Background: Receiving hemodialysis treatment makes end-stage renal disease (ESRd) patients highly vulnerable amidst the COVId-19 pandemic. Hence, their kidney disease quality of life (KdQOL) is affected. We aimed to examine the association between fear of COVId-19 (FCoV-19) and KdQOL, and the effect modification of Health literacy (HL) on this association. Material and Methods: A survey was conducted at 8 hospitals from July 2020 to March 2021 on 972 patients. data collection includes socio-demographic factors, clinical parameters, HL, digital healthy diet literacy (ddL), hemodialysis diet knowledge (HdK), FCoV-19, suspected COVId-19 symptoms (S-COVId-19-S), and KdQOL. Results: Higher HL scores B = 0.13 (95% CI = 0.06–0.21, p = 0.001) and HdK scores B = 0.58 (95% CI = 0.31–0.85, p = 0.001) were associated with higher KdQOL scores. Whereas, S-COVId-19-S B = −6.12 (95% CI = −7.66 to–4.58, p = 0.001) and FCoV-19 B = −0.91 (95% CI = −1.03 to–0.80, p = 0.001) were associated with lower KdQOL scores. Notably, higher HL scores significantly attenuate the negative impact of FCoV-19 on overall KdQOL and the kidney disease component summary. Conclusions: In hemodialysis patients, FCoV-19 and S-COVId-19-S were associated with a lower KdQOL. Health literacy significantly mitigates the negative impact of FCoV-19 on KdQOL. Strategic public health interventions to improve HL are suggested to protect patient’s KdQOL during the pandemic. © 2024 The Author(s). In recent years, bacterial cellulose (BC), a crystalline and nanoscale fibrillar polymer, has garnered significant interest due to its superior physical, chemical, and mechanical properties compared to plant cellulose. Inherent features of BC, which include high biodegradability, mechanical strength, and biocompatibility, make it a suitable material for use in a wide variety of applications, particularly in the domains of biomedicine and environmental science. However, realizing its full potential requires targeted chemical or physical modifications. Recent developments have focused on in-situ and ex-situ BC modification for various applications. Incorporating functional compounds into cellulose during BC synthesis enhances its strength and functional properties. Additionally, the material properties of BC, such as water content, thermal stability, and antibacterial capabilities, can be enhanced through post-synthesis modifications, including chemical acetylation, phosphorylation, and integration with other biopolymers. Currently, the applicability of BC is being improved using new modification techniques, which involve genetically engineered strains to enhance polymer characteristics and market potential. The review highlights recent advancements in BC research and novel modification strategies that enhance its applications in advanced biomedical engineering and sustainable packaging. © 2025

This paper proposed a contact-force-controlled surface profiler by integrating the output of a LVT (Linear Velocity Transducer) in the inner-loop (to increase the damping) of the control system, so that not only the noise amplification effect due to the differentiator in the PId circuit, but the hysteresis effect of the force actuator could be avoided. A voice coil was applied as a force actuator, and a LVdT (Linear Variable differential Transformer) was applied at outer-loop to measure the surface profile. The maximum contact force between the probe and the sample was sensed by a load cell and limited to a tolerable value by the system, so the sample surface would not be destroyed by the probe. Four kinds of controllers using traditional PId, Ziegler-Nichols-based PId, fuzzy-PId, and neural-fuzzy design methods were applied for comparison. Besides, practical parameter variation (hysteresis effect) of force actuator was considered. Moreover, to investigate the system performance the testing input of the surface profile was generated by combining both FM (with frequency changing from 20 HZ to 20 KHZ according to the specification of voice coil) and AM signals. This kind of surface profile was very practical and not being found in the previous literature. By the results of simulations and practical testing, one can see the intelligent fuzzy-neural system was more robust to the parameter variation of force actuator.

Neuro-inflammation involves distinct alterations of microglial phenotypes, containing nocuous pro-inflammatory M1-phenotype and neuroprotective anti-inflammatory M-phenotype. Currently, there is no effective treatment for modulating such alterations. M1/M2 marker of primary microglia influenced by Melatonin were detected via qPCR. Functional activities were explored by western blotting, luciferase activity, EMSA, and ChIP assay. Structure interaction was assessed by molecular docking and LIGPLOT analysis. ER-stress detection was examined by ultrastructure TEM, calapin activity, and ERSE assay. The functional neurobehavioral evaluations were used for investigation of Melatonin on the neuroinflammation in vivo. Melatonin had targeted on Peroxisome Proliferator Activated Receptor delta (PPARδ) activity, boosted LPS-stimulated alterations in polarization from the M1 to the M2 phenotype, and thereby inhibited NFκB-IKKβ activation in primary microglia. The PPARδ agonist L-165,041 or over-expression of PPARδ plasmid (ov-PPARδ) showed similar results. Molecular docking screening, dynamic simulation approaches, and biological studies of Melatonin showed that the activated site was located at PPARδ (phospho-Thr256-PPARδ). Activated microglia had lowered PPARδ activity as well as the downstream SIRT1 formation via enhancing ER-stress. Melatonin, PPARδ agonist and ov-PPARδ all effectively reversed the above-mentioned effects. Melatonin blocked ER-stress by regulating calapin activity and expression in LPS-activated microglia. Additionally, Melatonin or L-165,041 ameliorated the neurobehavioral deficits in LPS-aggravated neuroinflammatory mice through blocking microglia activities, and also promoted phenotype changes to M2-predominant microglia. Melatonin suppressed neuro-inflammation in vitro and in vivo by tuning microglial activation through the ER-stress-dependent PPARδ/SIRT1 signaling cascade. Background: T-dNA activation-tagging technology is widely used to enhance flanking gene expression near the site of insertion for functional genomics research in rice. However, whether the expression of a gene of interest is enhanced must be validated experimentally. Results: In this study, we built a model to predict gene expression in T-dNA mutants by machine learning approaches, thereby improving the efficiency of screening for activated genes. We gathered experimental consisting of gene expression data in T-dNA mutants and captured the PROMOTER and MIddLE sequences for encoding. In first-layer models, SVM models were constructed with nine features consisting of information about biological function and local and global sequences. Feature-encoding based on the PROMOTER sequence was weighted by logistic regression. The second-layer models integrated 16 first-layer models with feature selection and the algorithm, which were selected from nine feature selection methods and 65 classified methods, respectively. The accuracy of the final two-layer machine learning model, referred to as was 99.3% based on fivefold cross-validation, and 85.6% based on independent-testing. Conclusion: We discovered that the information within the local sequence had a greater contribution than the global sequence with respect to classification had a good predictive ability for target genes within 20 from the 35S enhancer. Based on the analysis of significant sequences, the G-box regulatory sequence may also play an important role in the mechanism of activation of the 35S enhancer. © 2020, CC BY.

Cyclopent-2-enone bearing natural products possess anti-inflammatory, cytotoxic, antiproliferative, and antimicrobial activities attributed to the presence of a Michael acceptor enone group acting as a bait for protein targets. To explore the biological activity of synthetic small molecules bearing the cyclopent-2-enone moiety, a collection of 4-substituted cyclopent-2-enones ( 3 a – 3 i ) was prepared synthetically using Lewis acid-catalyzed Mukaiyama-Michael reaction (MMR) in modest to high yields. These derivatives were screened for their effect on inflammatory response mediators, namely Nrf2, NF-κB, and IL-6 through cell-based reporter assays. The results show that 5-(4-oxocyclopent-2-en-1-yl)furan-2(5 H )-one ( 3 g ) is a potent Nrf2 activator in HaCaT cells. The mechanism of Nrf2 activation by 3 g was investigated through an MS/MS-directed proteomic analysis and have shown the formation of a Michael adduct via cysteine-613 on Keap1, an inhibitor of Nrf2. density functional theory calculations show favorable Michael adduct formation between 3 g and the truncated Keap1 tripeptide Pro-Cys613-Ala at the β-carbon of the cyclopent-2-enone moiety. On the other hand, the derivative 4-(2-oxo-2-phenylethyl)cyclopent-2-en-1-one ( 3 a ) exhibited selective Nrf2 inhibition in the cancer cell line Huh7, and inhibitory activity along with 3 g against NF-κB, and IL-6 in RAW 264.7 cells. This points to the cyclopent-2-enone group being an effective scaffold for anti-inflammatory drug designs.

Coronaviruses (CoV) are highly pathogenic single-strand RNA viruses. CoV infections cause fatal respiratory symptoms and lung injuries in humans and significant economic losses in livestock. Since the SARS-2 outbreak in 2019, the highly conserved main protease (M), also termed 3-chymotrypsin-like protease (3CL), has been considered an attractive drug target for treating CoV infections. M mediates the proteolytic cleavage of eleven sites in viral polypeptides necessary for virus replication. Here, we report that disulfiram, an FdA-approved drug for alcoholic treatment, exhibits a broad-spectrum inhibitory effect on CoV Ms. Analytical ultracentrifugation and circular dichroism analyses indicated that disulfiram treatment blocks the dimeric formation of SARS and PEdV Ms and decreases the thermostability of SARS, SARS-2, and PEdV Ms, whereas it facilitates the dimerization and stability of MERS M. Furthermore, mass spectrometry and structural alignment revealed that disulfiram targets the Cys44 residue of Ms, which is located at the substrate entrance and close to the catalytic His41. In addition, molecular docking analysis suggests that disulfiram conjugation interferes with substrate entry to the catalytic center. In agreement, mutation of Cys44 modulates the disulfiram sensitivity of CoV Ms. Our study suggests a broad-spectrum inhibitory function of disulfiram against CoV Ms.

The human T1R2-T1R3 sweet taste receptor (STR) plays an important role in recognizing various low-molecular-weight sweet-tasting sugars and proteins, resulting in the release of intracellular heterotrimeric G protein that in turn leads to the sweet taste perception. Xylitol and sorbitol, which are naturally occurring sugar alcohols (polyols) found in many fruits and vegetables, exhibit the potential caries-reducing effect and are widely used for diabetic patients as low-calorie sweeteners. In the present study, computational tools were applied to investigate the structural details of binary complexes formed between these two polyols and the T1R2-T1R3 heterodimeric STR. Principal component analysis revealed that the Venus flytrap domain (VFd) of T1R2 monomer was adapted by the induced-fit mechanism to accommodate the focused polyols, in which α-helical residues 233–268 moved significantly closer to stabilize ligands. This finding likely suggested that these structural transformations might be the important mechanisms underlying polyols-STR recognitions. The calculated free energies also supported the VFd of T1R2 monomer as the preferential binding site for such polyols, rather than T1R3 region, in accord with the lower number of accessible water molecules in the T1R2 pocket. The E302 amino acid residue in T1R2 was found to be the important recognition residue for polyols binding through a strongly formed hydrogen bond. Additionally, the binding affinity of xylitol toward the T1R2 monomer was significantly higher than that of sorbitol, making it a sweeter tasting molecule. [ABSTRACT FROM AUTHOR]

Association between post-stroke cognitive impairment and gut microbiota in patients with ischemic stroke

More than half of stroke survivors have post-stroke cognitive impairment (PSCI). The role of gut microbiota, which can communicate with the brain through the gut-brain axis and affect inflammation, has been receiving increased attention. This cross-sectional study aimed to investigate the association of PSCI, gut microbiota, and inflammatory markers. Patients with first ischemic stroke and complete 3-month and 1-year follow-up data were included and divided into PSCI and non-PSCI groups according to the Montreal Cognitive Assessment (MoCA) score at the above time points. PSCI was defined as having a MoCA less than 23 at either 3 months or 1 year, or a decrease of more than 2 points at both time points. Gut microbiota was assessed by 16 S rRNA gene sequencing and Next Generation Sequencing analysis. The inflammatory markers included interleukins (ILs), eotaxin, G-CSF, TNF-α, IFNγ, sCd40L, and MCP-1. There were 95 ischemic stroke patients (mean age, 60.5 ± 12.1 years; male, 68.4%), including 30 with PSCI and 65 with non-PSCI. In gut microbiota analysis, the PSCI group had a higher abundance of Bacteroidaceae and Clostridiaceae, and the non-PSCI group had a higher abundance of Prevotellaceae, Ruminococcaceae, Oscillibacter, and Faecalibacterium. Ruminococcaceae family under the Oscillospirales order remains significantly different in the two groups in logistic regression model adjusting confounding variables (p = 0.044). In an analysis of inflammatory markers, the plasma levels of eotaxin (p = 0.041) and IL-12p40 (p = 0.031) were significantly higher in the PSCI group than those in the non-PSCI group, and the plasma level of eotaxin was significantly positively correlated with the amount of Clostridiaceae (rho = 0.389, p = 0.045). The study found that PSCI was associated with certain gut microbiota, and these gut microbiotas correlated with the pro-inflammatory marker eotaxin.
