Luwak coffee is one of the Indonesia traditional coffee that known globally because of its unique aroma and flavor. Luwak coffee was produced by a fermentation process in luwak digestion by involving microbe that can be found in luwak digestion. Luwak coffee fermentation process using luwak animal has many weakness such as inefficient production processes, and final product quality that always makes consumers hesitate in terms of its hygiene and halal. Therefore some research to produce coffee with the quality of luwak coffee without using luwak began to be developed. Several methods of fermentation to produce coffee with quality luwak coffee are discussed in this paper, such as fermentation by in vitro technique

This review presents a comprehensive perspective on the genomic surveillance of SARS-CoV-2 in Taiwan, with a focus on next-generation sequencing and phylogenetic interpretation. This article aimed to explore how Taiwan has utilized genomic sequencing technologies and surveillance to monitor and mitigate the spread of COVID-19. We examined databases and sources of genomic sequences and highlighted the role of data science methodologies in the explanation and analyses of evolutionary data. This review addressed the challenges and limitations inherent in genomic surveillance, such as concerns regarding data quality and the necessity for interdisciplinary expertise for accurate data interpretation. Special attention was given to the unique challenges faced by Taiwan, including its high population density and major transit destination for international travelers. We underscored the far-reaching implications of genomic surveillance data for public health policy, particularly in influencing decisions regarding travel restrictions, vaccine administration, and public health decision-making. Studies were examined to demonstrate the effectiveness of using genomic data to implement public health measures. Future research should prioritize the integration of methodologies and technologies in evolutionary data science, particularly focusing on phylodynamic analytics. This integration is crucial to enhance the precision and applicability of genomic data. Overall, we have provided an overview of the significance of genomic surveillance in tracking SARS-CoV-2 variants globally and the pivotal role of data science methodologies in interpreting these data for effective public health interventions.

Furanoids are a class of contaminants prevalent in both airborne and occupational environments, with potential health implications through inhalation, oral ingestion, and skin penetration. Given their diminutive molecular size, there is a presumption that furanoids can readily permeate the skin. To systematically explore this presumption, we investigated the skin absorption and toxicity of a series of furans (furfuryl alcohol, furfuryl acetate, furfural, methyl 2-furoate, and 5-methylfurfural) using in silico, in vitro, and in vivo models. The in vitro permeation test (IVPT) from neat and aqueous suspension (5 mM) of furans demonstrated a facile absorption through pig and nude mouse skins. The lipophilicity of furans significantly influenced skin deposition, with higher lipophilicity displaying greater deposition. However, an opposing trend emerged in the receptor compartment accumulation. In barrier-defective skin simulating atopic dermatitis (AD) and psoriasis, enhanced deposition occurred with more hydrophilic furans but not with the more lipophilic ones. In the cell-based study, furanoids induced the proliferation of keratinocytes and skin fibroblasts except for the compounds with the aldehyde group (furfural and 5-methylfurfural). Both furfuryl acetate and 5-methylfurfural activated keratinocytes via the overexpression of COX-2 and PGE by 1.5‒2-fold. This stimulation involved the mitogen-activated protein kinase (MAPK) signaling pathway. For the in vivo mouse skin treatment, we selected furfuryl acetate (hydrophilic) and 5-methylfurfural (lipophilic). Both furans showed different patterns of skin lesions, where repeated application of furfuryl acetate caused epidermal hyperplasia and scaling, while 5-methylfurfural predominantly evoked skin inflammation and barrier disintegration. Cow's milk-intolerance is a digestive problem on people who not able to digest milk. This problem may relate to the variant of (3-casein (CSN2), especially A1, suggested due to (3-casomorphins (BCM-7) formation during enzymatic digestion, for that selecting cattle free BCM-7 become a concern to produce digestive friendly milk. This study aimed to differentiate A1 and A2 allele variant of CSN2 gene in selected population of Indonesian Holstein cattle. In total 70 cows DNA were collected, and fragment of CSN2 exon 7 which contain Single Nucleotide Polymorphism (SNP) rs43703011 and rs43703013 were amplified. Variant analysis was done by mutation site analysis using PCR-Restriction Fragment Length Polymorphism (PCR-RFLP) with Mspl restriction enzyme and DNA sequence for confirmation. Result shows 2 allele variants of (3-casein that are B type, representing A1 family variant, and A2 in mutation site rs43703013C>G. We found A2 allele in the studied population is superior in frequency than A1 (0.916 vs. 0.084). Of that, 8.6% cattle were heterozygotes that is BA2 and 91.4% were homozygotes A2A2. Cattle which carry A1 allele variant should be excluded from dairy cattle breeding program for further milk production free of BCM-7.

Fish and seaweed (Eucheuma cotonii) meatball for reducing constipation effect

Functions of dietary fiber include increasing stool mass and reducing intestinal transit time which can be used to prevent constipation. This study aims to determine the ability of mixed catfish meatballs and sea grass flour to reduce constipation through two stages. The first stage was to get the best meatball quality (ratio of tapioca composition and seaweed flour manipulated at 100%: 0%; 90%: 10%; 80%: 20%; 70%: 30%) followed by the second stage which was in vivo testing for rat with doses of meatballs manipulated at 2 g, 4 g, 6 g and 8 g. Research data were analyzed with one-way variance analysis with a confidence level of 95%. Composition of tapioca and seaweed flour at 70%: 30% was the best fish meatball formula resulting in these data: 64.90% moisture content, 2.00% ash, 5.76% fat, 13.91% protein, 13.43% carbohydrate, total food fiber 5.94%) with sensory value: appearance 8.67 (smooth, not hollow, and bright surface), flavor 7.00 (less fish flavor), taste 6.67 (less meat fish taste), texture 9 (solid, compact, springy). The in vivo test showed that administration of 2 g / 200 g weigh body of rat was able to produce rat feces back to normal condition (47.93% moisture content) with a frequency of defecation 21 times per day (no constipation any more).

Rodent tuber is a potential plant which can be developed as an anticancer drug. The rodent tuber plant has a low anticancer compound which does not have an economic value to be utilized on an industrial scale, particularly to explore as an anticancer drug. The material plant was used 30 clones of Rodent Tuber Pekalongan to increase its bioactive compounds using a combination of in vitro culture with gamma-ray irradiation. The selection process has been done until the 8th generation and obtained 14 somaclonal mutants that have higher bioactive compounds than control plant (non-mutant). This study aimed to analyze the genetic variation of 14 somaclonal mutants of rodent tuber using RAPD with 14 primers. The results were produced 513 total bands with the size of 200-5000 bp and 380 polymorphic bands between somaclonal mutants. OPB-18 primer produced the most polymorphic bands and five specific polymorphic bands of 1336 bp, 1070 bp, 901 bp, 861 bp and 728 bp in the somaclonal mutant M23 (20-4-2-1-1-1). OPD-20 primer produced seven specific polymorphic bands of 1574 bp, 1557 bp, 1501 bp, 1496 bp, 1234 bp, 1229 bp and 1086 bp in the M23 somaclonal mutant. The phylogenetic analysis showed that there were two main groups with the coefficient similarity between 0.77 and 0.83. The highest genetic variation obtained was found in the M23 somaclonal mutant with a genetic difference of 23% compared with control. There are five somaclonal mutants (M24, M22, M21, M16, M14) that have a genetic variation of up to 22% over the control. This study shows that OPB-18 and OPD-20 primers were efficient in detecting somaclonal mutant variants. © 2018 Institute of Physics Publishing. All rights reserved.
