The world is encountering a global threat: Climate change. It is estimated that the average air temperature will raise 3.7-7.8 °C by 2100. The possible impacts will vary, however, agriculture will be adversely affected marked with yield loss of staple crops such as rice and maize and altered post-harvest quality of horticulture products. On the other hand, the world food and Agriculture Organization (UN-FAO) has set a target 'zero hunger' by 2030', despite the five big challenges ahead: I) protracted conflict, ii) growing inequality, iii) rapid urbanization, iv) economic challenges, v) drought and natural disasters. The existing plant genetic resources, particularly the wild, neglected, and underutilized species has a great role and bears big potential to be exploited and targetted to achieve food and nutrition security, esp. in the densely populated regions with malnutrition problem e.g. in Asia and sub-Saharan regions. However, a key to future and sustainable crop production lies in the collection of crop and their wild relatives stored in ex-situ, or in-situ. This paper aims to explain the importance of plant genetic resources as well as the potential of some underutilized crops closely attributed with drought resistence, such as sorghum (Sorghum bicolor L. Moench), amaranth (Amaranthus sp.). These crops bear potential to support national food security program. © Published under licence by IOP Publishing Ltd.

Naja atra, the Chinese cobra, is a major cause of snake envenomation in Asia, causing hundreds of thousands of clinical incidents annually. The current treatment, horse serum-derived antivenom, has unpredictable side effects and presents manufacturing challenges. This study focused on developing new-generation snake venom antidotes by using microbial phage display technology to derive nanobodies from an alpaca immunized with attenuated N. atra venom. Following confirmation of the immune response in the alpaca, we amplified VHH genes from isolated peripheral blood mononuclear cells and constructed a phage display VHH library of 1.0 × 107 transformants. After four rounds of biopanning, the enriched phages exhibited increased binding activity to N. atra venom. Four nanobody clones with high binding affinities were selected: aNAH1, aNAH6, aNAH7, and aNAH9. Specificity testing against venom from various snake species, including two Southeast Asian cobra species, revealed nanobodies specific to the genus Naja. An in vivo mouse venom neutralization assay demonstrated that all nanobodies prolonged mouse survival and aNAH6 protected 66.6% of the mice from the lethal dosage. These findings highlight the potential of phage display-derived nanobodies as valuable antidotes for N. atra venom, laying the groundwork for future applications in snakebite treatment. Background Chlortetracycline (CTC) belongs to the class of tetracycline’s compounds, which is known for its antibacterial activity. CTC was largely consumed by farm animals than humans, which leads to the growth antibiotic – resistant bacteria and environmental contamination. So far no one reported the electrochemical detection and determination of CTC by using AuNPs@BiVO4 composite. Some reports have been reported using conventional chromatography which is most expensive, time-consuming and highly equipped method. Results Here, we report a simple method for preparing gold nanoparticle decorated bismuth vanadate (AuNPs@BiVO4), which was dropped cast on a screen-printed carbon electrode (SPCE). The AuNPs@BiVO4 and AuNPs@BiVO4/SPCE materials were characterized using TEM, SEM-EDX, XPS, DLS, ELS, EIS, and CV. The as-prepared AuNPs@BiVO4/SPCE was used to detect and determine chlortetracycline (CTC) by different pulse potentials in phosphate buffer pH 7.0. The electrode possessed a wide linear range of 0.99 – 19.06 µM, with obtained LOD = 0.15 µM, LOQ = 0.49 µM, and sensitivity = 0.059 μA μM-1 cm-2. It also exhibited good selectivity, stability, and repeatability, recovering 87.14% - 100% for detecting CTC in Mili Q and tap water. Significance Facile and rapid synthesis of AuNPs@BiVO4 nanocomposite film modified electrode at physiological pH conditions has been prepared and A highly electrochemical detection and determination of CTC using AuNPs@BiVO4 nanocomposite film has been developed for the first time. Thus, the proposed electrode could be applied for the detection and determination of CTC in water sources and has the potential to be used for real sample analysis. © 2024, The Authors. Background: Liposomal bupivacaine (LB) is a relatively new formulation that slowly releases bupivacaine to extend its efficacy for 72–96 h. It is inconclusive whether LB offers better efficacy than traditional periarticular injection (TPAI) following total knee arthroplasty (TKA). Methods: Relevant randomized controlled trials (RCTs) were searched using electronic databases, including PubMed, Cochrane Library, EMBASE, and Web of science. Review Manager 5.4.1 was used for calculations. Results: Sixteen RCTs were included in this meta-analysis. LB had better effects on morphine consumption equivalents during postoperative 24–48 h than TPAI. No significant difference was observed in pain relief, incidence of nausea and vomiting, or length of hospital stay between the two groups. Conclusion: LB administration during TKA is not superior to TPAI. Studies with larger sample size are needed to validate our findings. Mapping of rain water harvesting potential at Keduang Sub-watershed, Central Java, Indonesia

One of the main issues in agriculture in this climate change era is the water crisis. It caused a drought that affected the delay of rice cultivation. A study found that Wonogiri Regency has very critical with a value of 163.1%. Hence, it is necessary to have an alternative solution to overcome it. In this case, utilizing rain by harvesting rainwater (RWH) can be used as an alternative to meet water needs during the dry season and for various other uses. One of the RWH applications is a small-farm reservoir (SFR). To identify the potential locations for RWH is using location suitability modelling based on GIS. The determination of the location depends on biophysical criteria such as rainfall, slope, soil texture, stream order, and land use. The socio-economic attributes consist of the distance to the river, road, settlement, and agriculture. To obtain the suitability location, after all of the maps are generated, next is reclassify the value to 0 and 1. In the end, the maps calculate with a raster calculator to define the suitability location that has value 1. Location and dimension details can find out. This study aims to determine the potential of RWH for agriculture (in specific, SFR) at Keduang Sub-watershed as a consideration for stakeholders' decision. The results showed 226 areas are potential for SFR implementation and will make it easier for stakeholders to decide by comparing the suitability location map and the long-term strategic map.

Natural waterways near urban areas are heavily impacted by anthropogenic activities, including their microbial communities. A contaminant of growing public health concern in rivers is antibiotic resistant genes (ARGs), which can spread between neighboring bacteria and increase the potential for transmission of AR bacteria to animals and humans. In order to identify the matrices of most concern for AR, we compared ARG burdens and microbial community structures between sample types from the Scioto River Watershed, Ohio, United States from 2017-2018. Five environmental matrices (water, sediment, periphyton, detritus, and fish gut) were collected from 26 river sites. Due to our focus on clinically relevant ARGs, three carbapenem resistance genes (blaKPC, blaNDM, and blaOXA-48) were quantified via DropletDigitalTM PCR. At a subset of nine urbanized sites, we conducted16S rRNA gene sequencing and functional gene predictions. Carbapenem resistance genes were quantified from all matrices, with blaKPC being the most detected (88% of samples), followed by blaNDM (64%), and blaOXA-48 (23%). Fish gut samples showed higher concentrations of blaKPC and blaNDM than any other matrix, indicating potential bioaccumulation of ARGs, and risk of broader dissemination through aquatic and nearshore food webs. Periphyton had higher concentrations of blaNDM than water, sediment, or detritus. Microbial community analysis identified differences by sample type in community diversity and structure. Sediment samples had the most diverse microbial communities, and detritus, the least. Spearman correlations did not reveal significant relationships between the concentrations of the monitored ARGs and microbial community diversity. However, several differentially abundant taxa and microbial functions were identified by sample type that are definitive of these matrices’ roles in the river ecosystem and habitat type. Cordyceps militaris (C. militaris) is a unique medicinal fungus of the genus Cordyceps. It has high economic value due to various biological functions from the extracellular polysaccharides (EPSs) it produces. The aim of this study was to establish a mycelium-immobilization system for EPS production in a bubble column system. First, EPS production was optimized by 90.2% (from 0.72 to 1.37 g/L) using response surface methodology under an alginate-immobilized mycelium system with 6 days of cultivation. The highest EPS production was obtained (5.3 g/L) in a flask system through elongation to 15 days of cultivation. EPS production subsequently further increased to 10.42 g/L utilizing a bubble column reactor. Furthermore, the system can be reused for at least three rounds. In the EPS characteristics analysis, the produced EPSs were mainly composed of glucose and galactose. They presented strong antibacterial activity against Staphylococcus aureus. In terms of antioxidant activity, respective the EPS concentration that caused 50% scavenging of DPPH and ABTS free radicals values (IC50) were 4.65 and 7.92 mg/mL. In summary, these findings should be helpful for establishing a suitable industrial fermentation system for C. militaris and can be studied in the food and medical industries in future work. © 2024 The Authors

In Brazil, the use of Eucalyptus is focused on the production of wood or pulp for the paper industry but without any general recovery of waste, with leaves and branches being left on the ground. One possibility is to use these residues as raw materials in the production of industrially relevant and value-added compounds such as essential oil. The aim of the present study was to investigate the chemical composition, yield, anti-inflammatory/antinociceptive activities, and acute toxicity in mice, as well as the antimicrobial effects of essential oils from the leaves of 7 varieties of Eucalyptus and hybrids against Escherichia coli , Staphylococcus aureus, and Candida albicans . The extraction of oils was carried out using hydrodistillation, and they were analyzed by gas chromatography coupled to mass spectrometry. Urocam and Grancam were the plants that obtained the highest oil yield, with yields of 3.32 and 2.30%, respectively. The main chemical components identified in these plants were 1.8 cineole and α -pinene. The antinociceptive effect of the 7 oils (50 mg/kg, p.o.) was initially assessed in the acetic acid-induced writhing test. In this assay, a significant ( p < 0.05) antinociceptive/anti-inflammatory effect was observed from 4 tested essential oils ( E. benthamii, E. saligna, and the hybrids Urocam and Grancam) when compared to the vehicle-treated group. This effect was then confirmed in the formalin-induced paw licking test. No toxicological effects or alterations were observed in motor coordination after the administration of the studied oils to the animals. In the antimicrobial evaluation, the seven essential oils inhibited the growth of S. aureus, E. coli , and C. albicans at different concentrations. Collectively, these results demonstrate that the essential oil from the leaves and branches of Eucalyptus species and varieties present potential biomedical applications and represent a source of antimicrobial and/or anti-inflammatory compounds.

Grapevine (Vitis vinifera L.) is a plant containing many phenolic compounds, mostly distributed in the peel, pulp, and seeds. This study evaluates the centesimal composition and bioactive compounds in Shiraz grape (Vitis vinifera) peels using spectrophotometric and UHPLC techniques and develops different formulations of compound powders from the peels and arrowroot using conventional drying technology. The results demonstrate that Shiraz grape skin contains significant amounts of insoluble fiber (15.3%), phenolics (157.09 ± 6.96-149.11 ± 9.27 mg GAE g-1), and flavonoids (0.75 ± 0.50-2.00 ± 0.50 mg QE g-1), with excellent antioxidant capacity observed in the alcoholic extracts. The phenolic content in the developed powdered compounds ranged from 128.32 to 139.70 mg GAE g-1. In general, the compounds showed good antioxidant capacity (IC50 = 0.17 to 0.19 μg mL-1). According to the chromatographic evaluation, it was possible to quantify gallic acid, catechin, and epicatechin, the latter of which was found in the largest quantities in the six formulations. The EV5 formulation was the most efficient in terms of phenolic compounds and protein amounts.
