Tn antigen suppresses lipopolysaccharide-induced dermatitis via Clec10a

C-type lectin domain family 10 member A (Clec10a), also called macrophage galactose-type lectin 1 (MGL1) or Cd301a, is expressed on myeloid cells, including macrophages in the skin. Several pieces of evidence suggest that Clec10a physically binds to O-linked mucin-like molecules, but the functional ligand that mediates signaling in macrophages remains undetermined. We previously reported that house dust mite (HdM) treatment on the skin produced a greater amount of Toll-like receptor 4 (TLR4)-mediated inflammatory cytokines from macrophage in Clec10a-deficient mice than in wild-type (WT) mice, suggesting that a functional ligand for Clec10a contained in HdM inhibited TLR4-mediated skin macrophage activation. In this study, we investigated the glycan structure of a functional Clec10a ligand (Clec10a-L) contained in HdM. We showed that Clec10a bound to O-linked mucin-like molecules such as polyacrylamide (PAA)-conjugated Tn antigen (Tn antigen-PAA), Core1-PAA, LacNAc-PAA, Lewis A-PAA, and Lewis X-PAA. Among them, plate-coated Tn antigen-PAA, Lewis A-PAA, and Lewis X-PAA activated Clec10a expressed on the reporter cells. However, intradermal injection of only Tn antigen-PAA, but not Lewis A-PAA and Lewis X-PAA, ameliorated LPS-induced dermatitis in WT but not Clec10a-deficient mice. Moreover, Tn antigen-PAA suppressed LPS-induced production of inflammatory cytokines IL-6 and TNF-α by bone marrow-derived cultured macrophage in vitro . These results indicate that Tn antigen is a functional Clec10a-L in HdM that suppresses TLR4-induced macrophage activation in the skin.

In recent years, bacterial cellulose (BC), a crystalline and nanoscale fibrillar polymer, has garnered significant interest due to its superior physical, chemical, and mechanical properties compared to plant cellulose. Inherent features of BC, which include high biodegradability, mechanical strength, and biocompatibility, make it a suitable material for use in a wide variety of applications, particularly in the domains of biomedicine and environmental science. However, realizing its full potential requires targeted chemical or physical modifications. Recent developments have focused on in-situ and ex-situ BC modification for various applications. Incorporating functional compounds into cellulose during BC synthesis enhances its strength and functional properties. Additionally, the material properties of BC, such as water content, thermal stability, and antibacterial capabilities, can be enhanced through post-synthesis modifications, including chemical acetylation, phosphorylation, and integration with other biopolymers. Currently, the applicability of BC is being improved using new modification techniques, which involve genetically engineered strains to enhance polymer characteristics and market potential. The review highlights recent advancements in BC research and novel modification strategies that enhance its applications in advanced biomedical engineering and sustainable packaging. © 2025

University of Cincinnati Cincinnati OH David Greenberg is a Professor of Chemical Engineering at the University of Cincinnati a position he has held for the past 12 years. He was educated at Carnegie Tech (CMU) the Johns Hopkins University and Louisiana State University when he received his B.S. M.S. and Ph.D. Degrees in Chemical Engineering.

Techniques for the rapid determination of the presence and level of microorganisms have long been of concern to a host of scientific disciplines that span the medical pharmaceutical, and processing fields among others. It appears to be feasible that by using the quantitative, nonfluorescent substrate probe procedures, a measure of bacterial presence and characterization can be achieved. An order of magnitude light-scattering bacterial concentration determination in conjunction with the dAF fluorescent probe technique would significantly increase microorganism characterization. K//M values, however, appear to be of limited utility as a tool in microorganism determination, other than possibly providing information on the class of enzyme that is present.

Mitochondria, often referred to as the "powerhouses" of eukaryotic cells, play a critical role in maintaining cellular homeostasis and supporting essential life functions, particularly in cell survival and apoptosis. This study introduces a composite formulation—combining carrageenan (Car), calcium peroxide (CaO2), and microparticles, derived from soybean oil lecithin, enhanced with photothermally active polypyrrole (PPy)—to create Car-Lec-PPy MP, aimed at promoting mitochondrial activation in cells. The investigation focuses on intercellular mitochondrial transfer and activation as a therapeutic strategy. The formulation seeks to stimulate mitochondrial biogenesis and enhance cellular resilience through targeted mitochondrial activation, leveraging the synergistic effects of near-infrared (NIR) therapy. In the in vivo experiment, phototherapeutic Car-Lec-PPy MP was administered to rodent muscle tissue. The results showed increased expression of heat-shock proteins and reduced oxidative stress in the treated muscle. Preliminary findings suggest that this approach not only boosts mitochondrial activity but also improves cell proliferation and survival, presenting significant potential for applications in regenerative medicine and cellular therapy. © 2025 Elsevier B.V.

Berm soils at small arms firing ranges (SAFRs) are contaminated with metals as bullets impact the soil and corrode. These berms generally lack vegetation and frequently suffer from erosion that may transport heavy metals (e.g. lead) into surface water and, occasionally, into groundwater. Plants are generally useful for stabilizing metals in SAFR soils by reducing particulate runoff. However, results have shown that plants appear to increase lead solubility in surface runoff and leachate. Studies were conducted to investigate this phenomenon that plants enhance lead solubility and complexation. Hydroponic bermuda plant culture solutions were prepared. Plant culture and soluble lead solutions were reacted to form lead complexes. Ion exchange filtration was performed to separate lead complexes based on relative molecular sizes. Results showed that bermuda cultures increased lead solubility and formed measurable lead complexes for fractionated sizes. Continued research will determine the effect of soluble plant organic compounds on metal dissolution. © 2010 ASCE.

Moral Reasoning and Political Beliefs of dutch Adolescents and Young Adults

His research interests are cognitive moral and political development of adolescents and young adults. TOM VERBOGT studied at Catholic University of Nijmegen and is completing his Ph.D. at Utrecht University. His research interests are moral and political development. WILMA VOLLEBERGH completed her Ph.D. at Utrecht University. She is currently the coordinator of a research program on education and the development of adolescents in different ethnic groups. Her research interests include the etiology of psychopathology cultural orientations identity development and psychosocial problems in adolescents.

The relation between moral reasoning and political beliefs is studied by distinguishing between two different domains of political beliefs. These domains were related to the level of moral reasoning in adolescents and young adults. A sample of 1,968 dutch adolescents and young adults responded in 1991 to the defining Issues Test (J. R. Rest, 1979a) and seven political attitude scales. This procedure was repeated in 1994 for 1,498 dutch adolescents and young adults, of whom 911 individuals had also participated in the 1991 survey. Thus, longitudinal data were available for this latter group. Results showed, first, that a distinction between economic and cultural dimensions of political beliefs is useful. Second, the level of moral reasoning related to political beliefs, but only in the cultural domain. Third, during adolescence, this correlation increased with age and educational level. The relation between moral reasoning and political beliefs is discussed in terms of similarities in content versus cognitive determinants.

Mesona procumbens Hemseley is a well-known traditional herbal medicine used for heat-related ailments. In Taiwan, boiled extracts of M. procumbens are also used as desserts called grass jelly. In this study, the hexane extract from 75% EtOH of M. procumbens showed potent activities on inhibition of E. coli β-glucuronidase (eβG) and NO production and cytotoxicity against MCF-7 and HepG2 cancer cell lines. Furthermore, using various flash columns and HPLC chromatography on the bioactive layer led to the isolation of twelve compounds (1–12), including a new ent-kaurene, mesokaurol A (1), and a new germacrene derivative, mesogermapene A (2). Their structures were elucidated by extensive spectroscopic analyses, especially 2 d NMR and mass data. Biological assays showed that compound 9 (linolenic acid) had specific activity on inhibition of eβG (68.27%) at 100 µg/mL but was non-inhibitory to human β-glucuronidase. Compound 1 possessed significant cytotoxicity against MCF-7 (EC₅₀ = 9.76 µM) and HepG2 (EC₅₀ = 8.64 µM) cancer cell lines. © 2022 Informa UK Limited, trading as Taylor & Francis Group.
