Background An autoimmune disease that causes inflammation and bone and cartilage deterioration is rheumatoid arthritis (RA). In the pathogenesis of RA, oxidative stress and pro-inflammatory cytokines are important factors. The main polyphenol in black tea, theaflavins (TFs), has been used medically to treat a variety of inflammatory illnesses by reducing inflammation and reactive oxygen species (ROS) Objectives The medications available to treat RA have a variety of side effects. The current study was designed to assess the anti-arthritic properties of theaflavin in a mouse collagen-induced arthritis model. Methods In order to induce arthritis in dBA/1 mice, type II collagen was administered intradermally. From days 21 through days 42, different doses of theaflavin (50 and 100 mg/kg/day) were orally administered. To determine the effect of theaflavin on collagen-induced arthritis, histological analyses were conducted. In addition, the generation of reactive oxygen species (ROS), nitric oxide, and the activities of enzymatic antioxidants (superoxide dismutase, glutathione peroxidase, catalase, and glutathione reductase) in the joint homogenate of mice were examined. The levels of TNF-, IL-6, and IL-1β were also measured by ELISA to detect inflammation. Results Our results showed anti-oxidant and anti-inflammatory effects of theaflavin in arthritic mice. Histopathological studies corroborated the anti-arthritic properties of theaflavin. The compound was found to be effective in lowering ROS and nitric oxide levels while increasing enzymatic antioxidant levels. Theaflavin therapy also reduced TNF-, IL-6, and IL-1β levels. Conclusion In mice with arthritis, theaflavin was successful in reducing inflammation and oxidative stress. These results suggest that theaflavin may be used in conjunction with other treatments to manage RA. REFERENCES: NIL. In this study, we demonstrated a novel technique in selective absorption spectra (AS), on a transparent glass (GT) formed by self-assembled Nanoparticle array (SANP) and observed a narrow band absorption in the green wavelength region. The single layer deposition on a transparent glass is achieved by using the in-house developed self-assembly deposition technique. The monolayer retains the shape of hexagonal arrangement in between Nanoparticles and forms a regular array structure. Copyright © (2018) by Chemical and Biological Microsystems Society. Neuro-inflammation involves distinct alterations of microglial phenotypes, containing nocuous pro-inflammatory M1-phenotype and neuroprotective anti-inflammatory M-phenotype. Currently, there is no effective treatment for modulating such alterations. M1/M2 marker of primary microglia influenced by Melatonin were detected via qPCR. Functional activities were explored by western blotting, luciferase activity, EMSA, and ChIP assay. Structure interaction was assessed by molecular docking and LIGPLOT analysis. ER-stress detection was examined by ultrastructure TEM, calapin activity, and ERSE assay. The functional neurobehavioral evaluations were used for investigation of Melatonin on the neuroinflammation in vivo. Melatonin had targeted on Peroxisome Proliferator Activated Receptor delta (PPARδ) activity, boosted LPS-stimulated alterations in polarization from the M1 to the M2 phenotype, and thereby inhibited NFκB-IKKβ activation in primary microglia. The PPARδ agonist L-165,041 or over-expression of PPARδ plasmid (ov-PPARδ) showed similar results. Molecular docking screening, dynamic simulation approaches, and biological studies of Melatonin showed that the activated site was located at PPARδ (phospho-Thr256-PPARδ). Activated microglia had lowered PPARδ activity as well as the downstream SIRT1 formation via enhancing ER-stress. Melatonin, PPARδ agonist and ov-PPARδ all effectively reversed the above-mentioned effects. Melatonin blocked ER-stress by regulating calapin activity and expression in LPS-activated microglia. Additionally, Melatonin or L-165,041 ameliorated the neurobehavioral deficits in LPS-aggravated neuroinflammatory mice through blocking microglia activities, and also promoted phenotype changes to M2-predominant microglia. Melatonin suppressed neuro-inflammation in vitro and in vivo by tuning microglial activation through the ER-stress-dependent PPARδ/SIRT1 signaling cascade. Recently, extracting inherent biological system information (e.g. cellular networks) from genome-wide expression profiles for developing personalized diagnostic and therapeutic strategies has become increasingly important. However, accurately constructing single-sample networks (SINs) to capture individual characteristics and heterogeneity in disease remains challenging. Here, we propose a sample-specific-weighted correlation network (SWEET) method to model SINs by integrating the genome-wide sample-to-sample correlation (i.e. sample weights) with the differential network between perturbed and aggregate networks. For a group of samples, the genome-wide sample weights can be assessed without prior knowledge of intrinsic subpopulations to address the network edge number bias caused by sample size differences. Compared with the state-of-the-art SIN inference methods, the SWEET SINs in 16 cancers more likely fit the scale-free property, display higher overlap with the human interactomes and perform better in identifying three types of cancer-related genes. Moreover, integrating SWEET SINs with a network proximity measure facilitates characterizing individual features and therapy in diseases, such as somatic mutation, mut-driver and essential genes. Biological experiments further validated two candidate repurposable drugs, albendazole for head and neck squamous cell carcinoma (HNSCC) and lung adenocarcinoma (LUAd) and encorafenib for HNSCC. By applying SWEET, we also identified two possible LUAd subtypes that exhibit distinct clinical features and molecular mechanisms. Overall, the SWEET method complements current SIN inference and analysis methods and presents a view of biological systems at the network level to offer numerous clues for further investigation and clinical translation in network medicine and precision medicine.

Objectives:Many cancer biomarkers such as the circulating tumor cells/microemboli (CTCs/CTM) have been reported significant associations with clinical outcomes. However, different biomarkers have different sensitivities and specificities for cancer types and cohort patients, and synergistic effects between certain biomarkers have also been observed, leading to the inaccurate, fluctuating, and even controversial results when multiple biomarkers are analyzed together. In this paper, a novel combinational index, P-score, was developed for monitoring and predicting the disease condition of lung cancer patients during follow-up visits. Materials and methods:There were totally 13 return patients with 54 blood samples involved in this study to examine the number of CTC and CTM. Information from one group of 7 patients including 27 blood samples with published clinical data was employed to develop while those from another group of 4 patients containing 14 blood samples with unpublished clinical data were used to validate the P score in prediction. Enumerations were based on immunofluorescent staining images. distributions of CTC/CTM and their frequencies in stratified patients were carefully examined and analyzed the ROC curve and AUC value to develop the P score and P score-based prediction model. Results and conclusion:We found that the predictive power of P-score was not only comparable to the traditional cancer marker, in comparison with individual CTC/CTM, more false positives could be corrected by using P-score, thereby to improve the accuracy of analysis. From our preliminary validation tests, the prognosis and disease progression monitored longitudinally by P-score were further confirmed by clinical outcome data from physicians and its sensitivity was even better than those from individual biomarkers. We believe that this novel combinational indicator could be a promising tool to interpret clinical outcomes more accurately from multiple factors, particularly useful

The development of Internet of Things (IoT) makes the application of smart homes grow rapidly. It is very popular to install smart appliances in the house. However, building a smart control system at home not only costs a lot but also has many limitations. For this reason, this study proposes a smart homes control system to easily integrate IoT, WSN, smart robot and single-board computer to implement smart home applications. We use wireless technology and automatic equipment to avoid excessive communication cable and to make the house more intelligent to keep in-house movement unimpeded and indoor space tidy and to let appliance adjust appropriate environment settings automatically. This system brings intelligence and convenience to home, making living environment more comfortable.

The paper presents a brief introduction to big data and its role in healthcare applications. It is observed that the use of big data architecture and techniques are continuously assisting in managing the expeditious data growth in healthcare industry. Here, initially an empirical study is performed to analyze the role of big data in healthcare industry. It has been observed that significant work has been done using big data in healthcare sector. Nowadays, it is intricate to envision the way the machine learning and big data can influence the healthcare industries. It has been observed that most of the authors who implemented the use of machine learning and big data analytics in disease diagnosis have not given significant weightage to the privacy and security of the data. Here, a novel design of smart and secure healthcare information system using machine learning and advanced security mechanism has been proposed to handle big data of medical industry. The innovation lies in the incorporation of optimal storage and data security layer used to maintain security and privacy. different techniques like masking encryption, activity monitoring, granular access control, dynamic data encryption and end point validation have been incorporated. The proposed hybrid four layer healthcare model seems to be more effective disease diagnostic big data system.

Silicon detectors are an extraordinary piece of equipment that has become the cornerstone of research in modern high energy physics. They are becoming increasingly useful in medical research as well. LHCbs VELO detector is itself, a microstrip silicon vertex detector. It is the heart of LHCb spectrometer and plays a vital role in the reconstruction of particle tracks. The data gathered by VELO is used in LHCb studies of CP violation and heavy flavour physics. This work presents the studies on the calibration parameters of the VELO-LHCb. The inference from the analysis, with the use of probabilistic programming, is then used to create a machine learning based anomaly detection system in the space of VELO calibration.
