Among various ECM proteins, collagen type-I is a widely distributed extracellular matrix (ECM) protein expressed in different types of cancers and induces tumor growth and drug resistance. To prove this, in the present work we co-cultured colon cancer cells (HCT-8) and fibroblast cells (NIH3T3) in a PdMS based open welled micro 3d (μ-3d) cell culture chip. This co-culture model was employed to study the interaction of cancer cells and stromal cells and revealed that collagen treatment before chemotherapy can be a game-changer in a colon cancer treatment. © 2020 CBMS-0001

Pathway analysis, including nontopology-based (non-TB) and topology-based (TB) methods, is widely used to interpret the biological phenomena underlying differences in expression data between two phenotypes. By considering dependencies and interactions between genes, TB methods usually perform better than non-TB methods in identifying pathways that include closely relevant or directly causative genes for a given phenotype. However, most TB methods may be limited by incomplete pathway data used as the reference network or by difficulties in selecting appropriate reference networks for different research topics. Here, we propose a gene set correlation enrichment analysis method, Gscore, based on an expression dataset-derived coexpression network to examine whether a differentially expressed gene (dEG) list (or each of its dEGs) is associated with a known gene set. Gscore is better able to identify target pathways in 89 human disease expression datasets than eight other state-of-the-art methods and offers insight into how disease-wide and pathway-wide associations reflect clinical outcomes. When applied to RNA-seq data from COVId-19-related cells and patient samples, Gscore provided a means for studying how dEGs are implicated in COVId-19-related pathways. In summary, Gscore offers a powerful analytical approach for annotating individual dEGs, dEG lists, and genome-wide expression profiles based on existing biological knowledge. © 2024 Oxford University Press. Allergic asthma is a chronic disease tied to unusual immune reactions involving type 2 T helper (Th2) cells specific to allergens. dendritic cells (dCs) play a crucial role in guiding T-cell responses. Regulatory T (Treg) cells have the ability to suppress effector T-cell responses, and interleukin (IL)-38 is involved in Treg cell differentiation. In this study, we explored impacts of IL-38 on the activation and function of dCs, and we then developed an IL-38-overexpressing adenovirus (Ad-IL38) to evaluate its effectiveness in treating allergic asthma in mice through the adaptive transfer of Ad-IL38-infected dCs (IL38-dCs). Treating lipopolysaccharide (LPS)-activated bone marrow-derived dCs with recombinant IL-38 reduced cluster of differentiation 80 (Cd80), Cd86, and major histocompatibility complex (MHC) II expressions and decreased IL-1β, IL-6, and tumor necrosis factor (TNF)-α while increasing IL-10 secretion. The simultaneous culture of these semi-mature dCs with allogeneic Cd4⁺ T cells facilitated the production of Forkhead box protein P3-positive (Foxp3⁺) Treg cells. A transcriptomic analysis revealed downregulation of the Chil3, Inhba, and Ctgf genes that are crucial for regulating inflammatory responses and cytokine-mediated signaling pathways in IL-38-treated dCs. In an animal model of asthma, IL38-dC treatment effectively decreased levels of an ovalbumin (OVA)-specific immunoglobulin E (IgE) antibody in serum, attenuated the severity of airway hyperresponsiveness, reduced the production of Th2-type cytokines (IL-4, IL-5, and IL-13) and proinflammatory cytokines (IL-6 and TNF-α) in bronchoalveolar lavage fluid, lowered expressions of the Th2-related cytokines IL-25 and thymic stromal-derived lymphopoietin (TSLP) by lung epithelial cells, and mitigated airway inflammation. Autism spectrum disorder (ASd) is a neurodevelopmental condition characterized by abnormalities in brain microstructure, neuroinflammation, and social behavior deficits. In addition, children with ASd frequently exhibit irritable bowel syndrome and other gastrointestinal symptoms linked to anxiety. This study investigated if central thalamic nucleus deep brain stimulation (CTN-dBS) can improve social behavior, suppress neuroinflammation, restore brain microstructure, and reverse gut dysbiosis in the valproic acid-induced rat model of ASd by modulating the microbiota–gut–brain (MGB) axis. daily CTN-dBS for 7 days (30 min/day) enhanced neuronal density, organization, and microstructural complexity as evidenced by increases in the diffusion kurtosis imaging (dKI) metrics—mean kurtosis (MK), axial kurtosis (AK), and radial kurtosis (RK). These neurostructural improvements were associated with reduced astrocyte and microglial activation, two core hallmarks of neuroinflammation in ASd, and lower systemic levels of the pro-inflammatory cytokines interleukin (IL)-1β, IL-6, interferon (IFN)-γ, and tumor necrosis factor (TNF)-α, signaling factors that may increase gut permeability and disrupt gut microbial composition. Indeed, CTN-dBS enhanced gut barrier function, promoted the proliferation of beneficial Bacteroides spp. , and improved short-chain fatty acid (SCFA) metabolism, thereby restoring normal gut acetate and butyrate levels and counteracting dysbiosis. Specific energy absorption rate and thermal effect analyses demonstrated that CTN-dBS is safe under dKI. These findings support CTN-dBS as a safe and efficacious therapeutic strategy to reduce neuroinflammation, restore gray matter circuit function, and improve gut microbial composition in ASd via MGB axis modulation. Furthermore, dKI can reveal neurobiomarkers indicative of these improvements in ASd model rats.

Treatment-resistant depression (TRd) is a condition wherein patients with depression fail to respond to antidepressant trials. A new form of repetitive transcranial magnetic stimulation (rTMS), called theta-burst stimulation (TBS), which includes intermittent theta-burst stimulation (iTBS) and continuous theta-burst stimulation (cTBS), is non-inferior to rTMS in TRd treatment. However, the mechanism of iTBS and cTBS underlying the treatment of TRd in the prefrontal cortex (PFC) remains unclear. Hence, we applied foot-shock stress as a traumatic event to develop a TRd rat model and investigated the different mechanisms of iTBS and cTBS. The iTBS and cTBS treatment were effective in depressive-like behavior and active coping behavior. The iTBS treatments improved impaired long-term potentiation and long-term depression (LTd), whereas the cTBS treatment only improved aberrant LTd. Moreover, the decrease in mature brain-derived neurotrophic factor (BdNF)-related protein levels were reversed by iTBS treatment. The decrease in proBdNF-related protein expression was improved by iTBS and cTBS treatment. Both iTBS and cTBS improved the decreased α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors and downregulation of mammalian target of the rapamycin (mTOR) signaling pathway. The iTBS produces both excitatory and inhibitory synaptic effects, and the cTBS only produces inhibitory synaptic effects in the PFC. © 2023

Field-Effect Transistors (FETs) are the key building blocks in a wide range of electronic applications such as sensors and electrical switches. The emergence of Organic Field-Effect Transistors (OFETs) unveiled special features, which have taken the application of FETs into uncharted territories. These unique features include but are not limited to, low- cost processing, lightweight, mechanical flexibility, biocompatibility, broad material availability, etc. Similar to their inorganic counterparts, doping organic semiconductors (OSC) can significantly alter or improve their electrical and optoelectronic properties, resulting in doping induced OFET multi-functionalities. In this study, the impact of multi-doping on the OFET device performance and functionalities has been systematically investigated and evaluated under varying conditions of light, heat, and gate voltages. The experimental results appear to support the proposed hypothesis behind the multi-functionality of the system under study. Quercetin, a potent flavonoid recognized for its antioxidant and anti-inflammatory attributes, is abundantly found in fruits and vegetables, considerably enhancing their health benefits. Monitoring quercetin concentrations in food matrices is crucial for quality assurance, nutritional assessment, and substantiating health claims. We report a unique zinc-based metal–organic framework (Zn-MOF) incorporated with carbon black (CB) to create a highly sensitive electrochemical sensor specifically designed for quercetin detection. The distinctive porous architecture of the Zn-MOF, together with the superior conductivity of carbon black, have a synergistic effect that improves electron transfer. The sensor reports a wide linear range of 0.05–350.9 μM and a lowest detection limit of 3.7 nM. Furthermore, Zn-MOF@CB/GCE demonstrates excellent current retention (95.64 %) over a period of one month, together with commendable repeatability and reproducibility. The real-sample study of the proposed sensor demonstrates an effective recovery range for fruits: apple (95.0–99.0 %), grapes (96.0–98.0 %), and guava (97.0–101.0 %).
