Mathematical competency is vital to success in engineering. Competency requires not only short-term mastery of mathematical concepts, but also long-term retention. Research in cognitive psychology shows that the more times information is retrieved from memory, the more likely it is to be remembered over the long-term. Research also shows that increasing the amount of time between retrievals increases long-term retention. Which of these interventions-increasing the amount or the temporal spacing of retrieval practice-has the greater impact on long-term retention? Supported by NSF's Improving Undergraduate STEM Education (IUSE) program, we answered this question by independently manipulating the amount and the spacing of retrieval practice in a precalculus course for engineering students. This was done by varying the number of quiz questions concerning key precalculus objectives (amount of practice) and varying whether those questions were massed on a single quiz or distributed across quizzes in multiple weeks (spacing of practice). Long-term retention was assessed in a test of precalculus knowledge administered one month after the end of the precalculus course. We found that students were significantly more likely to retain precalculus objectives when quiz questions had been spaced versus massed. Increasing the number of quiz questions did not significantly affect retention. These findings suggest that educators wishing to increase students' long-term retention of mathematics knowledge should increase the spacing, rather than the amount, of retrieval practice in their courses. © American Society for Engineering Education, 2018.

Recent advancements in the discovery of natural products have made significant progress in rapidly identifying known compounds from complex extracts using advanced spectroscopic technologies. Among these methods, one powerful approach called MS/MS molecular networking (MN) organizes chemical similarity based on MS/MS fragmentation data, serving as a valuable complement to traditional dereplication strategies. In our ongoing research focused on lead discovery from a sustainable medicinal source, an aquaculture soft coral Sarcophyton trocheliophorum, the MN profiling approach revealed the chemical diversity of biscembranoid analogs. By analyzing the MN pattern, biscembranoid derivatives with unique MS/MS fragments were prioritized for further investigation. Two novel biscembranoids, sarcotroxide A (1) and B (2), were successfully obtained through targeted isolation. These novel biscembranes contained an unprecedented 3-hydroperoxy-3-methylcyclohex-1-ene moiety compared with all biscembranoid analogs. The chemical structures of the two new biscembranoids were confirmed through spectroscopic analyses and in silico calculations. Their anti-inflammatory properties were assessed against the generation of superoxide anion (O₂^•−) and elastase release in activated human neutrophils. Sarcotroxides A (1) and B (2) exhibited anti-inflammatory activity by inhibiting O₂^•− accumulation (with 43.67% and 61.62% inhibition) and elastase release (with 38.21% and 61.62% inhibition) at 10 µM. These findings validated the potential of these biscembranoids from the cultured S. trocheliophorum as promising candidates for the future development of anti-inflammatory agents specifically designed to target neutrophils. © 2023 The Authors

Background: Cryptococcosis is one of the most frequent fungal eye infections in patients with immunosuppression. Currently, treatment approaches for non-meningeal, non-pulmonary cryptococcosis are based on those used for cryptococcal meningitis or pneumonia. Case presentation: We present a rare case of non-meningeal, non-pulmonary cryptococcosis with clinical manifestations limited to one eye of a cadaveric kidney transplant recipient with chronic-active antibody-mediated rejection. Typical manifestations, diagnosis, and treatments, including antifungal therapies, adjunctive therapies, and immunosuppression reduction, are discussed. After timely diagnosis and treatment, her visual acuity recovered to baseline without recurrence or sequelae of cryptococcosis. Conclusions: Clinicians should be aware of rare presentations of fungal infections, especially when a kidney transplant recipient with rejection has been treated with intensive immunosuppressants. Early diagnosis with individualized therapies may have a favorable prognosis. © 2023, BioMed Central Ltd., part of Springer Nature.

Mixture-of-experts variational autoencoder for clustering and generating from similarity-based representations

Clustering high-dimensional data, such as images or biological measurements, is a long-standing problem and has been studied extensively. Recently, Deep Clustering gained popularity due to the non-linearity of neural networks, which allows for flexibility in fitting the specific peculiarities of complex data. Here we introduce the Mixture-of-Experts Similarity Variational Autoencoder (MoE-Sim-VAE), a novel generative clustering model. The model can learn multi-modal distributions of high-dimensional data and use these to generate realistic data with high efficacy and efficiency. MoE-Sim-VAE is based on a Variational Autoencoder (VAE), where the decoder consists of a Mixture-of-Experts (MoE) architecture. This specific architecture allows for various modes of the data to be automatically learned by means of the experts. Additionally, we encourage the latent representation of our model to follow a Gaussian mixture distribution and to accurately represent the similarities between the data points. We assess the performance of our model on synthetic data, the MNIST benchmark data set, and a challenging real-world task of defining cell subpopulations from mass cytometry (CyTOF) measurements on hundreds of different datasets. MoE-Sim-VAE exhibits superior clustering performance on all these tasks in comparison to the baselines and we show that the MoE architecture in the decoder reduces the computational cost of sampling specific data modes with high fidelity. Validation of extracellular ligand-receptor interactions by Flow-TriCEPS

Objective: The advent of ligand-based receptor capture methodologies, allows the identification of unknown receptor candidates for orphan extracellular ligands. However, further target validation can be tedious, laborious and time-consuming. Here, we present a methodology that provides a fast and cost-efficient alternative for candidate target verification on living cells. Results: In the described methodology a ligand of interest (e.g. transferrin, epidermal growth factor or insulin) was conjugated to a linker (TriCEPS) that carries a biotin. To confirm ligand/receptor interactions, the ligand-TriCEPS conjugates were first added onto living cells and cells were subsequently labeled with a streptavidin-fluorophore and analyzed by flow cytometry (thus referred as Flow-TriCEPS). Flow-TriCEPS was also used to validate identified receptor candidates when combined with a siRNA knock down approach (i.e. reduction of expression levels). This approach is versatile as it can be applied for different classes of ligands (proteins, peptides, antibodies) and different cell lines. Moreover, the method is time-efficient since it takes advantage of the large variety of commercially available (and certified) siRNAs. © 2018 The Author(s).

Chondrichthyans (sharks, rays and chimeras) are a fascinating and highly vulnerable group of early branching gnathostomes. However, they remain comparatively poorly sampled from the point of view of molecular resources, with deep water taxa being particularly data deficient. The development of long-read sequencing technologies enables the analysis of phylogenetic relationships through a precise and reliable assembly of complete mtDNA genomes. The sequencing and characterization of the complete mitogenome of the opal chimeraChimera opalescensLuchetti, Iglésias et Sellos 2011, using the long-read technique PacBio HiFi is presented. The entire mitogenome was 23,411 bp long and shows the same overall content, i.e. 13 protein-coding genes, 22 transfer RNA and 2 ribosomal RNA genes, as all other examined Chondrichthyan mitogenomes. Phylogenetic reconstructions using all available Chondrichthyan mitogenomes, including 11 Holocephali (chimeras and ratfishes), placesC. opalescenswithin the Chimaeridae family. Furthermore, the results reinforce previous findings, showing the genusChimeraas paraphyletic and thus highlighting the need to expand molecular approaches in this group of cartilaginous fishes.

Massive usage of antibiotics harms the environment through animals’ excrement and squeezes open-wellbeing issues. Enrofloxacin (EF), an antimicrobial agent, treats bacterial diseases and infections in cattle, poultry and exotic animals and exhibits harmful side effects. The present work proposed a sensitive and rapid electrochemical scaffold based on the Cu-Ni bimetallic-organic framework (Cu-Ni-MOF) for the detection of harmful EF in real-time applications. The Cu-Ni-MOF was synthesized through solvothermal approach with the support of a multifunctional organic ligand, 4,4-biphenyldicarboxylicacid and characterized through various analytical tools. The electrochemical sensor for EF detection was constructed by drop-casting the Cu-Ni-MOF on a glassy carbon electrode and the fabricated sensor exhibited higher catalytic activity when compared to the parental counterparts by exhibiting 1.7-fold higher current for EF oxidation. Further, the fabricated sensor is highly active in the linear range response of 200 nM–2 mM with a limit of detection of 15.17 nM and sensitivity of 638.71 µA mM cm −2 under optimized environments. The higher electrocatalytic activity of the Cu-Ni-MOF electrode synergistically stems from the strong interaction between Cu 2+ of the MOF and pyridone and carboxylate moieties of the EF and facile electron transition facilitated by Ni 2+ . The practicability of the sensor was evaluated in milk, egg and river water samples and the obtained results were validated with Liquid Chromatography-Mass Spectrometric technique. Furthermore, the prepared catalyst was utilized for the photochemical decontamination of EF and the present Cu-Ni-MOF catalyst delivered excellent photocatalytic activity in the decontamination of EF under visible light at ambient temperature.

ABSTRACTABSTRACTAll-terrain vehicle (ATV) crashes are one of the leading causes of death and injury in agriculture across the United States. Use of ATVs is highly prevalent in rural areas and has seen an increase in their application for a variety of agricultural work tasks. Empirical research on the use of these vehicles for agricultural tasks and associated injuries is limited, especially in the United States. Moreover, little is known about the risk factors associated with ATV-related injuries while doing farm work. A comprehensive review was conducted to evaluate the current injury burden of ATV use in agriculture, the need for future research, and possible solutions related to agricultural ATV safety. Potential injury prevention approaches are evaluated based on the hierarchy of control, including elimination or substitution (using side-by-side vehicles instead of ATVs or modifying ATV design), engineering control (operator protection devices), administrative authority (regulations and standards in the United States and around the world), training, and use of personal protective equipment. In addition, vehicle nomenclature, risk factors, and crash mechanisms are reviewed. Recommendations to decrease the likelihood of ATV crashes and injuries are provided.
