The performance of an electrochemiluminescence (ECL) immunosensor was improved with a particle gradient. SiO2-coated magnetic beads were adopted as nanocarriers for gradient manipulation and immobilized with the primary antibody. Cadmium telluride quantum dots were coated with a layer of protein G for conjugation and orientation of the secondary antibody as signal labels. ECL immunosensor gradients on the electrode were formed by magnetolithography (ML) with magnetized nickel masks of column and stripe arrays. The immunosensor generally aggregated as an island on the substrate, leading to a decrease of efficiency in the characteristic signals. Stripe arrays of magnetized nickel were designed to generate cylindrical magnetic flux on the substrate to improve the particle manipulation with the gradient. Various gradients of the sandwich-structured immunosensor substantially affected the electrochemical performance. Compared to the gradient-free immunosensor, the gradient of the immunosensor generated by ML using a 3 μm stripe array mask enhanced the ECL intensity ∼2.2 times. The results of quantification of epithelial cell adhesion molecules (EpCAM) with the gradient immunosensor showed a broad linear range (15-420 pg mL-1), a low limit of detection (5.5 pg mL-1), and high reliability for EpCAM-spiked serum samples, indicating that the immunosensor gradient substantially enhances the performance of the ECL assay. © 2022 American Chemical Society.

Graph Neural Networks (GNNs) are powerful for the representation learning of graph-structured data. Most of the GNNs use a message-passing scheme, where the embedding of a node is iteratively updated by aggregating the information from its neighbors. To achieve a better expressive capability of node influences, attention mechanism has grown to be popular to assign trainable weights to the nodes in aggregation. Though the attention-based GNNs have achieved remarkable results in various tasks, a clear understanding of their discriminative capacities is missing. In this work, we present a theoretical analysis of the representational properties of the GNN that adopts the attention mechanism as an aggregator. Our analysis determines all cases when those attention-based GNNs can always fail to distinguish certain distinct structures. Those cases appear due to the ignorance of cardinality information in attention-based aggregation. To improve the performance of attention-based GNNs, we propose cardinality preserved attention (CPA) models that can be applied to any kind of attention mechanisms. Our experiments on node and graph classification confirm our theoretical analysis and show the competitive performance of our CPA models. In recent years, many mammographic image analysis methods have been introduced for improving cancer classification tasks. Two major issues of mammogram classification tasks are leveraging multi-view mammographic information and class-imbalance handling. In the first problem, many multi-view methods have been released for concatenating features of two or more views for the training and inference stage. Having said that, most multi-view existing methods are not explainable in the meaning of feature fusion, and treat many views equally for diagnosing. Our work aims to propose a simple but novel method for enhancing examined view (main view) by leveraging low-level feature information from the auxiliary view (ipsilateral view) before learning the high-level feature that contains the cancerous features. For the second issue, we also propose a simple but novel malignant mammogram synthesis framework for upsampling minor class samples. Our easy-to-implement and no-training framework has eliminated the current limitation of the CutMix algorithm which are unreliable synthesized images with random pasted patches, hard-contour problems, and domain shift problems. Our results on Vindr-Mammo and CMMd datasets show the effectiveness of our two new frameworks for both multi-view training and synthesizing mammographic images, outperforming the previous conventional methods in our experimental settings. © 2023, CC BY-NC-SA.

This work presents oxidation behaviours of Al-bearing Cr-based alloys GA1 (51Cr-22Al-13Mo-9Ta-5Si in at%) and GA2 (40Cr-33Al-13Mo-9Ta-5Si in at%) at 1100–1400 ˚C for up to 24 h. The formation of oxides on the surfaces was influenced by constituent phases;BCC phase was rich in Al and formed only α-Al2O3, complex oxides (Cr2O3, AlTaO4, CrTaO4, and Ta2O5) occurred on top of C14 Laves phase. Sufficient Al content is crucial to the formation of protective α-Al2O3, as GA2 was able to resist severe oxidation at 1400 ˚C with an oxidation weight gain of only 3.3 mg/cm2 after 24 h. © 2024 Elsevier Ltd

Translational readthrough of UGA stop codons by selenocysteine-specific tRNA (tRNASec) enables the synthesis of selenoproteins. Seryl-tRNA synthetase (SerRS) charges tRNASec with serine, which is modified into selenocysteine and delivered to the ribosome by a designated elongation factor (eEFSec in eukaryotes). Here we found that components of the human selenocysteine incorporation machinery (SerRS, tRNASec, and eEFSec) also increased translational readthrough of non-selenocysteine genes, including VEGFA, to create C-terminally extended isoforms. SerRS recognizes target mRNAs through a stem-loop structure that resembles the variable loop of its cognate tRNAs. This function of SerRS depends on both its enzymatic activity and a vertebrate-specific domain. Through eCLIP-seq, we identified additional SerRS-interacting mRNAs as potential readthrough genes. Moreover, SerRS overexpression was sufficient to reverse premature termination caused by a pathogenic nonsense mutation. Our findings expand the repertoire of selenoprotein biosynthesis machinery and suggest an avenue for therapeutic targeting of nonsense mutations using endogenous factors. © 2023 The Author(s). Cyclin-dependent protein kinase 8 (CdK8) plays important roles in regulating fibrotic growth factors and inflammatory signaling pathways. Long-term chronic inflammation of the lungs can lead to idiopathic pulmonary fibrosis (IPF). Abnormal alveolar epithelial regeneration leads to the release of various fibrotic growth factors and the activation of inflammatory cells. CdK8 regulates profibrotic cytokines broadly implicated in the pathogenesis of fibrosis. Therefore, inhibition of CdK8 is considered a promising strategy for treating IPF. Here, CdK8 inhibitors were designed and optimized using a fragment-based drug design strategy. Testing results revealed that 71% of the synthesized compounds inhibited CdK8 activity better than the original compound E966–0530. Of these compounds, compound 4k exhibited the strongest CdK8 enzyme-inhibiting activity (IC₅₀ =129 nM). Notably, it displayed a 13-fold increase in potency when compared to E966–0530. Experiments on toxicity and inhibition of epithelial-mesenchymal transition (EMT) protein expressions showed that compound 4k can inhibit EMT protein expressions, but with no significant cytotoxicity for alveolar epithelial cells. Compound 4k showed a potent inhibitory effect in cell migration assays. Furthermore, compound 4k significantly inhibited the phosphorylation of p-Smad3 and RNA Pol II, which are critical mediators in the fibrotic response signaling pathway. Compound 4k remarkably reduced TGF-β1-induced oxidative stress. The above results reveal optimized CdK8 inhibitors with potential use for IPF therapeutic treatment. © 2023 The Authors

Augmented reality (AR) is a technology that instantly merges digital pictures with the real scene. The perspective near-eye display is a key module of AR, through which computer-generated pictures or videos can be displayed in the existing environment. The purpose of this research is to establish the system structure of an augmented reality near-eye display device and to design the off-axis two-piece reflective aspherical mirror. Because AR glasses are wearable and are worn close to the eyes, not only the optical performance of the display but also the clear vision of the user needs to be considered. In this study, an off-axis two-piece reflective aspherical mirror is used as the optical system of the AR for displaying the digital image. The micro-display is placed at the focal point of the optical system to form a digital image, and the horizontal movement of the micro-display along the optical axis is controlled by a precise displacement mechanism so that the patient with uncorrected myopia or hyperopia can see the digital picture and real scene. Because the right and left micro-displays move horizontally independently, the user can see the images on the AR device, which becomes the biggest feature of this study.

In this study, metalloanthocyanin-inspired, biodegradable packaging films were developed by incorporating purple cauliflower extracted (PCE) anthocyanins into alginate (AL)/carboxymethyl chitosan (CCS) hybrid polymer matrices based on complexation of metal ions with these marine polysaccharides and anthocyanins. PCE anthocyanins-incorporated AL/CCS films were further modified with fucoidan (Fd) because this sulfated polysaccharide can form strong interactions with anthocyanins. Metals-involved complexation (Ca2+ and Zn2+-crosslinked films) improved the mechanical strength and water vapor permeability but reduced the swelling degree of the films. Zn2+-cross-linked films exhibited significantly higher antibacterial activity than did pristine (non-crosslinked) and Ca2+-cross-linked films. The metal ion/polysaccharide-involved complexation with anthocyanin reduced the release rate of anthocyanins, increased the storage stability and antioxidant capability, and improved the sensitivity of the colorimetric response of the indicator films for monitoring the freshness of shrimp. The anthocyanin-metal-polysaccharide complex film showed great potential as active and intelligent packaging of food products. © 2022 Elsevier Ltd
