Octacalcium phosphate is the precursor of biological apatite, that is why it is of special interest in bone regeneration due to it's biocompatibility and biodegradability. This study was performed to examine the in vitro stability of octacalcium phosphate granules in physiological solutions. Powder X-ray diffraction (XRD), infrared spectroscopy (IR), plasma atomic emission spectroscopy (AES) and scanning electron microscopy (SEM) data were obtained before and after granules immersion. The variation of Ca2+ and HPO42- concentration in the soaking solutions suggests that the material transformation occurs through octacalcium phosphate dissolution followed by hydroxyapatite precipitation. © Published under licence by IOP Publishing Ltd.

A common cause of loss of vision for humans is lens opacity, or cat aracts, which prevent patients from having a full field of vision and lower their visual acuities. Cataracts can be corrected for by replacing the opaque lens with a toric intraocular lens. The calculation for the placement of the toric lens is exceedingly important for the patient´s recovery of vision. Some houses, take date form medical equipment and perform the calculation, but more depends on the experience of the treating physician to ensure the desired effectiveness and efficiency, For this reason, in this investigation, the proposal is presented to perform the calculation, in addition, the approach to simulation of the placement of the lens inside the eye, especially in its axial length and depth in the anterior chamber, determinants for short, medium and long distance with an automatic focus and appropriate, in like manner, the results are exposed to the specialists of the clinic "Santa Lucia" in the city of Quito. © 2020, Springer Nature Switzerland AG.

Valence Tautomerism Induced Proton Coupled Electron Transfer:X−H Bond Oxidation with a Dinuclear Au(II) Hydroxide Complex

We report the preparation and characterization of the dinuclear Au II hydroxide complex Au II 2 (L) 2 (OH) 2 (L= N , N′ -bis (2,6-dimethyl) phenylformamidinate) and study its reactivity towards weak X−H bonds. Through the interplay of kinetic analysis and computational studies, we demonstrate that the oxidation of cyclohexadiene follows a concerted proton-coupled electron transfer (cPCET) mechanism, a rare type of reactivity for Au complexes. We find that the Au−Au σ-bond undergoes polarization in the PCET event leading to an adjustment of oxidation levels for both Au centers prior to C(sp 3 )−H bond cleavage. We thus describe the oxidation event as a valence tautomerism-induced PCET where the basicity of one reduced Au−OH unit provides a proton acceptor and the second more oxidized Au center serves as an electron acceptor. The coordination of these events allows for unprecedented radical-type reactivity by a closed shell Au II complex.

Limited therapeutic options are available to effectively preventing atherosclerosis. Inflammatory endothelial cells, foamy macrophages, and high protease levels contribute to atherosclerotic plaque formation. Studies have shown that catechins effectively scavenge reactive oxygen species (ROS), inhibit monocyte adhesion and reduce cholesterol levels, while nitric oxide (NO) enhances endothelial function. However, due to the poor stability and bioavailability of catechins and the toxicity from the burst release of current synthetic small molecules NO donor, effective delivery of these bioactive compounds to treat atherosclerosis is still a challenge. Herein, a catechin/protein-based NO donor co-delivery nanosystem was designed for combinatorial anti-atherosclerotic therapy. We engineered a (−)-epigallocatechin-3-gallate (EGCG)/NO-releasing protein co-assembled nanocomplex based on specific catechin-protein interactions. Furthermore, the nanocomplex was surface modified with fucoidan (Fu), a sulfated polysaccharide with anti-inflammatory activity. This nanocomplex exhibits sensitivity to ROS, pH, and enzymes. The Fu-functionalized nanoparticles specifically accumulates in atherosclerotic plaques mediated by P-selectin on inflamed endothelial cells and scavenger receptor A (SR-A) on foamy macrophages. Under environmental stimuli that simulate the condition of plaque, the nanoparticles are readily activated to release EGCG and NO in response to excess ROS and high protease levels, exerting the multi-synergistic anti-atherosclerosic effects on reducing monocyte adhesion, promoting NO production to proliferate endothelial cells, lowering ROS levels, and decreasing the foam cell formation in vitro, and reducing lipid accumulation, plaque size, and inflammatory cytokines release in high-fat diet-induced atherosclerosis model in ApoE−/− mice. TITLE: 從加護病房家屬觀點探究重症病情告知的經驗. 背景: 病情告知是減少醫病認知落差的重要步驟，且能減輕家屬心理壓力。現階段研究仍缺乏家屬對重症病情告知經驗及需求之探討。. 目的: 瞭解重症家屬病情告知經驗及過程需求。. 方法: 採描述現象學，立意取樣5位加護病房疾病嚴重度≥ 20分的病人家屬進行深度訪談。以Giorgi現象學分析法，利用Nvivo 11分析歸納資料。. 結果: 整體經驗包含四大主題：（一）訊息聽了無法懂，希望疑問詳細解、（二）無助找尋為解答，期望護理來幫忙、（三）託付專業卻無奈，渴望醫療多些愛、（四）重重難關多牽絆，盼望見解來割斷。. 結論／實務應用: 病情告知著重傳遞結果，家屬無法理解病情變化原因。本研究建議醫療專業人員應體認家屬認知程度，並顧及其情緒反應，納入家屬需求及期待，以病人為中心提供個別化告知。.;BACKGROUND: Truth-telling is an important step toward reducing the cognitive gap between physicians and patients as well as reducing the psychological pressures applied to physicians by family members. There is a lack of research on the truth-telling experience and needs in the intensive care unit from the perspective of patient family members. PURPOSE: This study is designed to explore the experiences and needs of families in the intensive care unit. METHODS: A descriptive phenomenology method was used in this study. In-depth interviews were conducted with five participants who had family members assessed with acute physiology and chronic health evaluation II scores ≥ 20. Data were analyzed using Giorgi's phenomenological methods and Nvivo 11. RESULTS: Four experience themes were examined, including (1) nothing is clear, requires explanation;(2) helpless to find answers, need a nurse to resolve this issue;(3) professional conduct makes us feel helpless, longing for love from the medical team;(4) decisions are very difficult, hoping to get more help. CONCLUSIONS / IMPLICATIONS FOR PRACTICE: The family members expressed that they were unable to understand the underlying causes of the progression in patient condition because the medical team only presented outcomes to the family and did not discuss related causes. Thus, it is recommended that medical teams learn to recognize the cognitive processes of patient family members and consider their emotions, including their needs and expectations, in order to provide individualized explanations based on a patient's status and progress.

Massive usage of antibiotics harms the environment through animals’ excrement and squeezes open-wellbeing issues. Enrofloxacin (EF), an antimicrobial agent, treats bacterial diseases and infections in cattle, poultry and exotic animals and exhibits harmful side effects. The present work proposed a sensitive and rapid electrochemical scaffold based on the Cu-Ni bimetallic-organic framework (Cu-Ni-MOF) for the detection of harmful EF in real-time applications. The Cu-Ni-MOF was synthesized through solvothermal approach with the support of a multifunctional organic ligand, 4,4-biphenyldicarboxylicacid and characterized through various analytical tools. The electrochemical sensor for EF detection was constructed by drop-casting the Cu-Ni-MOF on a glassy carbon electrode and the fabricated sensor exhibited higher catalytic activity when compared to the parental counterparts by exhibiting 1.7-fold higher current for EF oxidation. Further, the fabricated sensor is highly active in the linear range response of 200 nM–2 mM with a limit of detection of 15.17 nM and sensitivity of 638.71 µA mM cm −2 under optimized environments. The higher electrocatalytic activity of the Cu-Ni-MOF electrode synergistically stems from the strong interaction between Cu 2+ of the MOF and pyridone and carboxylate moieties of the EF and facile electron transition facilitated by Ni 2+ . The practicability of the sensor was evaluated in milk, egg and river water samples and the obtained results were validated with Liquid Chromatography-Mass Spectrometric technique. Furthermore, the prepared catalyst was utilized for the photochemical decontamination of EF and the present Cu-Ni-MOF catalyst delivered excellent photocatalytic activity in the decontamination of EF under visible light at ambient temperature.

The Augmented Whole-body Scanning via Magnifying PET (AWSM-PET) device is a novel technology designed to improve the detectability of lesions in whole-body PET/CT imaging. We have developed a prototype AWSM-PET device that consists of two panels of high-resolution time-of-flight (TOF) PET detectors, integrated with the standard readout electronics and event processing architecture of the Biograph Vision (Siemens Healthineers). The system exhibits excellent spatial, energy and timing resolution, with an average coincidence resolving time of $\sim 217$ ps FWHM and an average energy resolution of $\sim 11 \%$ FWHM at 511 keV. Custom firmware and software have been developed to encode the large number of detector elements and to form coincidences between the AWSM-PET detectors and the Biograph Vision scanner. We have pioneered a specialized image reconstruction approach utilizing a GPU-based list-mode framework, which incorporates the system’s point-spread function (PSF) to optimize image quality and spatial resolution. Continuous-bed motion (CBM) whole-body imaging protocols have been implemented for the AWSM-PET operation to improve the uniformity of sensitivity across the patient’s body. Sensitivity enhancement was evaluated using a point source. The first human subject study was a head-andneck cancer patient scheduled for a standard-of-care (SOC) FDG PET/CT scan following treatment. Both the SOC-PET/CT images and the AWSM-PET images were acquired simultaneously for a total of 17 minutes using the CBM mode. Results show high-resolution and high-quality AWSM-PET images. We anticipate to image more patients in the summer, refine AWSM-PET image reconstruction parameters and correction techniques. Comparison between the AWSM-PET and the SOC-PET/CT will be presented at the 2024 IEEE MIC conference.

Calorie restriction (CR) and fasting affect lifespan, disease susceptibility and response to acute injury across multiple animal models, including ischaemic injuries such as myocardial infarction or kidney hypoxia. The cargo and function of circulating extracellular vesicles (EV) respond to changes in host physiology, including exercise, injury, and other interventions. Thus, we hypothesised that EVs induced following CR may reflect some of the beneficial properties of CR itself. In a pilot study, EVs were isolated from mice following 21 days of 30 % CR, and from eight human donors after 72 h water-only fasting. EV size, concentration and morphology were profiled by NTA, western blot and cryoEM, and their function was assessed using multiple assays related to ischaemic diseases. We found that EVs from post-fasting samples better protected cardiac cells from hypoxia/reperfusion (H/R) injury compared to pre-fasting EVs. However, there was no difference when used to treat H/R-injured kidney epithelial cells. Post-fasting derived EVs slowed the rate of fibroblast migration and slightly reduced macrophage inflammatory gene expression compared to pre-fasting derived EVs. Lastly, we compared miRNA cargos of pre- and post-fasting human serum EVs and found significant changes in a small number of miRNAs. We conclude that fasting appears to influence EV cargo and function, with varied effects worthy of further exploration. © 2023 The Authors. The outbreak of pandemics (e.g., severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2 in 2019), influenza A viruses (H1N1 in 2009), etc.), and worldwide spike in the aging population have created unprecedented urgency for developing new drugs to improve disease treatment. As a result, extensive efforts have been made to design novel techniques for efficient drug monitoring and screening, which form the backbone of drug development. Compared to traditional techniques, microfluidics-based platforms have emerged as promising alternatives for high-throughput drug screening due to their inherent miniaturization characteristics, low sample consumption, integration, and compatibility with diverse analytical strategies. Moreover, the microfluidic-based models utilizing human cells to produce in-vitro biomimetics of the human body pave new ways to predict more accurate drug effects in humans. This review provides a comprehensive summary of different microfluidics-based drug sensing and screening strategies and briefly discusses their advantages. Most importantly, an in-depth outlook of the commonly used detection techniques integrated with microfluidic chips for highly sensitive drug screening is provided. Then, the influence of critical parameters such as sensing materials and microfluidic platform geometries on screening performance is summarized. This review also outlines the recent applications of microfluidic approaches for screening therapeutic and illicit drugs. Moreover, the current challenges and the future perspective of this research field is elaborately highlighted, which we believe will contribute immensely towards significant achievements in all aspects of drug development. © 2022 Elsevier B.V.
