Protein isolation and identification of Pterois volitans spine venom coagulant activity

Pterois volitans, or commonly referred to lionfish, are fish species originating from Indo-Pacific waters but are becoming invasive in other regions such as the Caribbean and Atlantis. Various efforts have been made to reduce the number of lionfish, and one of them is by utilizing the venom on the spine. The venom extraction of P. volitans spines is done mechanically using sonication and centrifugation, and then protein isolation is carried out using salt. Coagulant activity from extract (crude venom) and lionfish venom protein isolate was done by counting PT (prothrombin time) and aPTT (activated partial thromboplastin time) which resulted that the crude venom and protein isolate of lionfish venom can accelerate blood clot (procoagulant) respectively up to 8.5 seconds and 6 seconds. Protein identification was made using LC-MS/MS device. The LC-MS/MS analysis showed that the protein isolate of lionfish venom contains Nomega-nitro-L-arginine methyl ester (L-NAME) compounds known to have procoagulant effects. From a series of tests mentioned, it concluded that P. volitans venom have procoagulant activity and one of the compounds responsible for it is L-NAME

The population of Pterois volitans has caused significant damage to other fish populations and coral reef ecosystems. Population control of P. volitans consumes a considerable cost so that the utilization of these fish needs to be sought to be useful along with controlling the population. This fish is known to contain the enzyme Phospholipase A2 (PLA2), which used as an antibiotic against some bacteria. This study will examine the antibacterial activity of the phospholipase A2 enzyme extracted from P. volitans venom to Escherichia coli, Bacillus subtilis, and Staphylococcus aureus. The method used to isolate the enzyme PLA2 is by using precipitation of ammonium sulfate (AS) and precipitation with ethanol. The results of the precipitation tested with the Lowry protein concentration test, the Marinetti PLA2 activity test, and the identification of the SDS-PAGE protein. The agar diffusion disc method is used to test the antibacterial activity. The results obtained from this research are that 80% ammonium sulfate precipitation method has the highest protein and enzyme activity with a ratio of 1.32 times compared to toxic extract. For antibacterial activity test results, an 80% ammonium sulfate sample may inhibit the activity of S. aureus bacteria but does not affect B. subtilis and E. coli.

Pterois volitans is an invasive native predatory fish species in the Indo-Pacific ocean that disrupt the food chain and damage coral reefs which cause ecosystem imbalances. These fish have venomous spines that made them inedible and avoided by a predator. Due to its rapid growth, utilization of this venom can be useful along with controlling the population. Recent studies show the benefits of phospholipase A2, which is a protein compound that contained in the venom has antibacterial activity and expected to be an antibacterial agent. In this study, we used heating and gradual purification to obtain the optimum concentration of Phospholipase A2. The protein isolates were analyzed using the Marinetti method, determination of concentration by Lowry test, identification of protein with SDS-Page, and the antibacterial activity test using agar diffusion. The results of phospholipase A2 obtained from the extract of P. volitans venom by purification method ammonium sulfate at 80% saturation with a heating time of 35 minutes had a specific enzyme activity of 0.0206 units / µg and can inhibit E. coli bacteria 98.81% and Salmonella sp. inhibit 89.28% with a concentration of 3.77 µg / ml.

Sensitivity is the effect of changes in any variables on changes in the risk results. The most common Risk Assessment Methodology in Indonesia is Muhlbauer, Modified Muhlbauer & Risk-Based Inspection. This study evaluated six natural gas distribution pipeline segments. In general, there are two objectives, firstly comparing the risk result between Risk Assessment Methodology. The following objectives are to compare the sensitivity. The Risk Assessments methodology is translated into a mathematic model and computed in Monte Carlo based simulation software. The risk value result shows that in high risk & medium risk pipeline segments, all methodology is valid with 24% and 13% deviation, respectively. The sensitivity analysis result shows that Muhlbauer & Modified Muhlbauer’s methodology is sensitive to third party activity. Risk-Based Inspection is sensitive to intrinsic damage mechanisms. This sensitivity data can be adopted to develop a more sensitive Risk Assessment Methodology for gas pipeline in the future.

Purpose of Review: This study aims to review the literature on the use of Nile tilapia-derived extracellular matrix (ECM) proteins as naturally derived scaffold material for tissue regeneration. Recent Findings: Nile tilapia has emerged as a particularly relevant specie for the development of fish-derived biomaterials. The skin, scale, and spine of various fish species have been extensively demonstrated as a potential source of native ECM proteins for use in regenerative medicine. Summary: When compared to synthetic polymers, collagen-based biomaterials introduce some disadvantages due to their lower biomechanical stiffness and rapid biodegradation. However, collagen-based biomaterials continue to be one of the most highly investigated natural materials for bone regeneration. Studies have pointed to tilapia as a satisfactory source of collagen for its quality and cost benefit, with good responses in vitro and in vivo, especially studies involving the skin as the main source of production. For both soft and hard tissue regenerations, Nile tilapia skin is being considered as a good source of collagen. Other sources (scales and spine) are less addressed in comparison to skin, which in turn, brings more consistent perspective in relation to clinical use. © 2020, Springer Nature Switzerland AG.

Background: Scleroderma-associated interstitial lung disease (SSc-ILD) is often observed in patients with systemic scleroderma (SSc) and its diagnosis contributes to early treatment decisions 1,2 . Objectives: The present study aims to automatically quantify SSc-ILD from high-resolution chest-computed tomography (HRCT) and to evaluate the association between interstitial lung disease (ILD) extension and lung function impairment. Methods: Ninety-four patients with SSc and 27 lung-healthy subjects matched for gender, weight, height, and age underwent HRCT, spirometry and carbon monoxide diffusion capacity (DL CO ). SSc-ILD was determined as the tissue mass present between -500 and +100 Hounsfield Units normalized by the total lung tissue mass (TLM). Cut off was the highest value obtained in the control group (25% of TLM). All data are presented as mean and standard deviations (Table I). An ANOVA test followed by Bonferroni post-hoc correction was used for comparisons among groups. Results: From 94 patients with SSc, 64 were classified as having pulmonary involvement (SSc-ILD) and 30 as not having pulmonary involvement (SSc No-ILD). In SSc-ILD subjects, there was a significant reduction in forced vital capacity (FVC), carbon monoxide diffusion capacity (DL CO ) and carbon monoxide diffusion capacity normalized by alveolar ventilation (DL CO /A) when compared with SSc No-ILD and control group. Lionfish (Pterois volitans) is an invasive species originating from Indo-Pacific Sea. The rapid growth and the absence of natural predators cause Lionfish to breed rapidly and reduce the native fish populations by up to 80%. Therefore, the study of the Lionfish must be intensified to obtain the usefulness of these animals. Studies that have stated that the extract of poison lionfish spines has the potential to become an anticancer agent for cervical cancer. The aim of the present work is to Investigation the Anticancer activity of Pterois volitans from Indonesia to human cervical carcinoma cell (HeLa cell). Crude venom (CV) that is resulted from the extraction process by sonicating of fresh venomous Pterois volitans as a sample was used. The PLA 2 sample of crude venom was heated on (50, 55, 60, 65, 70) °C for 30 min and purified by (20, 40, 60, 80) % ammonium sulfate. The concentration and purity are analyzed using Lowry method, Marinette’s, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and toxicity tested using the BSLT method (Brine Shrimp Lethality Test). The results of sample testing showed that the sample purified with 80% ammonium sulfate at 70°C had the highest protein activity and purity. After determining the purest and safest sample, the chosen sample then tested into HeLLa cell at 2, 250, 500, and 750 ppm, and compared to the cell control sample. The result has shown that the sample 80% of ammonium sulfate at 70°C is highest inhibition level toward HeLLa cell culture at 750 ppm.

Lionfish (Pterois volitans) is a native species of the Indo-Pacific Ocean. This species is known as destructive invasive species that invade in many areas. The invasion of Lionfish gives negative impacts on the ecology and economic condition of the invaded regions. Therefore, to reduce the number of Lionfish in the ocean and to attract people to exploit Lionfish, the study about Lionfish’s benefit is necessary. The recent study revealed that venom extract from Lionfish spines has a potential cytolytic effect that widely uses for antitumor studies. In this study, to obtain the optimum concentration of the solvent that used for protein isolation and to observe the antitumor properties of Lionfish further, we isolated the protein of Lionfish venom extract with ammonium sulfate and tested it on HeLa cell as the model of tumor cells. The Lowry and BSLT test showed that the highest protein concentration and activity obtained when the highest level of ammonium sulfate isolated crude venom. The highest percentage of inhibition in HeLa cells reach 17-19% at the highest dose of Lionfish protein isolate. Indeed, the inhibition effect on HeLa cells can be enhanced because SDS-PAGE results showed that there are several proteins with different effects on tumor cells contained in the protein isolate of Lionfish venom. Thus, eliminating these proteins might increase the inhibition effects of Lionfish venom protein in tumor cells. These results suggest that Lionfish has an antitumor effect, but need further purification step and more evaluation of the protein in Lionfish venom extract that can enhance the antitumor effect.

Lionfish Pterois volitans is a tropical Indo-Pacific native species and invaded on marine ecosystem due to its rapid growth and predatory properties that were highly affecting fish and coral populations. Recent studies show the benefits of natural antioxidants, especially as the prevention of cancer and cardiovascular disease. Crude Venom (CV) was extracted using sonication and precipitated using ethanol and Ammonium Sulfate (AS) with 20, 40, 60, and 80% saturation. The protein concentration of each sample detected using Lowry’s method, and SDS-PAGE was done to find out the Molecular Weight (MW) of protein samples. Toxicity analysis to determine the Lethal Concentration (LC50) value was done using Brine Shrimp Lethality Test (BSLT) and antioxidant activity tested using 2,2-diphenyl-1-picrylhydrazyl (DPPH) method to determined Inhibitory Concentration (IC50) value. AS 80% had the higher value of protein concentration 361.76 µg/mL. SDS-PAGE analysis showed that AS 80% had a protein band at 7.9, 46.2 and 52.7 kD which supposed to be the protein that had antioxidant activity. The test showed the LC50 of AS 80% 101.93 µg/mL. Antioxidant analysis showed AS 80% had the higher value of inhibition 57.08 % with IC50 1.56 mg/mL. This research showed that AS 80% has the highest potential as an antioxidant compound due to its ability to inhibit free radicals.
