assess the efficacy of TBS or dilucidate the reasons to probably false positives.

Surgical resection is the major method for treating invasive or recurrent noninvasive bladder cancer. However, SR still causes additional risks of urinary tract infection, bleeding and some complications that may be life-threatening. Therefore, it is of urgent need for new diagnosis and therapy scheme for BC. Herein, we proposed a strategy that combines dual-modality 3D NIR-II PA and OCTA molecular imaging to guide visually interventional PTT for BC. Additionally, we developed an excellent NIR-II hyaluronic acid-IR-1048 with liposome-coated (HAPO-1048) photothermal agent for PTT of CD44 overexpressing orthotopic BC, which showed strong NIR-II optical absorption, preferable tumor targeting, excellent biocompatibility and high PTT efficacy. Combined with the NIR-II PA response of the HAPO-1048, we can achieve high sensitivity and speciﬁcity (about 2 mm diameter) dynamic monitoring and investigate the therapeutic efficacy of PTT at larger penetration depth (4.38 mm under the tissue) in orthotopic BC. Simultaneously, we can map the dynamic changes of 3D PA signals and HAPO-1048 NPs enrichment, which is closely related to the PTT efficacy. Significantly, the whole changes of complex microvascular morphometrics and angiogenesis of BC were well surveilled and assessed in vivo using OCTA. Lastly, minimally invasive interventional PTT strategy was implemented to achieve a controllable and precise therapy for BC based on laparoscopy and US guidance. In conclusion, we present a multimodal NIR-II optical imaging synergy laparoscopy interventional scheme, which should expand the current PTT technology, leading to a significant ongoing in treatment of primary tumors of urinary system. © 2022, The Authors.
