BACKGROUNd:The patient population with stage III non-small-cell lung cancer (NSCLC) is heterogeneous, with varying staging characteristics and diverse treatment options. despite the potential practice-changing implications of randomized controlled trials evaluating the efficacy of perioperative epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs), concerns have been raised due to conflicting overall survival (OS) results. Few real-world studies have examined the survival outcomes of patients with resected EGFR-mutant stage III adenocarcinoma receiving perioperative chemotherapy and EGFR-TKIs. METHOdS:In this retrospective observational study, we enrolled patients with resected stage III adenocarcinoma with EGFR mutations between January 2011 and december 2021. Patients were classified into two groups: perioperative chemotherapy and perioperative EGFR-TKIs. Outcomes and prognostic factors were analyzed using Cox proportional hazards regression analysis. RESULTS:Eighty-four patients were enrolled in the analysis. Perioperative EGFR-TKIs led to longer progression-free survival (PFS) than chemotherapy (38.6 versus 14.2 months; p = 0.019). However, only pathological risk factors predicted poor PFS in multivariate analysis. Patients receiving perioperative chemotherapy had longer OS than those receiving EGFR-TKIs (111.3 versus 50.2 months; p = 0.052). Multivariate analysis identified perioperative treatment with EGFR-TKIs as an independent predictor of poor OS (HR: 3.76; 95% CI: 1.22-11.54). CONCLUSION:Our study demonstrates that chemotherapy should be considered in the perioperative setting for high-risk patients, when taking pathological risk factors into consideration, and that optimized sequencing of EGFR-TKIs might be the most critical determinant of OS.

Analysis of Factors that Influence the production of wetland rice in Banten Province

Banten Province has a harvested area of paddy fields on the 2017 year of 415,687 ha and production of 2,369,731 tons of milled dry grain (MdG). The objectives of this study are: 1) Knowing the condition of wetland rice farming in Banten Province, 2) Knowing the factors that influence the production of wetland rice in Banten Province, 3) Knowing the efficiency of the use of production inputs. The sampling method used purposive sampling with 123 respondents. The analytical method used multiple linear of Cobb douglas production functions and descriptive analysis. The results of the study are: 1) Rice paddy farming in Rainy Season (RS) 2017/2018 has a B/C ratio of 1.7. The productivity of paddy rice is 5.91 tons harvested dry grain (HdG)/ha. 2) Significant factors affecting the production of paddy rice in RS 2017/2018 are The amount of use of solid organic fertilizer, The amount of solid herbicide use, The amount of use of human labor rent, and the arable land area, 3). The cumulative elasticity is 0.97, is inelastic, meaning that the addition of 1% of the production factor will increase production by 0.97% or decreasing return to scale. So rice farming is relatively inefficient.

Activation of mitogen-activated protein kinase (MAPK) and PI3K signaling confers resistance against sorafenib, a mainstay treatment for advanced hepatocellular carcinoma (HCC). Antrocin and ovatodiolide constitute as the most potent secondary metabolites isolated from Antrodia camphorata and Anisomeles indica, respectively. Both natural compounds have recently gained a lot of attention due to their putative inhibition of MAPK and PI3K signaling in various solid cancers. However, whether their combination is effective in HCC remains unknown. Here, we investigated their effect, alone or in various combinations, on MAPK and PI3K signaling pathways in HCC cells. An array of in vitro study were used to investigate anticancer and stemness effects to treat HCC, such as cytotoxicity, drug combination index, migration, invasion, colony formation, and tumor sphere formation. drug effect in vivo was evaluated using mouse xenograft models. In this study, antrocin and ovatodiolide synergistically inhibited the SNU387, Hep3B, Mahlavu, and Huh7 cell lines. Sequential combination treatment of Huh7 and Mahlavu with ovatodiolide followed by antrocin resulted stronger cytotoxic effect than did treatment with antrocin followed by ovatodiolide, their simultaneous administration, antrocin alone, or ovatodiolide alone. In the Huh7 and Mahlavu cell lines, ovatodiolide→antrocin significantly suppressed colony formation and proliferation as well as markedly downregulated ERK1/2, Akt, and mTOR expression. Inhibition of ERK1/2 and Akt/mTOR signaling by ovatodiolide→antrocin suppressed ribosomal biogenesis, autophagy, and cancer stem cell-like phenotypes and promoted apoptosis in Huh7 and Mahlavu cells. The sorafenib-resistant clone of Huh7 was effectively inhibited by synergistic combination of both compound in vitro. Passive interest turbulent drag reduction, effective means to improve air vehicle fuel consumption costs. Most turbulent problems happening to the nature and engineering applications were exactly the turbulence problem frequently caused by one or more turbulent shear flows. This study was operated with incompressible 3-d channels with cyclic wavy boundary to explore the physical properties of turbulence flow. This research measures the distribution of average velocity, instant flowing field shapes, turbulence and pressure distribution, etc. Furthermore, the systematic computation and analysis for the 3-d flow field was also implemented. It was aimed to clearly understand the turbulence fields formed by wavy boundary of tube flow. The purpose of this research is to obtain systematic structural information about the turbulent flow field and features of the turbulence structure are discussed.

Assessing the effect of different natural dissolved organic matters on the cytotoxicity of titanium dioxide nanoparticles with bacteria

Titanium dioxide nanoparticles （TiO2 NPs） are among the most widely manufactured nanomaterials on a global scale. However, prudent and vigilant surveillance, incumbent upon the scientific community with the advent of new technologies, has revealed potentially undesirable effects of TiO2 NPs on biological systems and the natural environment during their application and discharge. Such effects are likely best evaluated by first assessing the fate of the TiO2 NPs in natural environments. In this study, the effects of terrestrial humic acid （HA） and tannic acid （TA）, two major members of the collective： dissolved organic matter （dOM）, on the cytotoxicity of TiO2 NPs to Escherichia coli were investigated in the presence and absence of natural sunlight. Qualitative （transmission electron microscopy （TEM）） and quantitative （LCso） analyses were employed in this study. In addition, the production of reactive oxygen species （ROS） in the form of .OH was further assessed-as HA or TA increased the production of ROS decreased. The inhibition of bacterial viability in the light treatment groups, with respective treatment organics at concentrations of 10 ppm, was less in TA than in terrestrial HA. SAS was used to analyze the treatment effect of individual factors of light irradiation, dOM, and concentration of TiO2 NPs.

Understanding of elementary school teachers on physical concepts

To avoid errors in understanding the concept of physics sustainably, misconception of physics needs to be diagnosed, both students and teachers in elementary or secondary education. However, this diagnosis is still rarely done in teachers at elementary school. This research is to describe and identify misconceptions in understanding on physical concept in elementary school teachers. This is a qualitative research. The respondent are 14 elementary school teachers in Surakarta, with stratified purposive sampling. Collecting data with test and interview, with open response essay test and an interview. The process of analysis through data collection, data reduction, and withdrawal of conclusions, done inductively. The results: 14,54% teachers have a scientific understanding of the concept, misconception category of 80,12%, and 4,17% not understand category. Misconceptions absolutely occur in the understanding of the time object to arrive at the ground. Misconceptions are categorized high on the concept of 1) the force of gravity (92,86%), 2) light velocity (92,86%), 3) frequency and amplitude (85,71%), 4) static electric force 78,57%), and 5) visible object by eye (57,15%). The conclusion that the understanding of elementary school teachers on the concept of physical science shows different levels and varies.

Myrosinase (ß-thioglucosidase glucohydrolase (EC 3.2.3.1)) is a glycoprotein responsible for the hydrolysis of glucosinolates, a group of sulfur-containing secondary metabolites present in Brassicaceae plants. Sulforaphane, a powerful anti-cancer compound, comes from the hydrolysis of glucoraphanin, the main glucosinolate found in broccoli. The aim of this work was to investigate the interaction between broccoli myrosinase and different substrates through molecular modelling. Currently the amino acid sequence and the molecular structure of broccoli myrosinase are unknown. The amino acid sequence was deduced from the nucleotide sequence, which in turn was obtained experimentally. The primary sequence of broccoli myrosinase used for modelling consisted of 548 amino acids. The three-dimensional model was generated by an ab initio/threading method using the crystal structure of 1MYR_A and 1dWF_M of Sinapis alba, 1CBG_A of Trifolium repens L, 3PTK_A of Oryza sativa and 3WQ4_A of Camellia sinensis as templates, employing the I-TASSER server. The interaction with glucoraphanin (GRA), sinigrin (SIN), glucoiberin (GIB) and N-acetylglucosamine (NAG) was investigated through molecular docking. The binding energy of GRA, SIN, GIB and NAG to myrosinase were -3.80;-4.71;-4.77 and -5.55 (Kcal/mol), respectively. The simulations of the interaction between GRA and myrosinase suggested that the residues of the active site responsible for substrate stabilization by hydrogen bonding were Gln207, Gly431, Ser433 and Trp 477. The residue involved in the hydrolysis of the ß-d-thioglucose binding was Tyr352. NAG interacts with the active site, as inhibitor. Our results will allow further studies about molecular mechanisms of broccoli myrosinase. Copyright © 2018, AIdIC Servizi S.r.l.
