Activity-based E2 conjugating enzyme (E2)-ubiquitin (Ub) probes have recently emerged as effective tools for studying the molecular mechanism of E3 ligase (E3)-catalyzed ubiquitination. However, the preparation of existing activity-based E2-Ub probes depends on recombination technology and bioconjugation chemistry, limiting their structural diversity. Herein we describe an expedient total chemical synthesis of an E2 enzyme variant through a hydrazide-based native chemical ligation, which enabled the construction of a structurally new activity-based E2-Ub probe to covalently capture the catalytic site of Cys-dependent E3s. Chemical cross-linking coupled with mass spectrometry (CXMS) demonstrated the utility of this new probe in structural analysis of the intermediates formed during Nedd4 and Parkin-mediated transthiolation. This study exemplifies the utility of chemical protein synthesis for the development of protein probes for biological studies.

In NH 3 capture technologies, the desorption process is usually driven by high temperature and low pressure (such as 150–200 °C under vacuum), which accounts for intensive energy consumption and CO 2 emission. Developing light responsive adsorbent is promising in this regard but remains a great challenge. Here, we for the first time designed and synthesized a light responsive azophenol-containing covalent organic framework (COF), COF-HNU38, to address this challenge. We found that at 25 °C and 1.0 bar, the cis -COF exhibited a NH 3 uptake capacity of 7.7 mmol g −1 and a NH 3 /N 2 selectivity of 158. In the adsorbed NH 3 , about 29.0 % could be removed by vis-light irradiated cis-trans isomerization at 25 °C, and the remaining NH 3 might be released at 25 °C under vacuum. Almost no decrease in adsorption capacity was observed after eight adsorption-desorption cycles. As such, an efficient NH 3 capture and low energy release strategy was established thanks to the multiple hydrogen bond interactions (which are strong in total but weak in individuals) between NH 3 and the smart COF, as well as the increased polarity and number of hydrogen bond sites after the trans-cis isomerization.

Regression technique such as partial correlation analysis has been widely used as tool of prediction in business, finance and biomedical field. However, the application of predictive analysis in chemical process, specifically palm oil refinery process has rarely been done. Therefore, the objective of this paper is to present a quality prediction and diagnosis tool using partial correlation analysis, with the aim to predict the quality of refined palm oil and to diagnose the crude palm oil and process variables. Several statistical analysis are applied in data pre-process to obtain statistical sample size, optimum sampling and processing time of the process. The predictor coefficient is developed using partial correlation analysis while control chart is used to monitor the process behavior of both predicted and actual output value. The monitored out-of-control behavior is then diagnosed using SPE-contribution plot to identify the faulty input variables, thus pre-treatment can be executed before the refining process. The predicted model is successfully developed with MSE value less than 0.01 and three faulty variables are identified.

In this work we describe the design process of a grating spectrometer with fast superconducting THz detectors based on Niobiumnitride. The main application is the use as a diagnostic tool for accelerator-based sources of THz radiation like synchrotrons, where interactions of the electron bunches in storage ring lead to fluctuations in spectrum and intensity of the emitted radiation. Guidelines for the design of the two main parts, grating and detector design, are given. Here, a blazed reflection grating for a frequency range of 1 THz to 5 THz has been developed and successfully fabricated in a high-precision milling process out of brass. Also three different planar antenna designs (double-slot, annular slot and log-per spiral) in combination with quasioptical bandpass filters for the THz range have been simulated and evaluated for best spectral resolution and power coupling. Finally, we have verified grating performance for pulsed THz radiation of 1 THz in an experiment conducted at the synchrotron light source of the KIT. Excellent agreement of the grating performance to the simulations has been found.
