Natural products are in the spotlight of novelty for obtaining ecofriendly agents against pests. In this context, searching for new antifungal alternatives for control of grain microorganism is recommended. In this paper we show a brief description of nanoemulsification as a valuable tool for modification of essential oil properties. Low energy method was used for the preparation of Eugenia sulcata nanoemulsion. This approach was also used for the preparation of this type of colloid with β-caryophyllene, the main component of this essential oil. Especially regarding Aspegillus parasiticus, it was possible to observe increment of bioactivity by nanoemulsification of E. sulcata. This short communication contributes to better knowledge of mechanism in the rationalization of antifungal nanoemulsions based in essential oil and its main constituents, providing relevant insights that can be used for other complex volatile mixtures. © 2024 The Authors

The COVID-19 era has triggered profound effects on normal human life and the environment, as well as freshwater ecosystems worldwide. Despite the numerous influences, a strict COVID-19 lockdown might lead to improving the surface water quality and thus provide an unprecedented opportunity to restore the degraded freshwater resource. Thus, we intend to investigate the spatiotemporal water quality and preliminary health risks of heavy metals (loids) in the Karatoya River basin (KRB), a tropical urban river in Bangladesh. A total of 70 water samples were collected from 35 stations in KRB in 2019 and 2022 during the dry season. The results showed that the concentrations of Ni, Cu, Zn, Pb, Cd, and Cr were significantly reduced by 89.3–99.7% during the post-lockdown period (p © 2022, The Authors. All rights reserved.

For our knowledge, the roadside soils end up being the deposit of various residues discarded by drivers or passengers, plus, that coming from the runoff of rainwater. Basically, we do not know the impacts that this pollution causes on animals which inhabit these environments. Thus, in this study, our objective was to evaluate how the presence of plastic microfibers (MPFs), organic compounds and heavy metals affect the redox and cholinesterase homeostasis of mound-building termite [Cornitermes cumulans (workers) adults]. As a result, we noticed that MPFs were present in all sampled areas, being higher in road area (RA). Regardless of the presence of these pollutants, animals sampled in the RA were those in which we observed greater production of reactive oxygen species (ROS), hydrogen peroxide (HO) and nitric oxide (NO) (via nitrite), whose higher activities of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT), was not able to counterbalance the oxidative stress suggested by the evaluated biomarkers. Moreover, we observed increase in acetylcholinesterase (AChE) activity in these same animals, which suggests a cholinesterasic effect. Such alterations were positively correlated with the contamination of soil samples by Cd, Pb, Zn, Fe and Cu, as well as with the presence of the 11,10-guaiane-type sesquiterpenoid compound, identified only in the RA. Thus, our unique study reveals that the contamination of roadside soils constitutes an additional environmental stressor to populations of C. cumulans, which reinforces the need for greater attention and further investigation to be given to the pollution of these environments.

Cryptococcus gattii is one of the etiological agents of cryptococcosis. To achieve a successful infection, C. gattii cells must overcome the inhospitable host environment and deal with the highly specialized immune system and poor nutrients availability. Inside the host, C. gattii uses a diversified set of tools to maintain homeostasis and establish infection, such as the expression of remarkable and diverse heat shock proteins (Hsps). Grouped by molecular weight, little is known about the Hsp12 subset in pathogenic fungi. In this study, the function of the C. gattii HSP12.1 and HSP12.2 genes was characterized. Both genes were upregulated during murine infection and heat shock. The hsp12.1∆ null mutant cells were sensitive to plasma membrane and oxidative stressors. Moreover, HSP12 deletion induced C. gattii reactive oxygen species (ROS) accumulation associated with a differential expression pattern of oxidative stress-responsive genes compared to the wild type strain. Apart from these findings, the deletion of the paralog gene HSP12.2 did not lead to any detectable phenotype. Additionally, the double-deletion mutant strain hsp12.1∆/hsp12.2∆ presented a similar phenotype to the single-deletion mutant hsp12.1∆, suggesting a minor participation of Hsp12.2 in these processes. Furthermore, HSP12.1 disruption remarkably affected C. gattii virulence and phagocytosis by macrophages in an invertebrate model of infection, demonstrating its importance for C. gattii pathogenicity. © 2022, The Authors. The transient receptor potential vanilloid channel 4 (TRPV4) is associated with the development of several pathologies, particularly gastric disorders. However, there are no studies associating this receptor with the pathophysiology of gastric erosions. The aim of this study was to investigate the role of TRPV4 in the development of ethanol-induced gastric damage in vivo. Gastric lesions were induced by ethanol in Swiss mice pretreated with TRPV4 antagonists, GSK2193874 (0.1;0.3 and 0.9 mg/kg) or Ruthenium red (0.03;0.1 or 0.3 mg/kg) or its agonist, GSK1016790A (0.9 mg/kg). Gastric mucosal samples were taken for histopathology, immunohistochemistry, atomic force microscopy and evaluation of antioxidant parameters. The gastric mucus content and TRPV4 mRNA expression were analyzed. Ethanol exposure induced upregulation of gastric mRNA and protein expression of TRPV4. TRPV4 blockade promoted gastroprotection against ethanol-induced injury on macro- and microscopic levels, leading to reduced hemorrhage, cell loss and edema and enhanced gastric mucosal integrity. Moreover, an increase in superoxide dismutase (SOD) and glutathione (GSH) activity was observed, followed by a decrease in malondialdehyde (MDA) levels. TRPV4 blockade during alcohol challenge reestablished gastric mucus content. The combination of TRPV4 agonist and ethanol revealed macroscopic exacerbation of gastric damage area. Our results confirmed the association of TRPV4 with the development of gastric injury, showing the importance of this receptor for further investigations in the field of gastrointestinal pathophysiology and pharmacology. © 2021 Elsevier B.V.
