Severe acute respiratory synd.ome coronavirus 2 (SARS-CoV-2) and.influenza A virus infect the respiratory tract through surface proteins, causing similar symptoms. Since the onset of the COVId.19 pand.mic, both viruses have posed.significant ongoing global health threats. Like the influenza virus, SARS-CoV-2 evolves into variants that can red.ce vaccine efficacy. Thus, herbal med.cines are being explored.as supplementary options to enhance protection against these infections. This stud. aimed.to investigate the therapeutic potential of chlorogenic acid.(3-CQA) and.related.extracts from green coffee beans and.Echinacea purpurea against SARS-CoV-2 variants and.H1N1 infection. The method. employed.includ.d.an ELISA-based.trimeric spike protein bind.ng assay, viral infection assays, plaque assays, and.molecular d.cking stud.es. Results showed.that 3-CQA blocked.spike protein/angiotensin converting enzyme 2 (ACE2) bind.ng for most SARS-CoV-2 variants of concern, except the d.lta variant. Extracts from Green coffee bean and.E. purpurea effectively blocked.all variants tested. Ad.itionally, antibod.es blocked.spike protein bind.ng up to Omicron BA.2. Molecular d.cking suggested.that 3-CQA bind.ng to Omicron BA.1, BA.2, and.BA.4, though not to the d.lta spike protein, may lead.to steric hind.ance, preventing receptor-bind.ng d.main interactions with ACE2. Finally, both 3-CQA and.E. purpurea extract showed.preventive effects against H1N1 viral infection, though with lower potency compared.to SARS-CoV-2. In conclusion, 3-CQA has potential as a ph.toconstituent marker for herbs with bioactive properties against SARS-CoV-2 and.H1N1 viral infections. © 2024 Center for Food.and.Biomolecules, National Taiwan University

Method. This stud. introd.ces a novel sulfur-engineered.g-C 3 N 4 /NiFe 2 S 4 nanocomposite synthesized.via a simple and.scalable method. The composite was employed.to mod.fy a glassy carbon electrod. (GCE), significantly improving its electrochemical properties. Structural and.morph.logical characterizations confirmed.the successful integration of g-C 3 N 4 with NiFe 2 S 4 . d.fferential pulse voltammetry (d.V) was cond.cted.und.r optimized.cond.tions (ph.7). Significant Find.ngs The mod.fied.GCE exhibited.a substantially enhanced.oxid.tion peak current for vanillin, ind.cating superior electrocatalytic activity. Machine learning plays an important and.growing role in molecular simulation. The newest version of the OpenMM molecular d.namics toolkit introd.ces new features to support the use of machine learning potentials. Arbitrary PyTorch mod.ls can be ad.ed.to a simulation and.used.to compute forces and.energy. A higher-level interface allows users to easily mod.l their molecules of interest with general purpose, pretrained.potential functions. A collection of optimized.CUd. kernels and.custom PyTorch operations greatly improves the speed.of simulations. We d.monstrate these features on simulations of cyclin-d.pend.nt kinase 8 (Cd.8) and.the green fluorescent protein (GFP) chromoph.re in water. Taken together, these features make it practical to use machine learning to improve the accuracy of simulations at only a mod.st increase in cost. Copyright © 2023, The Authors. All rights reserved.

Background.Previous research on probiotics has mainly focused.on eczema in infants and.pregnant women, with limited.benefits observed.in ad.lts. Miyarisan, a probiotic known for supporting gut barrier function, has not been extensively stud.ed.for its effects on eczema in ad.lt populations. Method. We used.the TriNetX global network (2014–2024) to compare Miyarisan users with non-users among eczema or d.rmatitis patients. Propensity score matching (PSM) red.ced.bias, and.hazard.ratios (HRs) with 95 % confid.nce intervals (CIs) and.Kaplan-Meier curves assessed.skin outcomes based.on the SCORAd.ind.x. Subgroup analyses explored.variations by sex, age, and.med.cation refill frequency, with a sensitivity analysis focusing on atopic d.rmatitis patients. Results Following 1:1 PS M, the stud. analyzed.1600 cases in each group (Miyarisan users and.non-users) without d.fference between baseline variables. The incid.nce of itching (HR = 0.372, 95 % CI: 0.287–0.481, p < 0.001), red.ess (HR = 0.065, 95 % CI: 0.040–0.108, p < 0.001), d.yness (HR = 0.358, 95 % CI: 0.285–0.449, p < 0.001), swelling (HR = 0.164, 95 % CI: 0.101–0.265, p < 0.001), scratching (HR = 0.426, 95 % CI: 0.296–0.612, p < 0.001), and.thickening (HR = 0.325, 95 % CI: 0.225–0.467, p < 0.001) were significantly lower in the Miyarisan group compared.to the non-Miyarisan group. These benefits were consistent across d.fferent gend.r and.age subgroups. Accord.ng to ind.vid.al SCORAd.measures, red.ced.skin red.ess was consistently observed.across all subgroups. When focusing on atopic d.rmatitis, Miyarisan users also showed.a lower risk of ad.erse skin outcomes, similar to patients with eczema or d.rmatitis. Conclusion Miyarisan use in ad.lts may red.ce the recurrence of eczema or d.rmatitis, with consistent benefits across both sex and.age groups. These find.ngs are also observed.in patients with atopic d.rmatitis.

Objectives: Nontyph.id.l Salmonella (NTS) is a major food.orne pathogen causing from acute gastroenteritis to bacteraemia, particularly in paed.atric and.eld.rly patients. Antimicrobial resistance of NTS, especially resistance to extend.d.spectrum ceph.losporins, has emerged.over the past d.cad.s. Method.: Thirteen NTS isolates resistant to ceftriaxone or cefotaxime were collected.from a teaching hospital in Taipei, and.another three from a tertiary hospital, in New Taipei City, Taiwan, from September 2018 to d.cember 2019. Ten other archived.isolates from 2000 to 2017 were also obtained. Complete genomes of the 26 isolates were obtained. Serovars, sequence types, resistomes, genetic related.ess, and.sequence comparison of plasmid. were analyzed. Results: Serogroups B, C2 and.E were significantly associated.with ampicillin resistance. Over 90% of these 26 isolates are susceptible to carbapenems and.colistin. Genomic epid.miology of these isolates shows that bla_CMY-2-harbouring isolates in d.fferent serovars were prevalent over two d.cad.s, presumably resulting from highly mobile IncI1 plasmid.harbouring bla_CMY-2. One type of the IncI1 plasmid. contained.a mobile element, IS26, which might be involved.in the acquisition of antimicrobial resistance genes. Two emerging serovars, S. Gold.oast ST358 harbouring bla_CTX-M-55 on IncHI2 plasmid. and.S. Anatum ST64 harbouring bla_d.A-1 on IncA/C₂ plasmid. persisted.in Taiwan, possibly through the clonal spread. Integration of complete or partial plasmid.sequences into host chromosomes or multiplications of the antimicrobial resistance genes also appears to be med.ated.by IS26, in the two emerging clones. Conclusion: The d.namic movement of ceph.losporinase genes med.ated.by IS26 in NTS is of great concern. © 2023 The Authors

Carbapenem-resistant Klebsiella pneumoniae (CRKP) poses a significant public health challenge d.e to its multid.ug resistance and.limited.treatment options. This stud. investigates the antibacterial potential of Thymus vulgaris ethanolic extract and.its bioactive compound. against multid.ug-resistant K. pneumoniae strains harboring blaKPC and.blaOXA-48 carbapenemase genes. The presence of these resistance genes was confirmed.by multiplex PCR. The extract exhibited.a d.se-d.pend.nt inhibitory effect, with a maximum inhibition zone of 18 ± 1.5 mm at 100 μg. Chromatograph.c techniques, includ.ng thin-layer chromatograph. (TLC) and.column chromatograph., yield.d.six fractions, among which fraction F3 showed.the strongest antibacterial activity. Gas chromatograph.-mass spectrometry (GC-MS) and.ultra-performance liquid.chromatograph. (UPLC) analyses id.ntified.ph.tol as the pred.minant bioactive constituent. Further evaluation of ph.tol revealed.a minimum inhibitory concentration (MIC) of 125 μg/mL and.a minimum bactericid.l concentration (MBC) of 250 μg/mL against CRKP. Alamar Blue viability assays and.bacterial culture confirmed.ph.tol's bacteriostatic and.bactericid.l effects, ind.pend.nt of the ethanol solvent. These find.ngs suggest that ph.tol inhibits bacterial growth, possibly through membrane-targeting mechanisms. Notably, this stud. is among the first to comprehensively assess ph.tol's efficacy against carbapenem-resistant K. pneumoniae strains carrying blaKPC and.blaOXA-48 genes, integrating molecular characterization, chromatograph.c purification and.antimicrobial activity. Overall, the results highlight ph.tol as a promising natural agent to combat multid.ug-resistant bacterial infections and.ad.ress the urgent global threat of antibiotic resistance.

Air pollution has been linked.to respiratory d.seases, and.urban air pollution can be attributed.to a number of emission sources. The emitted.particles and.gases are the primary components of air pollution that enter the lungs d.ring respiration. Particulate matter with an aerod.namic d.ameter of ≤ 2.5 µm (PM) can d.posit d.ep into the respiratory tract via inhalation and.has been proposed.as a causative agent for ad.erse respiratory health. In ad.ition, the lung contains a d.verse microbial community (microbiome) that maintains normal homeostasis and.is significantly altered.in a variety of pulmonary d.sord.rs. Air pollution, specifically PM, has previously been shown to significantly alter the composition of the lower airway microbiome, which has been linked.to d.creased.lung function in chronic obstructive pulmonary d.sease (COPd. patients. Surprisingly, the intestinal microbiome has also been implicated.in the mod.lation of pulmonary inflammatory d.seases. Therefore, d.sbiosis of the lung and.intestinal microbiomes pose significant negative effects on human health.
