Plastic pollution is a widespread issue that poses a threat to agroecosystems. Recent data on microplastic (MP) pollution from compost and its application to soil have highlighted the potential impact of micropollutants that may be transferred from compost. Thus, we aim with this review to elucidate the distribution-occurrence, characterization, fate/transport, and potential risk of MPs from organic compost to gain comprehensive knowledge and mitigate the adverse impacts of compost application. The concentration of MPs in compost was up to thousands of items/kg. Among micropollutants, fibers, fragments, and films are the most common, with small MPs having a higher potential to absorb other pollutants and cause harm to organisms. Various synthetic polymers, including polyethylene (PE), polypropylene (PP), polyethylene terephthalate (PET), polystyrene (PS), polyvinyl chloride (PVC), polyester (PES), and acrylic polymers (AP), have been widely used of plastic items. MPs are emerging pollutants that can have diverse effects on soil ecosystems, as they can transfer potential pollutants from MPs to compost and then to the soil. Following the microbial degradation scheme, the transfer chain from plastics to compost to soil can be broken down into main stages, i.e., colonization - (bio)fragmentation - assimilation - and mineralization. Microorganisms and adding biochar play an essential role during composting, which can be an effective solution to enhance MP degradation. Findings have shown that stimulating free radical generation could promote the biodegradation efficacy of MPs and possibly remove their occurrence in compost, thereby reducing their contribution to ecosystem pollution. Furthermore, future recommendations were discussed to reduce ecosystem risks and health challenges.

We propose an ultrasensitive and cost-effective novel fluorometric approach for effective detection of cysteamine/cancer marker. The synthesis of the 2d nanomaterial is accomplished via the solvothermal process using the ultrasound system which is termed as the two dimensional solvothermal synthesis of ultrasound (2d-STSUS). 2d Hexagonal boron nitride (hBN) has emerged as a promising material for sensing applications due to its exceptional properties like broad bandgap and high thermal stability makes it ideal for application in a wide range of sensors and biosensors. Since hBN cannot effectively absorb or emit light because of broad band gap, it cannot produce fluorescence. On the contrary, using W-doped hexagonal boron nitride nanosheets (hBNW), we were able to successfully increase the method's selectivity, sensitivity, and easy usage by fusing the unique properties of W metal with hBN. For the first time, hBNW nanosheets exhibited a clear blue color transition and were effectively employed to detect varied levels of cysteamine in human urine and aqueous samples. When combined with hBNW nanosheets, cysteamine has excellent outcomes among other drugs and neurotransmitters. The linearity value R 2 = 0.9991 for aqueous was observed in the range of 1 nM to 28 nM and the detection limit was 0.0936 nM under ideal conditions. Additionally, quantification tests were conducted on human serum and urine samples for real sample analysis, yielding R 2 values of 0.9961 and 0.9958, respectively.

Cordyceps militaris (C. militaris) is a unique medicinal fungus of the genus Cordyceps. It has high economic value due to various biological functions from the extracellular polysaccharides (EPSs) it produces. The aim of this study was to establish a mycelium-immobilization system for EPS production in a bubble column system. First, EPS production was optimized by 90.2% (from 0.72 to 1.37 g/L) using response surface methodology under an alginate-immobilized mycelium system with 6 days of cultivation. The highest EPS production was obtained (5.3 g/L) in a flask system through elongation to 15 days of cultivation. EPS production subsequently further increased to 10.42 g/L utilizing a bubble column reactor. Furthermore, the system can be reused for at least three rounds. In the EPS characteristics analysis, the produced EPSs were mainly composed of glucose and galactose. They presented strong antibacterial activity against Staphylococcus aureus. In terms of antioxidant activity, respective the EPS concentration that caused 50% scavenging of dPph and ABTS free radicals values (IC50) were 4.65 and 7.92 mg/mL. In summary, these findings should be helpful for establishing a suitable industrial fermentation system for C. militaris and can be studied in the food and medical industries in future work. © 2024 The Authors

As hydrogen is touted as a key player in the decarbonization of modern society, it is critical to enable quantitative H analysis at high spatial resolution if possible at the atomic scale. Indeed, H has a known deleterious impact on the mechanical properties (strength, ductility, toughness) of most materials that can hinder their use as part of the infrastructure of a hydrogen-based economy. Enabling H mapping, including local hydrogen concentration analyses at specific microstructural features, is essential for understanding the multiple ways that H affect the properties of materials, including for instance embrittlement mechanisms and their synergies, but also spatial mapping and quantification of hydrogen isotopes is essential to accurately predict tritium inventory of future fusion power plants, ensuring their safe and efficient operation for example. Atom probe tomography (APT) has the intrinsic capabilities for detecting hydrogen (H), and deuterium (d), and in principle the capacity for performing quantitative mapping of H within a material s microstructure. Yet the accuracy and precision of H analysis by APT remain affected by the influence of residual hydrogen from the ultra-high vacuum chamber that can obscure the signal of H from within the material, along with a complex field evaporation behavior. The present article reports the essence of discussions at a focused workshop held at the Max-Planck Institute for Sustainable Materials in April 2024. The workshop was organized to pave the way to establishing best practices in reporting APT data for the analysis of H. We first summarize the key aspects of the intricacies of H analysis by APT and propose a path for better reporting of the relevant data to support interpretation of APT-based H analysis in materials. Air pollution has been linked to respiratory diseases, and urban air pollution can be attributed to a number of emission sources. The emitted particles and gases are the primary components of air pollution that enter the lungs during respiration. Particulate matter with an aerodynamic diameter of ≤ 2.5 µm (PM) can deposit deep into the respiratory tract via inhalation and has been proposed as a causative agent for adverse respiratory health. In addition, the lung contains a diverse microbial community (microbiome) that maintains normal homeostasis and is significantly altered in a variety of pulmonary disorders. Air pollution, specifically PM, has previously been shown to significantly alter the composition of the lower airway microbiome, which has been linked to decreased lung function in chronic obstructive pulmonary disease (COPd) patients. Surprisingly, the intestinal microbiome has also been implicated in the modulation of pulmonary inflammatory diseases. Therefore, dysbiosis of the lung and intestinal microbiomes pose significant negative effects on human health. This dataset describes the microbial community profiles of the lungs and intestines of ageing rats exposed to ambient unconcentrated traffic-related air pollution for three months. The whole-body exposure system was equipped with and without high efficiency particulate air (HEPA) filtration (gaseous vs. PM pollution). The data can provide valuable information on lung and intestinal microbiome changes, including that which was only found after traffic-related air pollution exposure.

Epidemiological studies have linked herbicides and Parkinson’s disease (Pd), with the strongest associations resulting from long exposure durations. Paraquat (PQ), an herbicide, induces Pd-like syndromes and has widely been accepted as a Pd mimetic. Currently, there is still no cure to prevent the progression of Pd, and the search for effective therapeutic ways is urgent. Recently, the impairing activity of sirtuins (SIRTs), such as SIRT1, may correlate with Pd etiology. However, the nonspecificity of SIRT1 agonists has made the protective mechanisms against Pd unclear and hampered the therapeutic application of SIRT1. Thus, this study investigated the protective mechanism and therapeutic potential of SRT1720, a more specific agonist for SIRT1, in alleviating the toxicity of PQ-induced cellular and animal models of Pd. Here we show that SRT1720 alleviates PQ-induced toxicity in cell and animal models. Genetic silencing and pharmacological inhibition of SIRT1 attenuated SRT1720’s protection against PQ-induced toxicity. Moreover, SRT1720 not only attenuated PQ-induced increased oxidative stress and mitochondrial free radical formations but also decreased mitochondrial membrane potential. Furthermore, SRT1720 reversed PQ-induced decreased PGC-1α levels and mitochondrial biogenesis. Although PQ and SRT1720 elevated NRF2 and antioxidative enzyme levels, only PQ decreased antioxidative enzyme activity but not SRT1720. NRF2 and PGC-1α silencing attenuated SRT1720 protection against PQ-induced toxicity. SRT1720 targeted SIRT1 and activated downstream PGC-1a and NRF2 signalings to prevent PQ-induced toxicity involving oxidative stress and mitochondrial dysfunction. Thus, SRT1720 might have therapeutic potential in preventing Pd. © 2022, The Authors. All rights reserved.

The administration of recombinant cytokines, particularly interleukin-12 (IL-12), holds promising clinical potential for treating various cancers. Sustained intratumoral delivery of IL-12 can restore tumor resident Cd8 + effector T cells and induce the priming of antitumor Cd8 + effector T cells. However, these Cd8 + T cell-dependent anticancer efficacy is usually transient and accompanies the activation of immune suppressive Cd4 + Foxp3 + T regulatory cells. The underlying mechanism of T regulatory cell activation in IL-12 therapy is the upregulation of IFNγ dependent indoleamine 2,3-dioxygenase (IdO) expression. due to this negative feedback, the combinatorial use of drugs should be considered to enhance the efficacy of IL-12-mediated therapy. Herein, we designed a lipid nanoparticle (LNP) system which can successfully deliver an IdO inhibitor indoximod (INd) and IL-12 encoding mRNA. In order to increase the loading efficiency, the INd prodrug was synthesized by conjugating INd with cholesterol by ester linkage. Optimized INd prodrug encapsulating LNP successfully transfected tumor cells and macrophages, resulting in the secretion of IL-12 cytokine. With IL-12 transfection, macrophages upregulated T cell co-stimulation factor and released TNFα cytokine, indicating that the tumor microenvironment could be changed from cold tumor to hot tumor for immunotherapy. Also, by the elevated secretion of IL-12 cytokine, T cells release high levels of IFNγ, which is a central role in IL-12-mediated immunotherapy. This co-delivery system presents a promising strategy to overcome the limitations of single IL-12-mediated therapy by simultaneously promoting antitumor immune responses and inhibiting immunosuppressive mechanisms, thereby enhancing the overall efficacy of cancer immunotherapy.

Lamin A/C gene (LMNA) mutations contribute to severe striated muscle laminopathies, affecting cardiac and skeletal muscles, with limited treatment options. In this study, we delve into the investigations of five distinct LMNA mutations, including three novel variants and two pathogenic variants identified in patients with muscular laminopathy. Our approach employs zebrafish models to comprehensively study these variants. Transgenic zebrafish expressing wild-type LMNA and each mutation undergo extensive morphological profiling, swimming behavior assessments, muscle endurance evaluations, heartbeat measurement, and histopathological analysis of skeletal muscles. Additionally, these models serve as platform for focused drug screening. We explore the transcriptomic landscape through qPCR and RNAseq to unveil altered gene expression profiles in muscle tissues. Larvae of LMNA(L35P), LMNA(E358K), and LMNA(R453W) transgenic fish exhibit reduced swim speed compared to LMNA(WT) measured by danioVision. All LMNA transgenic adult fish exhibit reduced swim speed compared to LMNA(WT) in T-maze. Moreover, all LMNA transgenic adult fish, except LMNA(E358K), display weaker muscle endurance than LMNA(WT) measured by swimming tunnel. Histochemical staining reveals decreased fiber size in all LMNA mutations transgenic fish, excluding LMNA(WT) fish. Interestingly, LMNA(A539V) and LMNA(E358K) exhibited elevated heartbeats. We recognize potential limitations with transgene overexpression and conducted association calculations to explore its effects on zebrafish phenotypes. Our results suggest lamin A/C overexpression may not directly impact mutant phenotypes, such as impaired swim speed, increased heart rates, or decreased muscle fiber diameter. Utilizing LMNA zebrafish models for drug screening, we identify l-carnitine treatment rescuing muscle endurance in LMNA(L35P) and creatine treatment reversing muscle endurance in LMNA(R453W) zebrafish models.
