Massive usage of antibiotics harms the environment through animals’ excrement and squeezes open-wellbeing issues. Enrofloxacin (EF), an antimicrobial agent, treats bacterial diseases and infections in cattle, poultry and exotic animals and exhibits harmful side effects. The present work proposed a sensitive and rapid electrochemical scaffold based on the Cu-Ni bimetallic-organic framework (Cu-Ni-MOF) for the detection of harmful EF in real-time applications. The Cu-Ni-MOF was synthesized through solvothermal approach with the support of a multifunctional organic ligand, 4,4-biphenyldicarboxylicacid and characterized through various analytical tools. The electrochemical sensor for EF detection was constructed by drop-casting the Cu-Ni-MOF on a glassy carbon electrode and the fabricated sensor exhibited higher catalytic activity when compared to the parental counterparts by exhibiting 1.7-fold higher current for EF oxidation. Further, the fabricated sensor is highly active in the linear range response of 200 nM–2 mM with a limit of detection of 15.17 nM and sensitivity of 638.71 µA mM cm −2 under optimized environments. The higher electrocatalytic activity of the Cu-Ni-MOF electrode synergistically stems from the strong interaction between Cu 2+ of the MOF and pyridone and carboxylate moieties of the EF and facile electron transition facilitated by Ni 2+ . The practicability of the sensor was evaluated in milk, egg and river water samples and the obtained results were validated with Liquid Chromatography-Mass Spectrometric technique. Furthermore, the prepared catalyst was utilized for the photochemical decontamination of EF and the present Cu-Ni-MOF catalyst delivered excellent photocatalytic activity in the decontamination of EF under visible light at ambient temperature.

Background: Decades of steady improvements in life expectancy in Europe slowed down from around 2011, well before the COVID-19 pandemic, for reasons which remain disputed. We aimed to assess how changes in risk factors and cause-specific death rates in different European countries related to changes in life expectancy in those countries before and during the COVID-19 pandemic. Methods: We used data and methods from the Global Burden of Diseases, Injuries, and Risk Factors Study 2021 to compare changes in life expectancy at birth, causes of death, and population exposure to risk factors in 16 European Economic Area countries (Austria, Belgium, Denmark, Finland, France, Germany, Greece, Iceland, Ireland, Italy, Luxembourg, the Netherlands, Norway, Portugal, Spain, and Sweden) and the four UK nations (England, Northern Ireland, Scotland, and Wales) for three time periods: 1990–2011, 2011–19, and 2019–21. Changes in life expectancy and causes of death were estimated with an established life expectancy cause-specific decomposition method, and compared with summary exposure values of risk factors for the major causes of death influencing life expectancy. Findings: All countries showed mean annual improvements in life expectancy in both 1990–2011 (overall mean 0·23 years [95% uncertainty interval [UI] 0·23 to 0·24]) and 2011–19 (overall mean 0·15 years [0·13 to 0·16]). The rate of improvement was lower in 2011–19 than in 1990–2011 in all countries except for Norway, where the mean annual increase in life expectancy rose from 0·21 years (95% UI 0·20 to 0·22) in 1990–2011 to 0·23 years (0·21 to 0·26) in 2011–19 (difference of 0·03 years). In other countries, the difference in mean annual improvement between these periods ranged from –0·01 years in Iceland (0·19 years [95% UI 0·16 to 0·21] vs 0·18 years [0·09 to 0·26]), to –0·18 years in England (0·25 years [0·24 to 0·25] vs 0·07 years [0·06 to 0·08]). N-acetyltransferase 10 catalyzes RNA N4-acetylcytidine (ac4C) modifications and thus regulates RNA stability and translation efficiency. However, the deacetylase for ac4C is unknown. SIRT7 was initially identified as an NAD+-dependent protein deacetylase and plays essential roles in genome stability, circadian rhythms, metabolism, and aging. In this study, we identified SIRT7 as a deacetylase of the ac4C of ribosomal (r)RNA for the first time and found it to be NAD+-independent. Our data highlight the important role of SIRT7 in rRNA ac4C modification and suggest an additional epitranscriptional regulation of aging.

FoxO transcription factors actuate the formative pluripotency specific gene expression programme

Naïve pluripotency is sustained by a self-reinforcing gene regulatory network (GRN) comprising core and naïve pluripotency-specific transcription factors (TFs). Upon exiting naïve pluripotency, embryonic stem cells (ESCs) transition through a formative post-implantation-like pluripotent state, where they acquire competence for lineage choice. However, the mechanisms underlying disengagement from the naïve GRN and initiation of the formative GRN are unclear. Here, we demonstrate that phosphorylated AKT acts as a gatekeeper that prevents nuclear localisation of FoxO TFs in naïve ESCs. PTEN-mediated reduction of AKT activity upon exit from naïve pluripotency allows nuclear entry of FoxO TFs, enforcing a cell fate transition by binding and activating formative pluripotency-specific enhancers. Indeed, FoxO TFs are necessary and sufficient for the activation of the formative pluripotency-specific GRN. Our work uncovers a pivotal role for FoxO TFs in establishing formative post-implantation pluripotency, a critical early embryonic cell fate transition. © The Author(s) 2024.

Background and Objective: Mobile applications could be effectively used for dietary intake assessment, physical activity monitoring, behavior improvement, and nutrition education. The aim of this review is to determine the effectiveness of mobile applications in improving nutrition behaviors through a systematic review of literature. Methods: The review protocol was registered with PROSPERO: registration number CRD42018118809, and followed PRISMA guidelines. We involved original articles including mobile electronic devices for improving dietary intake, physical activity, and weight management in adult populations in this review. Data were retrieved from January 2010 to December 2018 with PubMed, Web of Science, Excerpta Medica Database (Embase), and Cumulative Index to Nursing and Allied Health Literature (CINAHL) as data sources. Authors individually screened the titles and abstracts, then full articles in order to obtain papers that met inclusion criteria. Results: The database search yielded 2962 records. After removing the duplicates and analyzing the full text papers a total of 8 original articles were reviewed. Two articles showed obvious bias and were not included in our results or discussion. The remaining six articles with low to moderate bias risk were included in this systematic review. Three selected studies were randomized control trials (RCTs) with over 180 participants each. The other three studies were a nested trial, a case-control trial, and a pilot RCT with 36, 162, and 24 participants respectively. All larger RCTs and the small case control trail showed significant improvements in some nutritional-health objectives measured. The other two trials showed insignificant improvements in outcomes measured between groups. Conclusion: This study highlights the potential significant health benefits acquirable through mobile health application-assisted nutrition interventions. Tetraheterocyclic compounds, derived from natural sources and contemporary pharmaceuticals, have shown promise as multitarget therapeutic agents. However, their mechanisms of action remain partially understood. In this study, we synthesized a series of 6H-thiochromeno[2,3-c]quinolin-12(12H)-one derivative, totaling 26 compounds, and assessed their potential for therapeutic application. We evaluated their effects on cell proliferation and conducted NCI-60 cell panel assays. MTT assays revealed that select compounds exhibited notable antiproliferative activity against two breast cancer cell lines (MCF-7 and MDA-MB-468). Notably, compounds 17 and 18 displayed the highest cytotoxicity against these cell lines. Furthermore, one-dose assays of the NCI-60 human tumor cell line screening program identified compounds 6, 7, 16, 18, 20, 24, 25, and 30 for further investigation. Subsequent five-dose cytotoxicity studies focused on compounds 18 and 20, which met the threshold inhibition criteria across a panel of cell lines. Our study highlights the effectiveness of compounds 18 and 20 in targeting breast cancer cell lines. Molecular docking simulations revealed that these compounds bind to the active sites of topoisomerase I (TOPO I). Our findings suggest that these novel compounds are promising anticancer agents, particularly against breast cancer, and are worthy of consideration as lead pharmacological candidates. © 2023 The Authors

BACKGROUND:The International System for Reporting Serous Fluid Cytopathology (ISRSFC) was introduced globally in 2019 in response to the absence of a standardized reporting system for serous fluid cytology. This study presents experiences implementing this system across three distinct hospitals in Taiwan. METHODS:A total of 6177 serous fluid specimens in three hospitals in Taiwan between 2018 and 2020 were retrospectively reviewed and reclassified according to the ISRSFC. Cytohistological correlation and chart review were further performed to investigate etiologies and risks of malignancy (ROMs). RESULTS:Reclassification showed that 34 (0.7%) of 4838 pleural effusions were nondiagnostic (ND), 4086 (84.5%) were negative for malignancy (NFM), 201 (4.2%) were atypia of undetermined significance (AUS), 92 (1.9%) were suspicious for malignancy (SFM), and 425 (8.8%) were malignant (MAL). The 1231 ascites cases contained 13 (1.1%) ND, 1004 (81.6%) NFM, 53 (4.3%) AUS, 31 (2.5%) SFM, and 130 (10.6%) MAL specimens. In pleural effusions, the ROM was 2.9% for ND, 14.0% for NFM, 52.2% for AUS, 85.9% for SFM, and 95.1% for MAL. In ascites, it was 15.4% for ND, 19.1% for NFM, 52.8% for AUS, 83.9% for SFM, and 92.3% for MAL. In pericardial effusions, it was 0.0% for ND, 11.6% for NFM, 30.8% for AUS, 100.0% for SFM, and 95.2% for MAL. Different effusions' most common benign and malignant etiologies were also disclosed. CONCLUSIONS:These multi-institutional data have determined the diagnostic usefulness of the ISRSFC, which provides pathologists and physicians with invaluable assistance in correctly classifying effusions for further management.

Engineering and characterizing proteins can be time-consuming and cumbersome, motivating the development of generalist CRISPR-Cas enzymes to enable diverse genome editing applications. However, such enzymes have caveats such as an increased risk of off-target editing. To enable scalable reprogramming of Cas9 enzymes, here we combined high-throughput protein engineering with machine learning (ML) to derive bespoke editors more uniquely suited to specific targets. Via structure/function-informed saturation mutagenesis and bacterial selections, we obtained nearly 1,000 engineered SpCas9 enzymes and characterized their protospacer-adjacent motif (PAM) requirements to train a neural network that relates amino acid sequence to PAM specificity. By utilizing the resulting PAM ML algorithm (PAMmla) to predict the PAMs of 64 million SpCas9 enzymes, we identified efficacious and specific enzymes that outperform evolution-based and engineered SpCas9 enzymes as nucleases and base editors in human cells while reducing off-targets. An in silico directed evolution method enables user-directed Cas9 enzyme design, including for allele-selective targeting of the RHO P23H allele in human cells and mice. Together, PAMmla integrates ML and protein engineering to curate a catalog of SpCas9 enzymes with distinct PAM requirements, and motivates the use of efficient and safe bespoke Cas9 enzymes instead of generalist enzymes for various applications.

Cobalt and nickel oxides with large theoretical capacitance, high redox activity and low-cost features are widely applied as electroactive material of battery supercapacitor hybrids (BSH) to reduce energy consumption and create renewable energy. Synthesizing cobalt and nickel oxides using zeolitic imidazolate framework 67 (ZIF67) can enhance conductivity and electroactivity and maintain surface area and porosity of ZIF67. Structure directing agent (SDA) can control morphology and improve electrochemical performance. It is attractive to apply SDA for controlling morphology of oxidized ZIF67 and achieving excellent energy storage. In this work, ammonium tetrafluoroborate (NH4BF4) is firstly applied as SDA to synthesize ZIF67 derivative, which is then annealed to form leaf-like cobalt and nickel oxides as electroactive material. The optimal oxidized ZIF67 derivative shows a larger specific capacitance (CF) of 828.9 F/g than those of 519.9 and 5.0 F/g respectively for the oxidized ZIF67 and pristine ZIF67 electrodes, due to preferable leaf-like structure and (311) facet for the former case. A BSH with oxidized ZIF67 derivative positive electrode and graphene negative electrode shows maximum energy density of 10.2 Wh/kg at 357 W/kg, and CF retention of 91% and Coulombic efficiency of 97% after 10,000 cycles. The novel SDA of NH4BF4 is expected to design other efficient electroactive materials for effective energy storage. © 2023 Elsevier Ltd

Two new non-withanolidal steroids with a cross-conjugated dienone structural unit in ring A, capnesterones A (1) and B (2), were isolated from the soft coral Capnella imbricata in the southeast waters of Taiwan. The compounds' structures were elucidated through a comprehensive analysis of spectroscopic data, including IR, ESIMS, ¹H NMR, and ¹³C NMR, and through comparison with previous literature. At a concentration of 10 μM, steroids 1 and 2 exhibited reductions in iNOS levels (60 % and 82 %) compared to LPS-stimulated control cells, along with increased release (134 % and 110 %) against COX-2 protein expressions. © 2024 Phytochemical Society of Europe
