To identify potent antiviral compounds, we introduced a high-throughput screen platform that can rapidly classify hit compounds according to their target. In our platform, we performed a compound screen using a lentivirus-based pseudovirus presenting a spike protein of coronavirus, and we evaluated the hit compounds using an amplified luminescence proximity homogeneous assay (alpha) test with purified host receptor protein and the receptor binding domain of the viral spike. With our screen platform, we were able to identify both spike-specific compounds (class I) and broad-spectrum antiviral compounds (class II). Among the hit compounds, thiosemicarbazide was identified to be selective to the interaction between the viral spike and its host cell receptor, and we further optimized the binding potency of thiosemicarbazide through modification of the pyridine group. Among the class II compounds, we found raloxifene and amiodarone to be highly potent against human coronaviruses including Middle East respiratory syndrome coronavirus (MERS-CoV), severe acute respiratory syndrome coronavirus (SARS-CoV), and SARS-CoV-2. In particular, using analogs of the benzothiophene moiety, which is also present in raloxifene, we have identified benzothiophene as a novel structural scaffold for broad-spectrum antivirals. This work highlights the strong utility of our screen platform using a pseudovirus assay and an alpha test for rapid identi-fication of potential antiviral compounds and their mechanism of action, which can lead to the accelerated development of therapeutics against newly emerging viral infections.

Contexte Au Cameroun, le ratio de mortalité maternelle était estimé en 2011 à 782 décès pour 100 000 naissances vivantes. La surveillance des décès maternels (DM) a débuté en 2014. Cependant, elle se heurte aux problèmes de sous-notification des cas et de mauvaise qualité des données. L'objectif de cette étude était d'évaluer le système de surveillance des DM dans le DS de Deïdo. Méthodes Les directives de CDC ont été utilisées pour l'évaluation des systèmes de surveillance. L'étude s'est déroulée de janvier à décembre 2017 dans huit fosas choisis par convenance dans le DS de Deïdo. Les données étaient recueillies auprès de tous les points focaux surveillance (PFS) des DM, des formulaires hebdomadaires des fosa et des registres des salles de travail et d'accouchement, des services d'hospitalisation, de chirurgie, et d'urgence. Des « score cards » ont été élaborées pour évaluer la qualité, la sensibilité, l'acceptabilité et l'utilité des données. Les données étaient analysées via les logiciels EPI Info et Microsoft Excel. Résultats Des neuf PFS des DM interrogés, six étaient des sages-femmes et huit énonçaient exactement la définition de DM. Sur les 62 DM enregistrés dans les registres de trois des huit fosas choisis par convenance, 61 % (38/62) ont été signalés au système. Le temps pour remplir un formulaire était jugé long pour deux répondants ; 78 % (324/417) fiches de notification examinées étaient correctement remplies et 80 % (373/468) de ces fiches étaient envoyées à temps au District. La surveillance des DM faisait partie du travail quotidien des neufs PFS. Le suivi des DM a conduit le DS à sensibiliser les fosas de niveau inférieur sur la référence rapide des urgences obstétricales. Discussion/Conclusion Malgré une faible sensibilité, la surveillance des DM est réactive, utile et acceptée par le personnel.
