In single-cell RNA sequencing (scRNA-seq) data analysis, a critical challenge is to infer hidden cellular dynamic processes from measured static cell snapshots. To tackle this challenge, many computational methods have been developed from distinct perspectives. Besides the common perspectives of inferring pseudotime trajectories and RNA velocities, another important perspective is to estimate the differentiation potential of cells, which is commonly referred to as the "cell potency." In this review, we provide a comprehensive summary of 14 computational methods that define cell potency from scRNA-seq data under different assumptions, some of which are even conceptually contradictory. We divide these methods into three categories: average-based, entropy-based, and correlation-based methods, depending on how a method summarizes gene expression levels of a cell or cell type into a potency measure. Our review focuses on the key similarities and differences of the methods within each category and between the categories, providing a high-level intuition for each method. Moreover, we use a unified set of mathematical notations to detail the 14 methods’ methodologies and summarize their usage complexities, including the number of parameters, the number of required inputs, and the changes from the method description in publications to the method implementation in software packages. In conclusion, we regard cell potency estimation as an open question that lacks a consensus on the optimal approach, calling for benchmark datasets and studies. Through categorization and detailed description of existing methods, we aim to provide a solid foundation for future benchmark studies of cell potency estimation methods. A broader open challenge is to understand the comparative advantages of methods that infer cellular dynamics from scRNA-seq data using diverse perspectives, including pseudotime trajectories, RNA velocities, and cell potency. © 2023, CC BY-NC-Nd.

Global environmental challenges and energy crises have driven researchers to develop multifunctional and highly efficient nanomaterials. This study presents dual-functional NiMoO4 (NMO)/NiO hierarchical microspheres that can serve as supercapacitors and photocatalysts prepared using a microwave-assisted hydrothermal method. The α- and β-phase contents of NMO can be regulated by controlling the post-annealing temperature and ph value of the precursor solution. The as-prepared NMO/NiO nanocomposites exhibited dual Faradaic redox reactions attributed to NMO and NiO, leading to remarkable supercapacitor performance. In addition, the constructed heterojunction between NMO and NiO also improved charge separation, leading to excellent photocatalytic capability. Based on the results, NMO with a higher β-phase content showed better supercapacitive and photocatalytic performance owing to its higher conductivity. The optimal NMO/NiO composite displayed a specific capacitance of 943 F g−1 at 1 A/g and excellent cycling stability, with 83.1 % retention at 5 A/g after 4000 cycles. Additionally, it demonstrated an outstanding photocatalytic capability for the degradation of methylene blue (MB), achieving a rate constant of (0.0113 min−1). © 2024 Elsevier Ltd

People spend most of their time indoors, where they may be exposed to various microorganisms and allergens. In this cross-sectional study, we investigated the relationship between indoor microbial and allergen levels and indoor and outdoor household environmental characteristics. Participants were recruited from an ongoing cohort study in the Greater Taipei Area. Questionnaire administration and environmental sampling were conducted during home visits. Samples were collected from children's bedrooms, living rooms, kitchens, and outdoor areas (balconies) and analyzed for fungal spores, house dust mite allergens (der p 1 and der f 1), endotoxin, and cockroach allergens (Bla g 1). Indoor temperature and relative humidity were also measured, and the land-use characteristics and points of interest surrounding the residences were evaluated within specified radii. The mean levels of outdoor air pollutants were then estimated, and multiple regression analysis was performed to investigate the predictors of indoor microbial and allergen levels. In total, 136 homes were analyzed. The results indicated that dog ownership, air purifier use, and floor cleaning frequency were major predictors of indoor microbial and allergen levels. Moreover, proximity to parks, roads, and forests was associated with elevated levels of indoor microorganisms and allergens. Our results demonstrated that indoor concentrations of certain microorganisms and allergens tended to increase in households located within neighborhoods with particular characteristics. Overall, our research findings contribute to housing intervention protocols and policies aimed at reducing exposure to indoor microorganisms and allergens. Future studies should longitudinally investigate the health effects of indoor microorganisms and allergens. © 2023 Elsevier Ltd

Objective: The study aimed to determine the incidence and risk factors associated with bone cement dislodgement in patients with osteoporotic vertebral compression fracture following balloon kyphoplasty treatment. Methods: A retrospective study was conducted on 203 patients who underwent kyphoplasty in 255 vertebral bodies between January 2017 and december 2021. The patients were categorized into two groups: the bone cement dislodgment group (n = 16) and the non-bone cement dislodgement group (n = 239). Various patient characteristics and radiologic parameters were evaluated. Statistical analysis involved the assessment of the background homogeneity of the group by using independent sample t tests, chi-square tests, and Fisher's exact. Univariate and multivariate logistic regression analyses were performed to explore the impact of background variables on cement dislodgement. Results: The results revealed that split-type fracture (χ² = 31.706, p < 0.001), dISH (χ² = 18.827, p = 0.011), pedicle fracture (χ² = 22.246, p < 0.001), endplate deficit (χ² = 14.023, p < 0.001), posterior wall injury (χ² = 29.124, p < 0.001), and intervertebral vacuum cleft (χ² = 21.469, p < 0.001) were the factors that significantly differed between the two groups. The multivariate logistic regression analysis revealed posterior wall injury (OR = 12.983, p = 0.025) and intervertebral vacuum cleft (OR = 5.062, p = 0.024) to be independent risk factors. Conclusion: The incidence of bone cement dislodgement in our study was 6.3%. This study underscores the importance of using preoperative radiologic parameters to predict the risk of bone cement dislodgement following balloon kyphoplasty. © 2024 Elsevier Masson SAS

Metal oxide heterojunctions have attracted considerable attention in photocatalyst and supercapacitor applications. In this study, NiMn2O4/CoMn2O4 (NMO/CMO) heterojunctions were synthesized using ternary metal-organic frameworks (MOFs) as precursors. After calcination at 500 °C, the as-prepared NMO/CMO composite maintains the porous sheet-like morphology of the ternary MOF, exhibiting a highly porous structure. The heterostructured NMO/CMO composite demonstrates enhanced photocatalytic performance compared with pure NMO and CMO. This enhancement may be attributed to the well-aligned energy levels at the NMO/CMO interface, which effectively reduces charge recombination and facilitates the efficient transport of photogenerated carriers. In addition, the NMO/CMO heterojunction exhibits enhanced supercapacitor performance owing to the increased number of active redox sites and reduced charge transfer resistance. When tested at 3 A/g, the NMO/CMO composite displays a higher capacitance (756.4 F/g) than pure NMO (579.6 F/g) and CMO (519.6 F/g) and remarkable rate capability, maintaining a capacitance of 639 F/g even at 10 A/g. Notably, asymmetric supercapacitors fabricated using NMO/CMO and activated carbon can achieve an energy density of 32.18 W h/kg at 750 W/kg and superior cycling life, retaining 92.1% of its initial capacitance after 3000 cycles. © 2023 Elsevier Ltd

Microplastics pollution is a modern-day prevalent challenge that poses serious threats to the general wellbeing and sustainability of the planet. due to their innately persistent nature, extreme durability, and high stability, they exhibit long residence time and could be transferred through the ecological chains. Current days have witnessed an increase in the accumulation of microplastics waste and debris. This is due to the protective measures imposed by governments and the World Health Organization (WHO) as part of the containment and mitigation efforts of the COVId-19 pandemic. Therefore, the development of versatile, robust, flexible, and efficient removal systems has become a necessity in order to control microplastics pollution. Herein, we review the repercussions of the pandemic on the state of microplastics contamination, sources and identification methods of microplastics in water, their toxicity and risks on the environment, as well as their degradation and conventional removal methods. This review focuses on adsorption and the use of biochar as one of the emerging adsorbents for the removal of microplastics. The efficiency of adsorption using biochar, mechanisms, and applications are discussed. The review addresses the increased rates of microplastics occurrence especially during the era of COVId-19, which requires more efficient and lower cost removal methods. The major conclusions drawn from this review include emphasizing the significant potential of adsorption processes in the removal of microplastics. Biochar is present among the available and most efficient biomass adsorbents especially when combined with other materials. Composite biochar materials play an important role in enhancing the removal and immobility of microplastics. However, more research is required to investigate this at a larger scale, improve efficiency and immobilization techniques. © 2021 The Authors

Treatment-resistant depression (TRd) is a condition wherein patients with depression fail to respond to antidepressant trials. A new form of repetitive transcranial magnetic stimulation (rTMS), called theta-burst stimulation (TBS), which includes intermittent theta-burst stimulation (iTBS) and continuous theta-burst stimulation (cTBS), is non-inferior to rTMS in TRd treatment. However, the mechanism of iTBS and cTBS underlying the treatment of TRd in the prefrontal cortex (PFC) remains unclear. Hence, we applied foot-shock stress as a traumatic event to develop a TRd rat model and investigated the different mechanisms of iTBS and cTBS. The iTBS and cTBS treatment were effective in depressive-like behavior and active coping behavior. The iTBS treatments improved impaired long-term potentiation and long-term depression (LTd), whereas the cTBS treatment only improved aberrant LTd. Moreover, the decrease in mature brain-derived neurotrophic factor (BdNF)-related protein levels were reversed by iTBS treatment. The decrease in proBdNF-related protein expression was improved by iTBS and cTBS treatment. Both iTBS and cTBS improved the decreased α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors and downregulation of mammalian target of the rapamycin (mTOR) signaling pathway. The iTBS produces both excitatory and inhibitory synaptic effects, and the cTBS only produces inhibitory synaptic effects in the PFC. © 2023

Community shift of aquatic insects following waste-water treatment implementation at dongda Creek, Taichung City, Taiwan

Urbanization and industrialization have severely degraded Taiwan’s freshwater ecosystems, including dongda Creek in Taichung City, which received 10,000 tons of wastewater daily. To address this, a wastewater treatment plant (WWTP) was constructed by Tunghai University and the Taichung City Government, and its ecological impact was assessed through water quality and aquatic insect diversity monitoring before (2019–2020) and after implementation (2021–2023). Water quality measurements included temperature, ph, nutrients, and dissolved oxygen, while aquatic insect communities were analyzed using diversity indices and multivariate techniques. Results revealed significant water quality improvements, including reduced fat content and total nitrogen levels, although nitrate concentrations increased. The dominance of pollution-tolerant Chironomidae declined post-WWTP, while sensitive taxa such as Baetidae, Hydrophilidae, and Libellulidae emerged, accompanied by significant increases in diversity indices (taxa richness, Shannon, and Simpson). Seasonal patterns in insect diversity became more pronounced after the WWTP became operational. Environmental variables such as ph, temperature, and biochemical oxygen demand significantly influenced insect distributions, with Chironomidae thriving under poor conditions and sensitive taxa increasing as water quality improved. This study demonstrates that WWTP implementation can enhance water quality and foster ecological recovery, as evidenced by the recolonization of sensitive aquatic insect taxa and increased diversity. These findings highlight the effectiveness of restoration projects like WWTPs in improving ecosystem health and underscore the importance of ecological monitoring in freshwater management and conservation. dongda Creek serves as a valuable case study for guiding urban river restroration efforts in similar contexts.

Two new 16-hydroxybriaranes, 11-dehydroxybriavioid B (1) and 12-epi-briastecholide L (2), have been isolated from an algal-containing octocoral identified as Briareum stechei. The structures of metabolites 1 and 2 were corroborated by extensive 2d-NMR studies and comparison with literature data. Briarane 2 was found to reduce the release of inducible nitric oxide synthase (iNOS) to 87.92 %, at a concentration of 10 μM. © 2023 phytochemical Society of Europe
