MiR-1 Is Regulated by Hydrogen Peroxide via MAPK and Limits Cell Migration and Invasion

MicroRNA-1 (miR-1) is a conserved microRNA that is highly expressed in skeletal and cardiac muscle tissues. Moreover, miR-1 regulates genes and has critical roles in cell migration and invasion. Downregulation of miR-1 has been found in many pathologies of numerous organs, including the lungs. What exactly contributes to the downregulation of miR-1 is not fully understood, and in the present study, we investigated whether ROS regulate miR-1 and its role in cell migration and invasion. A549 cells were grown and maintained in DMEM:F12 (1:1) and supplemented with 10% FBS and 1000 U of Penicillin/Streptomycin and maintained as recommended by the manufacturer (ATCC). Cell migration and invasion, IHC, Western blot, qPCR, ROS, miR-1 transfection, and qPCR were used to determine miR-1 regulation and its role in cell migration. Exogenous miR-1 decreased the formation of ROS and inhibited cell migration and invasion, whereas inhibition of miR-1 increased ROS formation and stimulated cell migration and invasion. Inhibition of miR-1 induced the formation of actin filaments contractile structures, whereas exogenous miR-1 limited the formation of these structures. Hydrogen peroxide significantly decreased miR-1 level, whereas inhibition of Nox4 had no effect on miR-1 level. Alpha amanitin did not decrease miR-1 level, whereas inhibition of NF-кB temporally decreased miR-1 level. This study demonstrates that ROS suppress miR-1 and that miR-1 is posttranscriptionally regulated via MAPK. Endogenous Nox4-dependent ROS are not involved in miR-1 regulation, whereas exogenous ROS regulates miR-1. NF-κB plays a key role in miR-1 regulation in both redox and nonredox environments. Moreover, Mir-1 limits cell migration and invasion even in the presence of ROS. TSP-1 is a major regulator of TGFβ and its expression is upregulated by ROS. Social concern regarding the environment has recently been increasing, and efforts are being made to reduce industrial waste and promote the effective utilization of construction waste. However, considering the discrepancy between the demand and supply of concrete waste, exploring new avenues for its utilization is crucial. In this study, we developed the perfect recycled aggregate concrete composed entirely using by-products and tested its production in a ready-mixed concrete plant for application to pavements. This is the first attempt of its kind in Japan. The results showed that concrete composed of by-products can be shipped and manufactured in actual plants by properly controlling the water absorption of recycled aggregates and establishing an appropriate mix proportion. This study confirmed the applicability of Japan’s first pavement material made exclusively using by-products of the constituent materials.

The COVID-19 pandemic has significantly reshaped global environmental dynamics, with coastal and marine ecosystems among the most affected areas. This adjustment period has also seen a surge in tourist activities in these regions, necessitating advanced strategies for marine debris management (MDM). Our study introduces an enhanced approach to MDM in the post-pandemic era, integrating essential attributes such as asset facilities, 3R flexibility (Reduce, Reuse, Recycle), responsive organizational structures, education on the impact of marine debris, and agency motivation within an adaptive capacity framework. By evaluating marginal willingness to participate (MWTP) across three hypothetical scenarios, our research highlights the comprehensive adaptation of the MDM scenario as the most effective, evidenced by the highest MWTP. This indicates a pronounced community readiness to engage in an integrated MDM approach, crucial in increasing tourist visits and challenging existing MDM frameworks. Our findings illustrate the necessity of an integrated approach to bolster MDM effectiveness in managing the heightened debris load from the post-pandemic resurgence of tourism. The incremental increase in MWTP across scenarios suggests that embracing more inclusive and unified strategies can significantly enhance community participation and the efficacy of MDM practices. © 2024 Elsevier Ltd

Accurate Structural Cracks Detection using NestedUNet from Drone and Handheld Camera Images

Crack detection has been significant in structural health monitoring and maintenance of concrete facilities. Many efforts have been made to automate the crack detection process using convolutional neural networks (CNNs). However, many previous CNN architectures and datasets have limitations that require improvements to reduce training time, GPU requirements, and model generalization capabilities. To mitigate these problems, we have developed a new dataset with different types of complex cracks utilizing a handheld camera and drone in different light, environmental conditions, and varying backgrounds. At the same time, we presented an enhanced CNN encoder-decoder architecture that achieves crack segmentation with higher accuracy with fewer parameters. To cope with spatial information loss effectively, we used nested skip connections in the encoder part, along with the EfficientNet-B7 encoder, for speed, accuracy enhancement, and parameter minimization. In the decoder part, residual convolution blocks are employed to alleviate the vanishing gradient problem and aid spatial information recovery. In addition, we added the convolutional block attention module (CBAM) within our architecture to enhance feature extraction by emphasizing crack details over the background. Experimental results demonstrate that our proposed method achieves a 96.15% accuracy, 85.78% precision, 82.23% recall, and 84.00% F1-score on our custom dataset. Comparisons with other models show that our model outperforms them on all evaluation matrices while having fewer parameters.

The present work investigated color and biochemical oxygen demand (COD) removal from treated landfill leachate via advanced oxidative processes (AOPs) artificially emitted. The AOP H₂O₂/UV, TiO₂/UV, and photolysis were tested in two bench-scale photoreactors: The first one with UV-C lamp and the other with UV-A lamp associated or not with coagulation / flocculation pretreatment. Crude leachate samples with pH ranging from 8 to 3 were used, and time varied in 30, 45, and 60 minutes. Experiments were performed in two stages: step 1 with a 20 mL volume from each sample and step 2 with repetition of the best results from the previous step, adopting the 150 mL volume. In step 1, the AOP applied in the crude leachate sample showed the best results, standing out H₂O₂/UV-C with 30 minutes and TiO₂/UV-A with 60 minutes. In step 2, H₂O₂/UV-C had a 60% color removal and 25% COD reduction, while TiO₂/UV-A had a 10% color removal and 20% COD reduction. Therefore, the H₂O₂/UV-C process was the most efficient, because the complex characteristics of the effluent interfered in the TiO₂/UV-A efficiency, but it is necessary to eliminate the process interferences. The use of artificial radiation is a viable alternative;however, it can be costly, being relevant in the associations between processes with artificial UV and solar UV, as the natural radiation becomes more attractive and allows the process operating costs reduction. © 2020 Wiley Periodicals, Inc.

Mississippi Burning: An Exploration of Community Noise Levels from a Rural Mississippi Community Grappling with Wood Pellet Production Manufacturing

Wood pellets are being touted as a renewable and sustainable form of energy. As a result of this growing popularity, the global wood pellet market is expected to reach $16 billion by 2026. Currently, there are approximately 100 pellet manufacturing plants in the United States, with most of the production occurring in predominantly Black and poor communities in the rural South. At the request of residents concerned about the noise pollution emanating from wood pellet production plants in their community, we carried out a community noise exposure assessment. From August 2023 to March 2024, and working closely with a local community organization we measured a variety of noise metrics in Gloster, a wood pellet impacted city in rural Mississippi, comparing these noise pollution levels to a background community with no wood pellet industrial activity. We observed statistically significant higher levels of A-weighted, C-weighted, and Z-weighted sound in Gloster compared to Mendenhall. These differences persisted across different times of the day and days of the week. When examining the decibel levels for each of the center frequencies of the 1:3 octave band, this pattern persisted. Our preliminary findings suggest that a rural community’s soundscape can be negatively impacted by unchecked wood pellet manufacturing. We argue that the sound level differences in communities impacted by wood pellet manufacturing and those that are not, combined with the strategic placement of wood pellet plants in vulnerable communities in the rural south, may also be an environmental justice issue. © 2024, The Authors. Water and energy shortages came due to rapid population growth, living standards and rapid development in the agriculture and industrial sectors. Desalination tends to be one of the most promising water solutions;however, it is a process of intense energy. Membrane Capacitive Deionization (MCDI) has received considerable interest as a promising desalination technology, and MCDI research has increased significantly over the last 10 years. In addition, there are no guidelines for the design of Capacitive Deionization (CDI) implementation strategies for individual applications. This study, therefore;provides an alternative of CDI’s recent application developments, with emphasis placed on hybrid systems to address the technological needs of different relevant fields. The MCDI’s energy consumption is compared with the reverse osmosis literature data based on experimental data from laboratory-scale system. The study demonstrates that MCDI technology is a promising technology in the next few years with an extreme competition in water recovery, energy consumption and salt removal for reverse osmosis.

This paper proposes a two-dimensonal positioning method based on a hidden Markov model map-matching scheme. The states of the hidden Markov model are generated by dividing the area of interest into a grid. At each time instant, the method considers two types of measurements: the platform’s heading and the two-dimensional distance between the platform and the single-anchor. A recursive Bayesian estimator exploits these measurements to estimate the platform’s position. The platform’s heading measurement is used to calculate the prior probability distribution. Following this, observation likelihood is computed by considering the two-dimensional distance measurement as the observation of the hidden Markov model. Finally, the most probable projection of these measurements on the states of the hidden Markov model is estimated as the platform’s position. The proposed method can be efficiently used, especially in constrained indoor and outdoor environments. Moreover, the method provides a two-dimensional positioning solution with an increased robustness thanks to the bounded error on the distance measurements. Simulation studies are provided to demonstrate the effectiveness of the proposed method.

Wood pellets are increasingly promoted as a renewable and sustainable energy source, driving the global wood pellet market to a projected value of $16 billion by 2026. However, the rapid expansion of this industry disproportionately affects predominantly Black and low-income communities in the rural South, where approximately 100 pellet manufacturing plants are currently located in the United States. From August 2023 to March 2024, we conducted a noise exposure assessment in Gloster, Mississippi, a community affected by pellet production, and compared it to Mendenhall, a nearby community without industrial activity. In collaboration with a local organization, we measured noise metrics and found significantly higher A-weighted, C-weighted, and Z-weighted sound levels in Gloster, along with elevated decibels at multiple center frequencies. These findings suggest that wood pellet manufacturing can severely alter the soundscape of rural communities, raising environmental justice concerns. Our findings underscore the importance of investigating the cumulative health impacts of noise and other environmental burdens on these vulnerable communities to better inform policy. © 2024

The importance of an accurate PNT (Position, Navigation, Timing) information is nowadays consolidated in the navigation field. Easy of use and robustness make Single Point Positioning (SPP) one of the most used techniques to compute navigation information, on the other hand, employing SPP may not lead to the achievement of the required performance, especially in challenging scenario. Different techniques or services could be used in order to achieve the target performance; in particular, the recent Galileo High Accuracy Service (HAS) could enhance the position accuracy. Several applications can benefit of HAS in particular geomatics, automotive, aviation, and maritime. This novel service provides corrections for orbits, code, phase, and satellite clock for Galileo and GPS (Global Positioning System) systems. Corrections are broadcast through the Galileo E6-B signal or other means (i.e., internet), and they have a global coverage. Even if HAS corrections are meant for the PPP (Precise Point Positioning) technique, in this study, they are applied to the SPP technique in order to assess the possible improvements in solution accuracy.
