Type 2 diabetes is an increasingly prevalent disease and patients do not always manage the disease properly. Therefore, creating tools that help diabetics self-manage their condition over time is of the utmost importance. Technological tools that include features meeting this population's needs, with an integrated personalized feedback system, in an environment of gamified incentives, may be the way for developing a sustained app's usage. This work aims to present a still in development, novel approach to managing diabetes type 2 with a focus on data analysis from user-inputs, gamification, and personalized coaching, with an accessible user interface. Thus, we will present some functionalities, in particular the theoretical and practical concept that is behind this development, namely the Transtheoretical model of behavior change to create different profiles, to provide customized feedback according to the user's behavioral stage, and also to ascertain if it induces any improvement at the behavioral level.

ABSTRACTABSTRACTAll-terrain vehicle (ATV) crashes are one of the leading causes of death and injury in agriculture across the United States. Use of ATVs is highly prevalent in rural areas and has seen an increase in their application for a variety of agricultural work tasks. Empirical research on the use of these vehicles for agricultural tasks and associated injuries is limited, especially in the United States. Moreover, little is known about the risk factors associated with ATV-related injuries while doing farm work. A comprehensive review was conducted to evaluate the current injury burden of ATV use in agriculture, the need for future research, and possible solutions related to agricultural ATV safety. Potential injury prevention approaches are evaluated based on the hierarchy of control, including elimination or substitution (using side-by-side vehicles instead of ATVs or modifying ATV design), engineering control (operator protection devices), administrative authority (regulations and standards in the United States and around the world), training, and use of personal protective equipment. In addition, vehicle nomenclature, risk factors, and crash mechanisms are reviewed. Recommendations to decrease the likelihood of ATV crashes and injuries are provided.

Introduction: Alzheimer's disease (AD) reports heterogeneity of neuropsychological symptoms misleading the differential diagnosis with other forms of dementia, such as dementia with Lewy bodies (DLB). About 50% of DLB patients are misdiagnosed as AD cases. Likewise, the diagnosis of both diseases is mainly based on clinical characteristics. However, differentiating AD of those with DLB based on neuropsychological symptoms and anatomical and functional brain changes remains challenging. Aim: To establish the main neuropsychological, anatomical, and functional similarities and differences in patients with AD and DLB. Methods: The present study followed the PRISMA guidelines and included studies from the PubMed, Scopus, and Web of sciences databases, published between January 2000 and July 2022. Results: Forty-one articles were included in this systematic review for critical analysis. Our results suggest that the cognitive key domains to consider in the differential diagnosis are memory, executive function, attention, visuospatial/visuoconstructive skills, and verbal fluency (both semantic and phonological). The stage and severity of both diseases would be essential for differential diagnosis. On the other hand, the anatomical and functional changes suggest a similar atrophy pattern between AD and DLB in the frontal, parietal, temporal, hippocampal, and precuneus regions. Conclusion: The differential diagnosis between AD and DLB is challenging in clinical practice. Three-dimensional (3D) genome dynamics are crucial for cellular functions and disease. However, real-time, live-cell DNA visualization remains challenging, as existing methods are often confined to repetitive regions, suffer from low resolution, or require complex genome engineering. Here, we present Oligo-LiveFISH, a high-resolution, reagent-based platform for dynamically tracking non-repetitive genomic loci in diverse cell types, including primary cells. Oligo-LiveFISH utilizes fluorescent guide RNA (gRNA) oligo pools generated by computational design, in vitro transcription, and chemical labeling, delivered as ribonucleoproteins. Utilizing machine learning, we characterized the impact of gRNA design and chromatin features on imaging efficiency. Multi-color Oligo-LiveFISH achieved 20-nm spatial resolution and 50-ms temporal resolution in 3D, capturing real-time enhancer and promoter dynamics. Our measurements and dynamic modeling revealed two distinct modes of chromatin communication, and active transcription slows enhancer-promoter dynamics at endogenous genes like FOS. Oligo-LiveFISH offers a versatile platform for studying 3D genome dynamics and their links to cellular processes and disease. © 2025 Elsevier Inc.

The Augmented Whole-body Scanning via Magnifying PET (AWSM-PET) device is a novel technology designed to improve the detectability of lesions in whole-body PET/CT imaging. We have developed a prototype AWSM-PET device that consists of two panels of high-resolution time-of-flight (TOF) PET detectors, integrated with the standard readout electronics and event processing architecture of the Biograph Vision (Siemens healthineers). The system exhibits excellent spatial, energy and timing resolution, with an average coincidence resolving time of $\sim 217$ ps FWHM and an average energy resolution of $\sim 11 \%$ FWHM at 511 keV. Custom firmware and software have been developed to encode the large number of detector elements and to form coincidences between the AWSM-PET detectors and the Biograph Vision scanner. We have pioneered a specialized image reconstruction approach utilizing a GPU-based list-mode framework, which incorporates the system’s point-spread function (PSF) to optimize image quality and spatial resolution. Continuous-bed motion (CBM) whole-body imaging protocols have been implemented for the AWSM-PET operation to improve the uniformity of sensitivity across the patient’s body. Sensitivity enhancement was evaluated using a point source. The first human subject study was a head-andneck cancer patient scheduled for a standard-of-care (SOC) FDG PET/CT scan following treatment. Both the SOC-PET/CT images and the AWSM-PET images were acquired simultaneously for a total of 17 minutes using the CBM mode. Results show high-resolution and high-quality AWSM-PET images. We anticipate to image more patients in the summer, refine AWSM-PET image reconstruction parameters and correction techniques. Comparison between the AWSM-PET and the SOC-PET/CT will be presented at the 2024 IEEE MIC conference.

Autonomic nervous system (ANS) has been suggested often associated with the isolated rapid eye movement (REM) sleep behavior disorder (iRBD), with iRBD, and heart rate variability (HRV) is a commonly used tool to assess it. However, the accuracy of HRV is heavily influenced by respiratory rate (RespR), potentially causing misinterpretation of frequency domain HRV indices. This study compares sleep-related ANS function between iRBD patients and a control group (CG) using respiratory-adjusted HRV spectral analysis. It involves 20 iRBD and 20 CG participants in each group, from which five-minute ECG signal epochs during N2, N3, and REM sleep phases are analyzed. RespR were estimated from ECG-derived respiratory signals. Both low and high frequency bands definition for HRV analysis were redefined using the RespR. The results indicate significant differences in HRV parameters between iRBD and CG subjects during REM sleep, highlighting disrupted autonomic regulation in iRBD. This finding is also evident in non-REM phases, thus emphasizing the need to incorporate RespR in HRV analysis, especially in sleep-related studies, for a comprehensive understanding of autonomic dysfunction.

The outbreak of pandemics (e.g., severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2 in 2019), influenza A viruses (H1N1 in 2009), etc.), and worldwide spike in the aging population have created unprecedented urgency for developing new drugs to improve disease treatment. As a result, extensive efforts have been made to design novel techniques for efficient drug monitoring and screening, which form the backbone of drug development. Compared to traditional techniques, microfluidics-based platforms have emerged as promising alternatives for high-throughput drug screening due to their inherent miniaturization characteristics, low sample consumption, integration, and compatibility with diverse analytical strategies. Moreover, the microfluidic-based models utilizing human cells to produce in-vitro biomimetics of the human body pave new ways to predict more accurate drug effects in humans. This review provides a comprehensive summary of different microfluidics-based drug sensing and screening strategies and briefly discusses their advantages. Most importantly, an in-depth outlook of the commonly used detection techniques integrated with microfluidic chips for highly sensitive drug screening is provided. Then, the influence of critical parameters such as sensing materials and microfluidic platform geometries on screening performance is summarized. This review also outlines the recent applications of microfluidic approaches for screening therapeutic and illicit drugs. Moreover, the current challenges and the future perspective of this research field is elaborately highlighted, which we believe will contribute immensely towards significant achievements in all aspects of drug development. © 2022 Elsevier B.V.

Cytokines and chemokines play important roles in inflammation and repair following brain injury. M1 microglia are pro-inflammatory which produce cytokines and lead to neural injury;in contrast, M2 microglia are anti-inflammatory, release neurotrophic factors, and promote neural repair processes. Chemokine CCL5, increased after TBI, shows a neuroprotective function by reducing oxidative stress. CCL5 may contribute to the balance between oxidative stress and immune responses after brain injury. Herein, we investigated the role of CCL5 in microglial polarization after mild traumatic brain injury, focusing on the cerebral cortex.C57BL/6 and CCL5 knockout (CCL5-KO) mice were given a mild traumatic brain injury (TBI) using weight-drop. Neurological parameters such as motor and sensory functions were analyzed by mNSS score, accelerating rotarod, beam walking, and adhesive removal tests. Oxidative stress and neuron damage were measured by NADPH oxidase activity, hypoxyloprobe staining and FJC. Performance of motor and sensory functions in both WT and CCL5-KO mice were impaired after brain injury which recovered after 7 days-post-injury (dpi) in the WT group but only after 14 days in the CCL5-KO mouse group. RT-qPCR analysis revealed that pro-inflammation cytokines - IL-1β, TNF-α, and IL-6 were higher in CCL5-KO mice compared to WT mice at 4 and 14 dpi. In contrast, M2-like microglia markers - IL-10 and Agr-1 were increased in WT mouse cortical tissue at 4 dpi. Oxidative stress increased both M1 and M2- related cytokine expression in BV2 cells treated with H 2 O 2;CCL5 treatment increased M2- but suppressed M1- related cytokine gene activation. Background and Objective: Mobile applications could be effectively used for dietary intake assessment, physical activity monitoring, behavior improvement, and nutrition education. The aim of this review is to determine the effectiveness of mobile applications in improving nutrition behaviors through a systematic review of literature. Methods: The review protocol was registered with PROSPERO: registration number CRD42018118809, and followed PRISMA guidelines. We involved original articles including mobile electronic devices for improving dietary intake, physical activity, and weight management in adult populations in this review. Data were retrieved from January 2010 to December 2018 with PubMed, Web of Science, Excerpta Medica Database (Embase), and Cumulative Index to Nursing and Allied health Literature (CINAHL) as data sources. Authors individually screened the titles and abstracts, then full articles in order to obtain papers that met inclusion criteria. Results: The database search yielded 2962 records. After removing the duplicates and analyzing the full text papers a total of 8 original articles were reviewed. Two articles showed obvious bias and were not included in our results or discussion. The remaining six articles with low to moderate bias risk were included in this systematic review. Three selected studies were randomized control trials (RCTs) with over 180 participants each. The other three studies were a nested trial, a case-control trial, and a pilot RCT with 36, 162, and 24 participants respectively. All larger RCTs and the small case control trail showed significant improvements in some nutritional-health objectives measured. The other two trials showed insignificant improvements in outcomes measured between groups. Conclusion: This study highlights the potential significant health benefits acquirable through mobile health application-assisted nutrition interventions.
