β-Fructofuranosidase and β -D-Fructosyltransferase from New Aspergillus carbonarius PC-4 Strain Isolated from Canned Peach Syrup: Effect of Carbon and Nitrogen Sources on Enzyme Production

β-fructofuranosidase (invertase) and β-D-fructosyltransferase (FTase) are enzymes used in industrial processes to hydrolyze sucrose aiming to produce inverted sugar syrup or fructooligosaccharides. In this work, a black Aspergillus sp. PC-4 was selected among six filamentous fungi isolated from canned peach syrup which were initially screened for invertase production. Cultivations with pure carbon sources showed that invertase and FTase were produced from glucose and sucrose, but high levels were also obtained from raffinose and inulin. Pineapple crown was the best complex carbon source for invertase (6.71 U/mL after 3 days of cultivation) and FTase production (14.60 U/mL after 5 days of cultivation). Yeast extract and ammonium chloride nitrogen sources provided higher production of invertase (6.80 U/mL and 6.30 U/mL, respectively), whereas ammonium nitrate and soybean protein were the best nitrogen sources for FTase production (24.00 U/mL and 24.90 U/mL, respectively). Fermentation parameters for invertase using yeast extract were Y_P/S = 536.85 U/g and P_P = 1.49 U/g/h. FTase production showed values of Y_P/S = 2,627.93 U/g and P_P = 4.4 U/h using soybean protein. The screening for best culture conditions showed an increase of invertase production values by 5.10-fold after 96 h cultivation compared to initial experiments (fungi bioprospection), while FTase production increased by 14.60-fold (44.40 U/mL) after 168 h cultivation. A. carbonarius PC-4 is a new promising strain for invertase and FTase production from low cost carbon sources, whose synthesized enzymes are suitable for the production of inverted sugar, fructose syrups, and fructooligosaccharides. © 2019 Gustavo Carvalho do Nascimento et al.

The aim of this work was to perform analysis of the a-tocopherol content by HPLC in Amazonian fruits consumed in Roraima state, Brazil, namely: ata brava, açaí, bacaba, buriti, buritirana, yellow murici, red murici, piaçaba braba, pupunha varieties red and yellow and tucumanzinho. A high concentration of the vitamin in the açaí pulp (1101.11 mg L-1) and of the ata brava seeds (644.27 mg L-1) was observed, but other fruits such as buritirana pulp (166.34 mg L-1), red murici (179.52 mg L-1), and piaçaba brava (8.77 mg L-1) presented low amounts of a-tocopherol. As for the yellow and red varieties of pupunha and the tucumanzinho, no presence of the isomer was detected. Identification and quantification of a-tocopherol cited in this work are unpublished for the ata brava, buritirana and red murici, as for the other fruits studied there were variations. Amazonian fruits may present a-tocopherol in its lipid composition, which is one of the most bioactive isomers of vitamin E, for this reason. Copyright © 2018, AIDIC Servizi S.r.l.

Fingerprints are unique to each individual who contributes to human identification in forensic cases. In this sense, the present work aimed to synthesize 1H-pyrazol-1-yl-4-phenylthiazoles and apply them as powder agents to reveal latent finger marks as an alternative and low cost related to powders found on the market. The compounds were characterized by infrared; fluorescence; 1H NMR; 13C NMR; laser diffractometer and SEM. In addition, cytotoxicity was also evaluated in oral keratinocytes showing high cell viability. The result of the hemolytic assay revealed that compound 2a does not present red cell cytotoxicity. The prepared powders were applied to fingermarks deposited on different surfaces providing quality visualization at level three so that human identification is possible.

Objectives: The 12-item Self-Compassion Scale–Short Form (SCS–SF) is a widely used instrument for the assessment of self-compassion. To date, there have been few examinations of this instrument’s psychometric properties, particularly across nations and languages. Therefore, we used data from the Body Image in Nature Survey (BINS) to assess measurement invariance of the SCS–SF across nations, languages, gender identities, and age groups. Methods: Participants (N = 56,968) from 65 nations completed the SCS–SF in 40 languages. Using these data, we tested various hypothesised models of the SCS–SF in the total sample and, using multi-group confirmatory factor analysis, tested for invariance of the optimal model across national groups, languages, gender identities, and age groups. Results: In the total dataset, we found that an 11-item, 2-factor model (i.e., SCS-11) provided best fit to the data, with the two factors tapping distinct constructs of compassionate and uncompassionate self-responding. The SCS-11 was found to be partially scalar invariant across national groups and languages, and fully scalar invariant across gender identities and age groups. There was wide variation in latent means for the two factors, particularly across national groups and languages. Further analyses showed negligible associations between the two factors and sociodemographic variables, including marital status, financial security, and urbanicity. Conclusions: Our results suggest that it may be possible to derive a stable 2-factor model of the SCS–SF for use in cross-cultural research, but also highlight the likelihood of cross-national and cross-linguistic variations in the way that self-compassion is understood. © The Author(s) 2025.

The aims of this study were to perform a detailed analysis of the chemical composition by gas chromatography, cinematic viscosity in function of temperature by viscometer Schott AVS 350, and biological tests of abóbora (Cucurbita argyrosperma) seed oil, collected in Mucajai city, Roraima state, Brazil. Thus, the abóbora seed samples were manually submitted to the oven at 50 °C for 72 hours, and crushed and sieved between 20-40 Mesh. The seed oil extraction was carried out in a Soxhlet apparatus with hexane for 3 hours, providing an oil yield of 29.8%. In the results were identified a total of 8 fatty acids by gas chromatography;of these 33.8% are saturated fatty acids and 65.7% are unsaturated fatty acids: palmitic (21.5%), stearic (11.6%), oleic (37.0%), linoleic (28.1%) and linolenic (0.3%). In the biological test, the oil was more active against Staphylococcus aureus (29.01%), Listeria monocytogenes (34.56%), Pseudomonas aeruginosa (23.86%) and Citrobacter freundii (32.74%), which features a characteristic low therapeutic index for these bacteria. The kinematic viscosity of the oil reduces in function of temperature (298.15 to 353.15 K) in an exponential curve, demonstrating to refer to a Newtonian fluid. In this work, nine fruits cultivated in the northern Amazon were studied: abiu (Pouteria caimito), acerola (Malpighia emarginata), araçá (Psidium cattleianum), bacuparí (Rheedia gardneriana), biribá (Rollinia mucosa), caçarí (Myrciaria dubia) (Annona squamosa), graviola (Annona muricata) and taperebá (Spondias mombin L.). The total phenolic compounds were evaluated in the pulp, seed and bark by means of the colorimetric reaction of Folin Ciocateau, as well as the antioxidant capacity in the different extracts. DPPH method and on the other hand by the iron reduction method. As the fruits that presented a greater quantity of phenolic compounds are in mg gallic acid. 100g-1 sample, we have: bark of camu camu (1241.1 ± 12.04), followed by the abiu bark with (1132,43 ± 8,10), araçá pulp (1080,21 ± 1,1) and Pulp of acerola (1071.4 ± 22.2). Evaluating the antioxidant capacity, the evaluated fruits that present a higher value of antioxidant capacity are the Araçá seed with EC50 value of (471,23 ± 21,23 g / g DPPH) and for iron reduction of (57,21 ± 4.11 mmol Fe2SO4 / g), followed by the EC50 of abiu bark (521.71 ± 1.34 g / g DPPH) and iron reduction of 411.43 ± 27.12 µmol Fe2SO4 / g), and for the camu-camu pulp (549.24 ± 21.13 g / g DPPH) and for reduction of iron (235.47 ± 11.44 mmol Fe2SO4 / g). Multivariate analysis methods were applied through Principal Component Analysis (PCA) with pulp having the highest correlation between data variability with 93.6% according to PCA. The Effect of Guava on the Improvement of Lipid Profile in Hypercholesterolemic Rats

Red guava is a fruit species with high vitamin C content and the highest fibers, in particular, soluble fiber (pectin) among tropical fruits and cereals. The soluble fiber is hypocholesterolemic in the body. Therefore, it has the potential to reduce cholesterol content. This study aimed to assess the potential of red guava in reducing cholesterol content in hypocholesterolemic rats. This study applied a pure experiment using a randomized pretestposttest group design on hypocholesterolemic Sprague Dawley rats. The rats were fed with 0.72 g of red guava and pectin supplement as a comparison. The total cholesterol content, LDL, HDL and triglyceride were measured by CHOD-PAP and GPO-PAP methods. Data analysis used t-test and ANOVA with significance rate of 0.05. Red guava intake to the hypocholesterolemic rats significantly reduced the contents of: total cholesterol to 57.8 mg/dL (32%), LDL to 25.5 mg/dL (43%), triglyceride to 19.7 mg/dL (18%). Conversely, it significantly increased HDL content to 10.6 mg/dL (18%). The red guava has proven to give hypocholesterolemic effect by reducing the total cholesterol LDL and triglyceride; Conversely, it significantly increased HDL.
