The hydrolysis of dNA serves as the foundational principle for gene engineering that enable precise gene cleavage at the molecular level. This process typically occurs in biological nucleases, which exhibit nucleobase-selective and catalytic hydrolysis capabilities rarely replicated in abiotic nanomaterials. Here, we demonstrate that molybdenum-terminated (Mo-terminated) edges of molybdenum disulfide (MoS2) possess the unique ability to abstract a proton (H+) from water molecules, thereby facilitating catalytic hydrolysis reactions that cleave the phosph.d.ester bonds in dNA through the action of hydroxide ions. The enhanced proton absorption at Mo-terminated edges of MoS2 significantly reduces the activation energy required for the dNA hydrolysis reaction. Furthermore, the favorable interaction between the Mo-terminated edges and thymine nucleobases promotes both charge transfer and P-O bond cleavage, enabling targeted dNA hydrolysis at 'TTTTTTT' sequences under dark conditions. This discovery underscores the potential of MoS2 as a stable, efficient nanosystem for precise genetic editing, heralding advanced applications in the field of gene engineering. © 2024, The Authors. Some argue that complementing climate change mitigation measures with solar radiation management (SRM) might prove a last resort to limit global warming to 1.5 °C. To make a socially responsible decision on whether to use SRM, it is important to consider also public opinion, across the globe and particularly in the Global South, which would face the greatest risks from both global warming and SRM. However, most research on public opinion about SRM stems from the Global North. We report findings from the first large-scale, cross-cultural study on the public opinion about SRM among the general public (N = 2,248) and students (N = 4,583) in 20 countries covering all inhabited continents, including five countries from the Global South and five ‘non-WEIRd’ (i.e. not Western, Educated, Industrialised, Rich, and democratic) countries from the Global North. As public awareness of SRM is usually low, we provided participants with information on SRM, including key arguments in favour of and against SRM that appear in the scientific debate. On average, acceptability of SRM was significantly higher in the Global South than in the ‘non-WEIRd’ Global North, while acceptability in the ‘WEIRd’ Global North was in between. However, we found substantial variation within these clusters, especially in the ‘non-WEIRd’ Global North, suggesting that countries do not form homogenous clusters and should thus be considered individually. Moreover, the average participants’ views, while generally neither strong nor polarised, differed from some expert views in important ways, including that participants perceived SRM as only slightly effective in limiting global warming. Still, our data suggests overall a conditional, reluctant acceptance. That is, while on average, people think SRM would have mostly negative consequences, they may still be willing to tolerate it as a potential last resort to fight global warming, particularly if they think SRM has only minor negative (or even positive) impact

As concerns about sugar-sweetened beverages are increasing, interest in natural sweeteners with zero calories is also growing. This study explores the health impacts of rebaudioside d and M, emerging sugar substitutes, compared to traditional alternatives and stevioside. In mouse models, we investigated the effects of rebaudioside d and M on obesity, liver health, thermogenic adipocytes, skeletal muscle, and gut microbiota. Results show no weight gain or exacerbation of obesity, no promotion of hepatic steatosis, no impairment of brown adipose tissue function and no changes in skeletal muscle metabolic proteins with either rebaudioside. Importantly, both of these sugar substitutes positively influence gut microbiota without adverse metabolic effects, suggesting their potential as safe sugar alternatives.

Cyclopent-2-enone bearing natural products possess anti-inflammatory, cytotoxic, antiproliferative, and antimicrobial activities attributed to the presence of a Michael acceptor enone group acting as a bait for protein targets. To explore the biological activity of synthetic small molecules bearing the cyclopent-2-enone moiety, a collection of 4-substituted cyclopent-2-enones ( 3 a – 3 i ) was prepared synthetically using Lewis acid-catalyzed Mukaiyama-Michael reaction (MMR) in modest to high yields. These derivatives were screened for their effect on inflammatory response mediators, namely Nrf2, NF-κB, and IL-6 through cell-based reporter assays. The results show that 5-(4-oxocyclopent-2-en-1-yl)furan-2(5 H )-one ( 3 g ) is a potent Nrf2 activator in HaCaT cells. The mechanism of Nrf2 activation by 3 g was investigated through an MS/MS-directed proteomic analysis and have shown the formation of a Michael adduct via cysteine-613 on Keap1, an inhibitor of Nrf2. density functional theory calculations show favorable Michael adduct formation between 3 g and the truncated Keap1 tripeptide Pro-Cys613-Ala at the β-carbon of the cyclopent-2-enone moiety. On the other hand, the derivative 4-(2-oxo-2-phenylethyl)cyclopent-2-en-1-one ( 3 a ) exhibited selective Nrf2 inhibition in the cancer cell line Huh7, and inhibitory activity along with 3 g against NF-κB, and IL-6 in RAW 264.7 cells. This points to the cyclopent-2-enone group being an effective scaffold for anti-inflammatory drug designs.

Idiopathic pulmonary fibrosis (IPF) is a chronic, progressive fibrotic lung disease of unknown etiology, affecting about 3 million individuals worldwide. Significant risk factors for IPF include advanced age and infectious agents. Current FdA-approved antifibrotic drugs slow the progression of pulmonary fibrosis with limited outcomes in overall survival, highlighting the need for novel pharmacological agents. Ugonin L (UL), a cyclized geranylflavonoid derived from Helminthostachys zeylanica , has been reported to have anti-inflammatory and antioxidant activities. However, the therapeutic potential of UL for pulmonary fibrosis remains unexplored. In this study, we demonstrated that UL mitigated pulmonary fibrosis in bleomycin (BLM)-induced mice. Specifically, UL improved the alveolar-capillary barrier integrity, decreasing inflammatory cytokines (TGF-β1, TNF-α, IL-1β, IL-6) in bronchoalveolar lavage fluid (BALF). UL ameliorated lesions in BLM-induced fibrotic lungs, reducing radiological signs of lung injury, alveolar septal thickening, and collagen deposition. In RNA-seq analysis, UL downregulated genes related to cell migration and ECM remodeling in TGF-β1-induced LL29 human lung fibroblasts. In particular, UL decreased cell migration, fibrotic marker expression, MMP-2 activity, and myofibroblast activation. In molecular modeling, UL interacted with key pharmacophores and the putative ATP-binding sites of the TβRI and TβRII kinase domains. Correspondingly, UL reduced the phosphorylation of key mediators in both the canonical (SMAd2/3) and non-canonical (ERK1/2 and PI3K) TGF-β signaling pathways. Furthermore, UL downregulated the PI3K/Akt/mTOR axis and promoted autophagy in TGF-β1-induced LL29 cells. Taken together, our findings demonstrate that UL acts as a novel TβRs inhibitor and shows therapeutic potential for pulmonary fibrosis.

An environmentally friendly absorbent sponge was successfully developed for diesel spill remediation via alginate polymer crosslinking followed by surface modification using tannic acid and dodecanethiol (TA/do). The optimal sponge was obtained with a 3 h crosslinking duration, achieving a balance between absorption performance and structural durability, while longer crosslinking times led to increased density and reduced efficiency. Surface modification imparted hydrophobicity to the alginate sponge, confirmed by a water contact angle of 120.2°, and the successful incorporation of TA/do was confirmed from FTIR and XPS analysis. The modified sponge (AA-3-TA/do) exhibited a diesel absorption capacity of 17.54 g/g—a 36 % increase over the unmodified form—and retained ∼90 % of this capacity after 34 absorption–desorption cycles. Kinetic modeling showed that diesel was rapidly absorbed during early contact, with faster uptake than water. A cytotoxicity assay confirmed its non-toxic nature, indicating environmental compatibility. This study demonstrates that a simple, scalable modification using accessible materials can significantly enhance oil sorbent performance without requiring complex techniques or hazardous reagents. © 2025 Elsevier B.V.

Unambiguously layer by layer (LBL) assembly of graphene quantum dots (GQds) and dye (GQds/dye) on TiO2 photoanode is the traditional and straightforward approach in the fabrication of graphene quantum dot-sensitized solar cells (QdSSCs). Unfortunately, limited light absorption and low affinity of GQds to TiO2 surface shadow the advantages of LBL and constrains its practical application. Herein, a new strategy of mixture configuration (GQds+dye) was investigated. A distinctive nanoporous honeycomb hexagonal carbon network of GQds was found with fewer defects single crystalline structure, and an average size of 9.87 nm was produced from cellulose. Experimental results demonstrated that LBL exhibited the highest efficiency of 16.76 % under low illumination but a lower efficiency (1.43%) than the mixture method (2.91%) under standard light. The increased Jsc (5.075 mA/cm2) and high charge collection efficiency (0.96) in the mixture sample indicated enhanced electron collection at TiO2. The less -OH groups on TiO2 provides a good surface intact of GQds and N719. In addition to that, the high surface potential (33.47 mV) of the premixed sample restricted the photogenerated electrons to go into a deep state, reducing back electron transfer. Therefore, mixture assembly of co-sensitization is an effective approach for light-harvesting in QdSSC. © 2021, CC BY.

This paper proposed a contact-force-controlled surface profiler by integrating the output of a LVT (Linear Velocity Transducer) in the inner-loop (to increase the damping) of the control system, so that not only the noise amplification effect due to the differentiator in the PId circuit, but the hysteresis effect of the force actuator could be avoided. A voice coil was applied as a force actuator, and a LVdT (Linear Variable differential Transformer) was applied at outer-loop to measure the surface profile. The maximum contact force between the probe and the sample was sensed by a load cell and limited to a tolerable value by the system, so the sample surface would not be destroyed by the probe. Four kinds of controllers using traditional PId, Ziegler-Nichols-based PId, fuzzy-PId, and neural-fuzzy design methods were applied for comparison. Besides, practical parameter variation (hysteresis effect) of force actuator was considered. Moreover, to investigate the system performance the testing input of the surface profile was generated by combining both FM (with frequency changing from 20 HZ to 20 KHZ according to the specification of voice coil) and AM signals. This kind of surface profile was very practical and not being found in the previous literature. By the results of simulations and practical testing, one can see the intelligent fuzzy-neural system was more robust to the parameter variation of force actuator.
