Short communication – The first inventory of the nest placement of horseshoe crab (Tachypleus gigas) in Indonesia

Previous studies have reported that extant horseshoe crabs, including Atlantic and Asian horseshoe crab, spawned in intertidal zone during high tide season. Limited information about spawning nest of the horseshoe crab especially Tachypleus gigas in Indonesia has not been known yet. Moreover, according to the IUCN, the conservation status of T. gigas is still data deficient. During the fieldwork, we found the location that indicated as the spawning ground or nest placement of T. gigas. This is the first time when the spawning area of horseshoe crab is found in Subang, Indonesia. There were three clutches of eggs and the total number was 375-534 eggs in varying depth (5-20 cm) below the sand. The second observation in the same location on January 16, 2020, found a pair of T. gigas laying their eggs and the number was 244 eggs in varying depth (5-15 cm). The characteristics of nest placement of T. gigas that found in Subang was located on the small island in the middle of the sea. The island is an uninhabited island that has mangrove area and sandy substrate. Thus, based on what was found Burung island indicated as the spawning ground of coastal horseshoe crab (T. gigas).

The first prototype of LST (LST-1) for the Cherenkov Telescope Array has been in commissioning phase since 2018 and already started scientific observations with the low energy threshold around a few tens of GeV. In 2021, LST-1 observed BL Lac following the alerts based on multi-wavelength observations and detected prominent gamma-ray flares. In addition to the daily flux variability, LST-1 also detected sub-hour-scale intra-night variability reaching 3–4 times higher than the gamma-ray flux from the Crab Nebula above 100 GeV. Direct inhibitors of M. tuberculosis enoyl-acyl carrier protein reductase (M. tuberculosis InhA) remain effective against variants with mutations associated with isoniazid resistance. In our previous study, structure-based virtual screening was employed to discover such inhibitors. However, most identified hits exhibited limited antimycobacterial activity, with minimum inhibitory concentration (MIC) values of >100 μg/mL. To address this challenge, we refined our virtual screening strategy by integrating ligand- and structure-based virtual screening approaches. The efficacy of this hybrid virtual screening approach was validated through biological assays measuring MIC and half-maximal inhibitory concentration (IC) for the inhibition of M. tuberculosis growth and InhA activity, respectively. Among 14 identified hits, compounds 3 and 10, classified as dipyrazole carboxamide derivatives, were validated as promising lead candidates, with MIC values of 25 and 50 μg/mL and IC values of 10.60 ± 0.56 and 5.08 ± 0.30 μM, respectively. The relatively low hit-to‑lead conversion rate (14 %) is ascribed to our observation that nine of the identified hits, including compounds 3 and 10, showed some level of precipitation in the MIC assay medium. Molecular dynamics simulations show that the dipyrazole carboxamide moiety in compounds 3 and 10 forms essential hydrogen bonds with nicotinamide adenine dinucleotide (oxidized form) (NAD) in the InhA binding pocket. Notably, both compounds 3 and 10 exhibit favorable safety profiles, with no toxicity observed in Caco-2 cells at concentrations up to 100 μg/mL. Consequently, we believe that these compounds present promising starting points for further lead optimization and development of novel antitubercular agents.

We have presented ‘trans-omics’, a discipline that mechanistically connects multiple omic layers. The relationships between molecules of distinct omic layers are governed by reaction rate equations such as v = k [S] (v: reaction rate;k: rate constant;[S]: concentration of molecule S). However, because of the limited availability of the reaction rate data for the left-hand side of the equation, many studies use changes in the amount of molecules in the right-hand side as a surrogate for the reaction rate. Recent advances in comprehensive measurement technologies enable to measure reaction rates in several omic layers. This allows attributing trans-omic network reconstruction to model selection problem of differential equation-based models. Thus, rate-oriented trans-omics enables more mechanistic network reconstruction, instead of an empirical network reconstruction that uses concentrations as readouts of reaction rates. © 2019 The Authors

Tree growth synchrony serves as a valuable ecological indicator of forest resilience to climate stress and disturbances. However, our understanding of how increasing temperature affects tree growth synchrony during rapidly and slowly warming periods in ecosystems with varying climatic conditions remains limited. By using tree-ring data from temperate broadleaf (Fraxinus mandshurica, Phellodendron amurense, Quercus mongolica, and Juglans mandshurica) and Korean pine (Pinus koraiensis) mixed forests in northeast China, we investigated the effects of climate change, particularly warming, on the growth synchrony of five dominant temperate tree species across contrasting warm-dry and cool-wet climate conditions. Results show that temperature over water availability was the primary factor driving the growth and growth synchrony of the five species. Growth synchrony was significantly higher in warm-dry than in cool-wet areas, primarily due to more uniform climate conditions and higher climate sensitivity in the former. Rapid warming from the 1960s to the 1990s significantly enhanced tree growth synchrony in both areas, followed by a marked reversal as temperatures exceeded a certain threshold or warming slowed down, particularly in the warm-dry area. The growth synchrony variation patterns of the five species were highly consistent over time, although broadleaves exhibited higher synchrony than conifers, suggesting potential risks to forest resilience and stability under future climate change scenarios. Growing season temperatures and non-growing season temperatures and precipitation had a stronger positive effect on tree growth in the cool-wet area compared to the warm-dry area. High relative humidity hindered growth in the cool-wet area but enhanced it in the warm-dry area. Overall, our study highlights that the diversity and sensitivity of climate-growth relationships directly determine spatiotemporal growth synchrony. The world's largest butterfly genus Delias, commonly known as Jezebels, comprises ca. 251 species found throughout Asia, Australia, and Melanesia. Most species are endemic to islands in the Indo-Australian Archipelago or to New Guinea and nearby islands in Melanesia, and many species are restricted to montane habitats over 1200 m. We inferred an extensively sampled and well-supported molecular phylogeny of the group to better understand the spatial and temporal dimensions of its diversification. The remarkable diversity of Delias evolved in just ca. 15–16 Myr (crown age). The most recent common ancestor of a clade with most of the species dispersed out of New Guinea ca. 14 Mya, but at least six subsequently diverging lineages dispersed back to the island. Diversification was associated with frequent dispersal of lineages among the islands of the Indo-Australian Archipelago, and the divergence of sister taxa on a single landmass was rare and occurred only on the largest islands, most notably on New Guinea. We conclude that frequent inter-island dispersal during the Neogene—likely facilitated by frequent sea level change—sparked much diversification during that period. Many extant New Guinea lineages started diversifying 5 Mya, suggesting that orogeny facilitated their diversification. Our results largely agree with the most recently proposed species group classification system, and we use our large taxon sample to extend this system to all described species. Finally, we summarize recent insights to speculate how wing pattern evolution, mimicry, and sexual selection might also contribute to these butterflies’ rapid speciation and diversification. © 2024 Elsevier Inc.

Indicine cattle, also referred to as zebu (Bos taurus indicus), play a central role in pastoral communities across a wide range of agro-ecosystems, from extremely hot semiarid regions to hot humid tropical regions. However, their adaptive genetic changes following their dispersal into East Asia from the Indian subcontinent have remained poorly documented. Here, we characterize their global genetic diversity using high-quality whole-genome sequencing data from 354 indicine cattle of 57 breeds/populations, including major indicine phylogeographic groups worldwide. We reveal their probable migration into East Asia was along a coastal route rather than inland routes and we detected introgression from other bovine species. Genomic regions carrying morphology-, immune-, and heat-tolerance-related genes underwent divergent selection according to Asian agro-ecologies. We identify distinct sets of loci that contain promising candidate variants for adaptation to hot semi-arid and hot humid tropical ecosystems. Our results indicate that the rapid and successful adaptation of East Asian indicine cattle to hot humid environments was promoted by localized introgression from banteng and/or gaur. Our findings provide insights into the history and environmental adaptation of indicine cattle.

Citronella oil is one of the essential oils derived from the leaves and stem of lemongrass (Cymbopogon nardus). At present, most of the lemongrass oil industry in Indonesia is still in the form of upstream industries which only provide crude purified oils with low purity for export with a fairly low sales value. technology in improving the purity of citronella oil needs innovation in following the development of the industrial revolution 4.0. This article presents an overview of the research opportunities in the development of distillation for fractionating citronella oil referring to automation of optimization processes in the vacuum fractional distillation accompanied by inspection of the quality of essential oil isolation run in the line process based on smart sensors integrated in a computer program. The combination technique of quality control of essential oils in line process system combined with the optimization technique of the vacuum fractional distillation process has not been studied by advanced researchers. Therefore, this study has a state of the art process engineering and smart sensor-based quality control systems that can be developed for strengthening the Indonesian citronella oil industry.
