The public bus service, which is one of the most frequently used public transportation methods, is an indispensable part of public lifestyle. Hence, government and companies have integrated the public bus service with smart device applications, enabling the public to check information such as bus schedules and instant information at any time. Their aim is to improve the efficiency of the public bus service. Unfortunately, their efforts have faced a variety of problems, such as the distrust of the public regarding the provided information, traffic situation, and unexpected incidents which disturb the bus schedule. In order to overcome the mentioned problems, a smart bus stop signboard system will be designed. The smart signboard will show the current position of the bus and provides a reservation service, which the public may access using smart devices. The proposed system allows bus drivers to obtain information about a particular bus stop to determine whether they should drive to that bus stop. Because the traffic situation and the embarkment and disembarkment of passengers affects the arrival time of the bus, the proposed system will take the change in the number of bus passengers and travel time into account in order to predict the arrival time of the bus.

Hair cortisol concentration (HCC) is a valuable biomarker for evaluating chronic stress in preschoolers. However, few studies have explored early life HCC and its associated factors. This prospective cohort study analysed the HCC in children aged 6-48 months and its associations with parental HCC as well as positive and negative parental mental health outcomes. We used data from the ongoing Longitudinal Examination Across Prenatal and Postpartum Health in Taiwan (LEAPP-HIT) project, conducted in Taipei between 2020 and 2024. Hair samples were collected from both parents and children in 177 families (91 samples obtained during pregnancy and 86 during the postpartum period). The parents also completed self-reported questionnaires. Multiple linear regression was conducted to analyse the data. We observed a significant positive correlation between parents' and preschoolers' HCC. Furthermore, maternal depression (adjusted beta coefficient [aβ] = 0.09, 95% confidence interval [CI] = 0.02, 0.16) and perceived stress (aβ = 0.15, 95% CI = 0.02, 0.26) were positively associated with preschoolers' HCC. By contrast, higher maternal eudaimonia was associated with lower HCC in preschoolers (aβ = -0.11, 95% CI = -0.20, -0.01). For parents, maternal depression, anxiety, and perceived stress were independently associated with an increased HCC during the postnatal period, whereas maternal eudaimonia was negatively associated with HCC. Our results indicate that both mothers and fathers affect children's responses to stress. Assessment of cortisol stress hormone concentrations through hair samples can be a key means of detecting preschoolers' stress levels and enabling early intervention.

The molecular mechanisms contributing to the regulation of Th17-mediated inflammation remain underexplored. We here report a SUMO-specific protease (SENP)2-mediated pathway induced in pathogenic Th17 cells that restricts the pathogenesis of inflammatory colitis. SENP2 regulates the maturation of small ubiquitin-like modifiers (SUMO) and recycles SUMO from the substrate proteins. We find higher levels of SENP2 in pathogenic Th17 cells. By deleting Senp2 in T-cell lineages in mice, we demonstrate that the lack of Senp2 exacerbates the severity of experimental colitis, which is linked to elevated levels of GM-CSFIL-17A pathogenic Th17 cells and more severe dysbiosis of the intestinal microbiome. Adoptive transfer experiments demonstrate the cell-autonomous effect of Senp2 in restraining Th17 differentiation and colitis. The enzymatic activity of SENP2 is important for deSUMOylation of Smad4, which reduces Smad4 nuclear entry and Rorc expression. Our findings reveal a SENP2-mediated regulatory axis in the pathogenicity of Th17 cells.

A general method is presented to calculate from first principles the full set of third-order elastic constants of a material of arbitrary symmetry. The method here illustrated relies on a plane-wave density functional theory scheme to calculate the Cauchy stress and the numerical differentiation of the second Piola-Kirchhoff stress tensor to evaluate the elastic constants. It is shown that finite difference formulas lead to a cancellation of the finite basis set errors, whereas simple solutions are proposed to eliminate numerical errors arising from the use of Fourier interpolation techniques. Applications to diamond, silicon, aluminum, magnesium, graphene, and a graphane conformer give results in excellent agreement with both experiments and previous calculations based on fitting energy density curves, demonstrating both the accuracy and generality of our new methodology to investigate nonlinear elastic behaviors of materials.

This paper proposed a contact-force-controlled surface profiler by integrating the output of a LVT (Linear Velocity Transducer) in the inner-loop (to increase the damping) of the control system, so that not only the noise amplification effect due to the differentiator in the PId circuit, but the hysteresis effect of the force actuator could be avoided. A voice coil was applied as a force actuator, and a LVdT (Linear Variable differential Transformer) was applied at outer-loop to measure the surface profile. The maximum contact force between the probe and the sample was sensed by a load cell and limited to a tolerable value by the system, so the sample surface would not be destroyed by the probe. Four kinds of controllers using traditional PId, Ziegler-Nichols-based PId, fuzzy-PId, and neural-fuzzy design methods were applied for comparison. Besides, practical parameter variation (hysteresis effect) of force actuator was considered. Moreover, to investigate the system performance the testing input of the surface profile was generated by combining both FM (with frequency changing from 20 HZ to 20 KHZ according to the specification of voice coil) and AM signals. This kind of surface profile was very practical and not being found in the previous literature. By the results of simulations and practical testing, one can see the intelligent fuzzy-neural system was more robust to the parameter variation of force actuator.

Thanks to advances in wireless communication technologies, the wireless sensor network (WSNs) have been attracting a lot of attention from academic communities and successfully applied to various domains. Along with developments of the Industry 4.0, the WSNs start to play a vital role in the construction of smart factories and realization of intelligent manufacturing. Although, the industrial WSNs (IWSNs) presents great quantity of advantages, there still have some drawbacks to overcome such as challenges of the quality of data for IWSNs. In order to resolve the data missing problems in the context of IWSNs, the Last Observation Carried Forward method is adopted to estimate the missing value and reconstruct the sensing dataset which takes into account the temporal characteristics of sensing data in IWSNs. Through experiments, this method is proved to be an easy and effective measurement for missing value imputation of the large multi-dimensional sensing data achieved by the IWSNs.

A descriptive study of variable discretization and cost-sensitive logistic regression on imbalanced credit data

Training classification models on imbalanced data tends to result in bias towards the majority class. In this paper, we demonstrate how variable discretization and cost-sensitive logistic regression help mitigate this bias on an imbalanced credit scoring dataset, and further show the application of the variable discretization technique on the data from other domains, demonstrating its potential as a generic technique for classifying imbalanced data beyond credit socring. The performance measurements include ROC curves, Area under ROC Curve (AUC), Type I Error, Type II Error, accuracy, and F1 score. The results show that proper variable discretization and cost-sensitive logistic regression with the best class weights can reduce the model bias and/or variance. From the perspective of the algorithm, cost-sensitive logistic regression is beneficial for increasing the value of predictors even if they are not in their optimized forms while maintaining monotonicity. From the perspective of predictors, the variable discretization performs better than cost-sensitive logistic regression, provides more reasonable coefficient estimates for predictors which have nonlinear relationships against their empirical logit, and is robust to penalty weights on misclassifications of events and non-events determined by their apriori proportions. We present the weak lensing analysis of 279 COdEX clusters using imaging data from 4200 deg2 of the dECam Legacy Survey (dECaLS) data Release 3. The cluster sample results from a joint selection in X-ray, optical richness in the range 20 ≤ λ 200c) and the richness relation with the expression hM200c|λi ∝ M0 (λ/40)Fλ. By measuring the COdEX cluster sample as an individual cluster, we obtain the best-fit values, M0 = 3.24+−00..2927 × 1014M, and Fλ = 1.00+−00..2222 for the richness scaling index, consistent with a power law relation. Moreover, we separate the cluster sample into three richness groups;λ = 20 - 30, 30 - 50 and 50 - 110, and measure the stacked excess surface mass density profile in each group. The results show that both methods are consistent. In addition, we find an excellent agreement between our weak lensing based scaling relation and the relation obtained with dynamical masses estimated from cluster member velocity dispersions measured by the SdSS-IV/SPIdERS team. This suggests that the cluster dynamical equilibrium assumption involved in the dynamical mass estimates is statistically robust for a large sample of clusters. Copyright © 2019, The Authors. Epidermal growth factor receptor (EGFR) abnormalities relevant to tumor progression. A newly developed strategy for cancer therapy is induction of EGFR degradation. GMI, an immunomodulatory protein from the medicinal mushroom Ganoderma microsporum, exhibits anticancer activity. However, its role in the intracellular trafficking and degradation of EGFR remains unclear. In this study, we discovered that GMI inhibits the phosphorylation of multiple tyrosine kinases. Specifically, GMI was discovered to suppress lung cancer cells harboring both wild-type and mutant EGFR by inhibiting EGFR dimerization and eliminating EGFR-mediated signaling. Functional studies revealed that GMI binds to the extracellular segment of EGFR. GMI interacts with EGFR to induce phosphorylation of EGFR at tyrosine1045, which triggers clathrin-dependent endocytosis and degradation of EGFR. Furthermore, in the mouse models, GMI was discovered to suppress tumor growth. Knockdown of EGFR in lung cancer cells abolishes GMI's anticancer activity in vivo and in vitro. Our results reveal the interaction mechanisms through which GMI induces EGFR degradation and abolishes EGFR-mediated intracellular pathway. Our study indicates that GMI is an EGFR degrader for inhibiting EGFR-expressing tumor growth.

Eight previously unreported glycosides, designated gomphandranosides I-VIII, along with nine known compounds, were isolated from the roots of Gomphandra mollis Merr. Their structures were elucidated through comprehensive spectroscopic analyses, including NMR, HR-ESI-MS, Cd, dP4+ probability, and ECd calculations. The anti-inflammatory activities of the isolated compounds were assessed using nitric oxide inhibition assays in LPS-stimulated RAW264.7 macrophages. Compounds 5, 10, and 16 exhibited significant anti-inflammatory activity with IC values of 7.56 ± 0.9, 15.0 ± 1.2, and 14.2 ± 0.9 μM, respectively. Network pharmacology, gene ontology, Kyoto Encyclopedia of Genes and Genomes analysis, and molecular docking studies identified IL-6, TNF-α, MMP9, PTGS2, and HIF1A as key targets, with potentially involved in the NF-κB and IL-17 signaling pathways. These findings highlight G. mollis as a promising candidate for the treatment of inflammation-related diseases.
