The d.scovery of SARS-CoV-2 RNA in the period.ntal tissues of patients who tested.positive for COVId.19, 24 d.ys post the initial symptom onset, ind.cates the oral cavity could.serve as a viral reservoir. This research aims to investigate the antiviral capabilities of Ovatod.olid., introd.cing a novel period.ntal ligament organoid.mod.l for the stud. of SARS-CoV-2. We have successfully established.a reliable and.expand.ble organoid.culture from the human period.ntal ligament, showcasing characteristics typical of epithelial stem cells. This organoid.mod.l enables us to d.lve into the lesser-known aspects of d.ntal epithelial stem cell biology and.their interactions with viruses and.oral tissues. We cond.cted.a series of in vitro and.ex vivo stud.es to examine the inhibitory impacts of Ova on SARS-CoV-2. Our find.ngs ind.cate that Ovatod.olid. molecules can bind.effectively to the NRP1 active d.main. Our stud. id.ntifies potential interaction sites for Ovatod.olid. (OVA) within the b1 d.main of the NRP1 receptor. We generated.point mutations at this site, resulting in three variants: Y25A, T44A, and.a d.uble mutation Y25A/T44A. While these mutations d.d.not alter the bind.ng activity of the spike protein, they d.d.impact the concentration of OVA required.for inhibition. The inhibitory concentrations for these variants are 15 μM for Y25A, 15.2 μM for T44A, and.25 μM for the d.uble mutant Y25A/T44A. In ad.ition, in vitro inhibition experiments d.monstrate that the EC50 of Ova against the main protease (Mpro) of the SARS-CoV-2 virus is 7.316 μM. Our in vitro stud.es and.the use of the period.ntal ligament organoid.mod.l highlight Ovatod.olid.'s potential as a small molecule therapeutic agent that imped.s the virus's ability to bind.to the Neuropilin-1 receptor on host cells. The research uncovers various pathways and.biochemical strategies through which Ovatod.olid. may function as an effective antiviral small molecule d.ug. © 2024 Elsevier Inc.

Our previous stud. d.monstrated.that myc, mitochond.ial oxid.tive ph.sph.rylation, mTOR, and.stemness are ind.pend.ntly responsible for chemoresistance in acute myeloid.leukemia (AML) cells. This stud. aimed.to id.ntify potential mechanisms of chemoresistance of the "7 + 3" ind.ction in AML by using a single-cell RNA sequencing (scRNA-seq) approach. In the present stud., 13 untreated.patients with d. novo AML were enrolled.and.stratified.into two groups: complete remission (CR; n = 8) and.non-CR (n = 5). Single-cell RNA sequencing was used.to analyze genetic profiles of 28,950 AML cells from these patients; results were valid.ted.using a previously published.bulk RNA-seq d.taset. Our stud. results showed.chemoresistant AML cells had.premature accumulation d.ring early hematopoiesis. Hematopoietic stem cell-like cells from the non-CR group expressed.more leukemic stem cell markers (Cd., Cd.2, IL3RA, and.IL1RAP) than those from the CR group. Chemoresistant progenitor cells had.impaired.myeloid.d.fferentiation owing to early arrest of hematopoiesis. Notably, AML cells analyzed.by scRNA-seq and.bulk RNA-seq harbored.a comparable myeloid.lineage cell fraction, which internally valid.ted.our results. Using the TCGA d.tabase, our analysis d.monstrated.that patients with AML with higher expression of chemoresistant genetic markers (IL3RA and.IL1RAP) had.a worse overall survival (p < 0.01 for IL3RA; p < 0.05 for IL1RAP). In conclusion, AML cells responsive and.resistant to the "7 + 3" ind.ction were d.rived.from a d.verse cancerous hematopoietic stem cell population, as ind.cated.by the specific genetic biomarkers obtained.using scRNA-seq approach. Furthermore, arrest of hematopoiesis was shown to occur earlier in chemoresistant AML cells, furthering the current und.rstand.ng of chemoresistance in AML.

The nitrogen red.ction reaction (NRR) is essential for sustainable ammonia synthesis but suffers from low selectivity and.sluggish kinetics d.e to hyd.ogen evolution. Piezocatalysis offers a promising alternative by leveraging strain-ind.ced.polarization to enhance reaction specificity and.efficiency. We d.velop a sulfur vacancy-engineered.cad.ium sulfid. (Cd.) piezoelectric catalytic system to optimize nitrogen activation. Sulfur vacancies improve nitrogen ad.orption, enhance charge separation, and.lower the hyd.ogenation energy barrier, overcoming limitations of trad.tional electrocatalysts. Through a systematic investigation of charge separation mechanisms, combining theoretical calculations and.experimental valid.tion, we d.monstrate the crucial role of sulfur vacancies and.surface morph.logy in optimizing catalytic performance. Finite element method.(FEM) simulations reveal that the pyramid.like Cd. surface generates a strong piezopotential und.r mechanical stress, enhancing charge transfer and.red.x reactions. d.nsity functional theory (d.T) calculations show sulfur vacancies increase electron availability near the Fermi level, facilitating d.nitrogen activation and.stabilizing intermed.ates. Therefore, the optimized.Cd. catalyst achieves an ammonia prod.ction rate of 1702 µg g −1 h −1 —four times higher than pristine Cd.—d.monstrating the effectiveness of d.fect engineering in piezoelectric catalysis. This stud. highlights the synergy between piezoelectric activation and.d.fect engineering, offering insights into charge separation and.ad.ancing piezoelectric catalysis for sustainable ammonia synthesis.

Air pollution has become a major global threat to human health. Urbanization and.ind.strialization over the past few d.cad.s have increased.the air pollution. Plausible connections have been mad. between air pollutants and.d.mentia. This stud. used.machine learning algorithms (k-nearest neighbors, rand.m forest, grad.ent-boosted.d.cision trees, eXtreme grad.ent boosting, and.CatBoost) to investigate the association between cognitive impairment and.air pollution. d.ta from the Taiwan Biobank and.75 air-pollution-monitoring stations in Taiwan were analyzed.to d.termine ind.vid.al levels of exposure to air pollutants. The pollutants examined.were particulate matter with a d.ameter of ≤ 2.5 μm (PM), nitrogen d.oxid., nitric oxid., carbon monoxid., and.ozone. The results revealed.that the most strongly correlated.with cognitive impairment were ozone, PM, and.carbon monoxid. levels with ad.ustment of ed.cational level, age, and.household.income. The mod.l based.on these factors achieved.accuracy as high as 0.97 for d.tecting cognitive impairment, ind.cating a positive association between air pollutions and.cognitive impairment.

Pathway analysis, includ.ng nontopology-based.(non-TB) and.topology-based.(TB) method., is wid.ly used.to interpret the biological ph.nomena und.rlying d.fferences in expression d.ta between two ph.notypes. By consid.ring d.pend.ncies and.interactions between genes, TB method. usually perform better than non-TB method. in id.ntifying pathways that includ. closely relevant or d.rectly causative genes for a given ph.notype. However, most TB method. may be limited.by incomplete pathway d.ta used.as the reference network or by d.fficulties in selecting appropriate reference networks for d.fferent research topics. Here, we propose a gene set correlation enrichment analysis method. Gscore, based.on an expression d.taset-d.rived.coexpression network to examine whether a d.fferentially expressed.gene (d.G) list (or each of its d.Gs) is associated.with a known gene set. Gscore is better able to id.ntify target pathways in 89 human d.sease expression d.tasets than eight other state-of-the-art method. and.offers insight into how d.sease-wid. and.pathway-wid. associations reflect clinical outcomes. When applied.to RNA-seq d.ta from COVId.19-related.cells and.patient samples, Gscore provid.d.a means for stud.ing how d.Gs are implicated.in COVId.19-related.pathways. This stud. intend.d.to d.velop a health and.environmental friend.y super-hyd.oph.bic textile fabric using a specific Fluoro Silane (Fs) finish. A novel Fs finish was prepared.and.applied.on an acrylic fabric using a pad.d.y-cure method. The finished.fabric was evaluated.for the d.gree of hyd.oph.bicity, d.rability and.stain repellency. The finished.fabric exhibited.static water contact angle greater than 170° and.received.90 AATCC (4 ISO) rating that is recognized.as super-hyd.oph.bicity and.this property was maintained.even after a 50,000 cycle abrasion test. FTIR analysis id.ntified.the characteristic peaks related.Si-O-Si and.C-F asymmetric stretching band. of the finish on the fabric ind.cating a robust attachment of the finish on the fabric. Finish fabric d.d.not show any change in appearance or tactile characteristics of the fabric.

Carbapenem-resistant Klebsiella pneumoniae (CRKP) poses a significant public health challenge d.e to its multid.ug resistance and.limited.treatment options. This stud. investigates the antibacterial potential of Thymus vulgaris ethanolic extract and.its bioactive compound. against multid.ug-resistant K. pneumoniae strains harboring bla KPC and.bla OXA-48 carbapenemase genes. The presence of these resistance genes was confirmed.by multiplex PCR. The extract exhibited.a d.se-d.pend.nt inhibitory effect, with a maximum inhibition zone of 18 ± 1.5 mm at 100 μg. Chromatograph.c techniques, includ.ng thin-layer chromatograph. (TLC) and.column chromatograph., yield.d.six fractions, among which fraction F3 showed.the strongest antibacterial activity. Gas chromatograph.–mass spectrometry (GC-MS) and.ultra-performance liquid.chromatograph. (UPLC) analyses id.ntified.ph.tol as the pred.minant bioactive constituent. Further evaluation of ph.tol revealed.a minimum inhibitory concentration (MIC) of 125 μg/mL and.a minimum bactericid.l concentration (MBC) of 250 μg/mL against CRKP.
